Assessment of experimental competencies is not yet well established. We just began an empirical pilot study, too. This study aims to examine if secondary school students may successfully use a predefined remote lab activity to
introduce themselves to atomic physics. The analysis of spectra is a fundamental component for the understanding of wave optics and color perception. Hence, every student should have the opportunity to conduct own optical emission experiments. Since spectrometers are expensive and an accurate calibration is necessary to achieve energy distribution spectra of high quality, we developed a remotely controlled laboratory. We evaluated the experimental set-up and the accompanying worksheet with groups of two to four students in a laboratory condition. Additionally, the emerged learning material was brought to school and tested as a homework activity with 9th-graders replacing the regular introduction to atomic physics. The results show that the experiment presented here can be used by ninth grade students and is useful in connection with the created material for the self-regulated introduction to atomic physics in the context of homework
Hypoxia. HIF-mediated articular chondrocyte function: prospects for cartilage repair
In a chronically hypoxic tissue such as cartilage, adaptations to hypoxia do not merely include cell survival responses, but also promotion of its specific function. This review will focus on describing such hypoxia-mediated chondrocyte function, in particular in the permanent articular cartilage. The molecular details of how chondrocytes sense and respond to hypoxia and how this promotes matrix synthesis have recently been examined, and specific manipulation of hypoxia-induced pathways is now considered to have potential therapeutic application to maintenance and repair of articular cartilage
The Perioperative Nursing Workforce Program in NSW: How a professional perioperative nursing association meets one of its mandates Part 1
The need to review and change the way nursing care is delivered in perioperative settings is predicated on nursing workforce shortages, the changing, increasingly technologised and risk-prone OR practice milieu, and increasing demand for surgery. In responding to members' concerns about these issues, the NSW Operating Theatre Association Inc. (NSW OTA) in conjunction with and with sponsorship from the NSW Chief Nursing and Midwifery Officer, initiated and oversaw the development of a program, called the Perioperative Nursing Workforce Program (PNWP). The aims of this program are to make better use of human resources, to improve the way care is provided and thus improve patient outcomes; and to empower perioperative nurses so they are capable of independently improving their working environment. The program, which takes a practice development approach, program participants and some of their projects are presented in this paper. What is known about the topic The role of perioperative professional nursing associations is to write standards for practice and to assist in the professional development of their members. Practice development is hypothesised to assist clinical nurses to 'see' their work contexts afresh and to implement changes to improve patient care by focusing on patient-centredness and the use of credible evidence. What this article contributes It describes the contents and the implementation of a perioperative nursing workforce program, initiated by the NSW OTA and auspiced by the Chief Nursing and Midwifery Officer, NSW Health. The PNWP uses the tenets of practice development (PD) to achieve its aims and this is possibly the first use of PD in perioperative settings, and on a statewide basis
Reconstitution of the complete pathway of ITS2 processing at the pre-ribosome
Removal of internal transcribed spacer 2 (ITS2) from pre-ribosomal RNA is essential to make functional ribosomes. This complicated processing reaction begins with a single endonucleolytic cleavage followed by exonucleolytic trimming at both new cleavage sites to generate mature 5.8S and 25S rRNA. We reconstituted the 7S -> 5.8S processing branch within ITS2 using purified exosome and its nuclear cofactors. We find that both Rrp44's ribonuclease activities are required for initial RNA shortening followed by hand over to the exonuclease Rrp6. During the in vitro reaction, ITS2-associated factors dissociate and the underlying 'foot' structure of the pre-60S particle is dismantled. 7S pre-rRNA processing is independent of 5S RNP rotation, but 26S -> 25S trimming is a precondition for subsequent 7S -> 5.8S processing. To complete the in vitro assay, we reconstituted the entire cycle of ITS2 removal with a total of 18 purified factors, catalysed by the integrated activities of the two participating RNA-processing machines, the Las1 complex and nuclear exosome

Interleukin-1β sequesters hypoxia inducible factor 2α to the primary cilium.
BACKGROUND: The primary cilium coordinates signalling in development, health and disease. Previously we have shown that the cilium is essential for the anabolic response to loading and the inflammatory response to interleukin-1β (IL-1β). We have also shown the primary cilium elongates in response to IL-1β exposure. Both anabolic phenotype and inflammatory pathology are proposed to be dependent on hypoxia-inducible factor 2 alpha (HIF-2α). The present study tests the hypothesis that an association exists between the primary cilium and HIFs in inflammatory signalling. RESULTS: Here we show, in articular chondrocytes, that IL-1β-induces primary cilia elongation with alterations to cilia trafficking of arl13b. This elongation is associated with a transient increase in HIF-2α expression and accumulation in the primary cilium. Prolyl hydroxylase inhibition results in primary cilia elongation also associated with accumulation of HIF-2α in the ciliary base and axoneme. This recruitment and the associated cilia elongation is not inhibited by blockade of HIFα transcription activity or rescue of basal HIF-2α expression. Hypomorphic mutation to intraflagellar transport protein IFT88 results in limited ciliogenesis. This is associated with increased HIF-2α expression and inhibited response to prolyl hydroxylase inhibition. CONCLUSIONS: These findings suggest that ciliary sequestration of HIF-2α provides negative regulation of HIF-2α expression and potentially activity. This study indicates, for the first time, that the primary cilium regulates HIF signalling during inflammation
Fertility preservation and management of pregnancy in melanoma patients requiring systemic therapy
Melanoma is one of the most common cancers in adolescents and adults at fertile age, especially in women. With novel and more effective systemic therapies that began to profoundly change the dismal outcome of melanoma by prolonging overall survival, the wish for fertility preservation or even parenthood has to be considered for a growing portion of melanoma patients—from the patients' as well as from the physicians' perspective. The dual blockade of the mitogen-activated protein kinase pathway by B-Raf proto-oncogene serine/threonine kinase and mitogen-activated protein kinase inhibitors and the immune checkpoint inhibition by anti-programmed cell death protein 1 and anti-cytotoxic T-lymphocyte-associated protein-4 monoclonal antibodies constitute the current standard systemic approaches to combat locally advanced or metastatic melanoma. Here, the preclinical data and clinical evidence of these systemic therapies are reviewed in terms of their potential gonadotoxicity, teratogenicity, embryotoxicity and fetotoxicity. Recommendations for routine fertility and contraception counseling of melanoma patients at fertile age are provided in line with interdisciplinary recommendations for the diagnostic work-up of these patients and for fertility-protective measures. Differentiated recommendations for the systemic therapy in both the adjuvant and the advanced, metastatic treatment situation are given. In addition, the challenges of pregnancy during systemic melanoma therapy are discussed
