Fusion of secretory vesicles isolated from rat liver
Secretory vesicles isolated from rat liver were found to fuse after exposure to Ca2+. Vescle fusion is characterized by the occurrence of twinned vesicles with a continuous cleavage plane between two vesicles in freeze-fracture electron microscopy. The number of fused vesicles increases with increasing Ca2+-concentrations and is half maximal around 10â6 m. Other divalent cations (Ba2+, Sr2+, and Mg2+) were ineffective. Mg2+ inhibits Ca2+-induced fusion. Therefore, the fusion of secretory vesiclesin vitro is Ca2+ specific and exhibits properties similar to the exocytotic process of various secretory cells.
Various substances affecting secretionin vivo (microtubular inhibitors, local anethetics, ionophores) were tested for their effect on membrane fusion in our system.
The fusion of isolated secretory vesicles from liver was found to differ from that of pure phospholipid membranes in its temperature dependence, in its much lower requirement for Ca2+, and in its Ca2+-specificity. Chemical and enzymatic modifications of the vesicle membrane indicate that glycoproteins may account for these differences
The WEBT BL Lacertae Campaign 2001 and its extension : Optical light curves and colour analysis 1994â2002
BL Lacertae has been the target of four observing campaigns by the Whole Earth Blazar Telescope (WEBT) collaboration. In this paper we present UBVRI light curves obtained by theWEBT from 1994 to 2002, including the last, extended BL Lac 2001 campaign. A total of about 7500 optical observations performed by 31 telescopes from Japan to Mexico have been collected, to be added to the âŒ15 600 observations of the BL Lac Campaign 2000. All these data allow one to follow the source optical emission behaviour with unprecedented detail. The analysis of the colour indices reveals that the flux variability can be interpreted in terms of two components: longer-term variations occurring on a fewday time scale appear as mildly-chromatic events, while a strong bluer-when-brighter chromatism characterizes very fast (intraday) flares. By decoupling the two components, we quantify the degree of chromatism inferring that longer-term flux changes imply moving along a âŒ0.1 bluerwhen- brighter slope in the B â R versus R plane; a steeper slope of âŒ0.4 would distinguish the shorter-term variations. This means that, when considering the long-term trend, the B-band flux level is related to the R-band one according to a power law of index âŒ1.1. Doppler factor variations on a âconvexâ spectrum could be the mechanism accounting for both the long-term variations and their slight chromatism.Reig Torres, Pablo, [email protected]
Multiwavelength behaviour of the blazar 3CÂ 279: decade-long study from Îł-ray to radio
We report the results of decade-long (2008â2018) Îł-ray to 1 GHz radio monitoring of the blazar 3C 279, including GASP/WEBT, Fermi and Swift data, as well as polarimetric and spectroscopic data. The X-ray and Îł-ray light curves correlate well, with no delay âł 3 h, implying general cospatiality of the emission regions. The Îł-rayâoptical fluxâflux relation changes with activity state, ranging from a linear to a more complex dependence. The behaviour of the Stokes parameters at optical and radio wavelengths, including 43 GHz Very Long Baseline Array images, supports either a predominantly helical magnetic field or motion of the radiating plasma along a spiral path. Apparent speeds of emission knots range from 10 to 37c, with the highest values requiring bulk Lorentz factors close to those needed to explain Îł-ray variability on very short time-scales. The Mgâii emission line flux in the âblueâ and âredâ wings correlates with the optical synchrotron continuum flux density, possibly providing a variable source of seed photons for inverse Compton scattering. In the radio bands, we find progressive delays of the most prominent light-curve maxima with decreasing frequency, as expected from the frequency dependence of the Ï = 1 surface of synchrotron self-absorption. The global maximum in the 86 GHz light curve becomes less prominent at lower frequencies, while a local maximum, appearing in 2014, strengthens toward decreasing frequencies, becoming pronounced at âŒ5 GHz. These tendencies suggest different Doppler boosting of stratified radio-emitting zones in the jet.First author draf
Emergence and preservation of a chronically sick building
STUDY OBJECTIVEâTo investigate the merits of case studies as complementary methodological approaches in the study of the sick building syndrome.âšSETTING AND PARTICIPANTSâA Swedish office building with longstanding health problems, and its inhabitants.âšDESIGNâThis paper is a case study based both on historical and present, quantitative as well as qualitative, documentary material, produced over the years by distinct parties, and on semi-structured interviews.âšRESULTSâLong drawn conflictive processes within the building were identified. It was revealed that the organisation for dealing with environmental problems was split, and ineffective with poor patterns of communication. It was suggested that this generated a situation of chronic stress leading to the persistence of symptoms.âšCONCLUSIONSâBy their capacity to identify internal processes within building contexts, case study methodology can contribute to a better understanding and management of sick building syndrome. The results of this study suggest that psychosocial factors, among them organisational structures and communication patterns, should be given close attention.âšâšâšKeywords: case study; sick building syndrom
