    The isolation of differentially expressed cDNA clones from the filarial nematode <i>Brugia pahangi</i>

    A cDNA library constructed from 3 day post-infective L3 of the filarial nematode Brugia pahangi was screened by differential hybridization with cDNA probes prepared from different life-cycle stages. Five cDNA clones hybridizing selectively to the mosquito-derived L3 probe were isolated and characterized. Northern blot analysis of 4 of the clones confirmed that each was most highly expressed in the mosquito-derived L3. The expression of each mRNA during parasite development in the mosquito vector was investigated using RT-PCR, and all were shown to be abundant in the immature L3. Four of the 5 cDNAs cloned coded for structural proteins: 2 cuticular collagens, and the muscle proteins tropomyosin and troponin. Further studies on troponin using an antiserum raised to the recombinant protein demonstrated that the protein, unlike the mRNA, was present in all life-cycle stages examined, while immunogold labelling demonstrated that it was localized to the muscle blocks

    The evolution of the structure factor is studied during the phase-ordering dynamics of the kinetic Ising model with conserved order parameter. A preasymptotic multiscaling regime is found as in the solution of the Cahn-Hilliard-Cook equation, revealing that the late stage of phase-ordering is always approached through a crossover from multiscaling to standard scaling, independently from the nature of the microscopic dynamics.Comment: 11 pages, 3 figures, to be published in Europhys.     Laboratory exploration of mineral precipitates from Europa's subsurface ocean

    The precipitation of hydrated phases from a chondritic-like Na-Mg-Ca-SO4-Cl solution is studied using in situ synchrotron X-ray powder diffraction, under rapid (360 K hour−1, T = 250-80 K, t = 3 hours) and ultra-slow (0.3 K day−1, T= 273-245K, t = 242 days) freezing conditions. The precipitation sequence under slow cooling initially follows the predictions of equilibrium thermodynamics models, however after ∼ 50 days at 245 K, the formation of the highly hydrated sulphate phase Na2Mg(SO4)2·16H2O, a relatively recent discovery in the Na2Mg(SO4)2-H2O system, was observed. Rapid freezing, on the other hand, produced an assemblage of multiple phases which formed within a very short timescale (≤4 minutes, ∆T = 2 K) and, although remaining present throughout, varied in their relative proportions with decreasing temperature. Mirabilite and meridianiite were the major phases, with pentahydrite, epsomite, hydrohalite, gypsum, bl¨odite, konyaite and loweite also observed. Na2Mg(SO4)2·16H2O was again found to be present and increased in proportion relative to other phases as the temperature decreased. Results are discussed in relation to possible implications for life on Europa and application to other icy ocean worlds

    Feline immunodeficiency virus (FIV) infection in domestic pet cats in Australia and New Zealand: Guidelines for diagnosis, prevention and management

    Despite the passage of over 30 years since its discovery, the importance of feline immunodeficiency virus (FIV) on the health and longevity of infected domestic cats is hotly debated amongst feline experts. Notwithstanding the absence of good quality information, Australian and New Zealand (NZ) veterinarians should aim to minimise the exposure of cats to FIV. The most reliable way to achieve this goal is to recommend that all pet cats are kept exclusively indoors, or with secure outdoor access (e.g., cat enclosures, secure gardens), with FIV testing of any in-contact cats. All animal holding facilities should aim to individually house adult cats to limit the spread of FIV infection in groups of animals that are stressed and do not have established social hierarchies. Point-of-care (PoC) FIV antibody tests are available in Australia and NZ that can distinguish FIV-infected and uninfected FIV-vaccinated cats (Witness™ and Anigen Rapid™). Although testing of whole blood, serum or plasma remains the gold standard for FIV diagnosis, PoC testing using saliva may offer a welfare-friendly alternative in the future. PCR testing to detect FIV infection is not recommended as a screening procedure since a negative PCR result does not rule out FIV infection and is only recommended in specific scenarios. Australia and NZ are two of three countries where a dual subtype FIV vaccine (Fel-O-Vax® FIV) is available and offers a further avenue for disease prevention. Since FIV vaccination only has a reported field effectiveness of 56% in Australia, and possibly lower in NZ, FIV-vaccinated cats should undergo annual FIV testing prior to annual FIV re-vaccination using a suitable PoC kit to check infection has not occurred in the preceding year. With FIV-infected cats, clinicians should strive to be even more thorough than usual at detecting early signs of disease. The most effective way to enhance the quality of life and life expectancy of FIV-infected cats is to optimise basic husbandry and to treat any concurrent conditions early in the disease course. Currently, no available drugs are registered for the treatment of FIV infection. Critically, the euthanasia of healthy FIV-infected cats, and sick FIV-infected cats without appropriate clinical investigations, should not occur

    Surface Modification of Polyethylene with Multi-End-Functional Polyethylene Additives

    We have prepared and characterized a series of multifluorocarbon end-functional polyethylene additives, which when blended with polyethylene matrices increase surface hydrophobicity and lipophobicity. Water contact angles of >112° were observed on spin-cast blended film surfaces containing less than 1% fluorocarbon in the bulk, compared to 98° in the absence of any additive. Crystallinity in these films gives rise to surface roughness that is an order of magnitude greater than is typical for amorphous spin-cast films but is too little to give rise to superhydrophobicity. X-ray photoelectron spectroscopy (XPS) confirms the enrichment of the multifluorocarbon additives at the air surface by up to 80 times the bulk concentration. Ion beam analysis was used to quantify the surface excess of the additives as a function of composition, functionality, and molecular weight of either blend component. In some cases, an excess of the additives was also found at the substrate interface, indicating phase separation into self-stratified layers. The combination of neutron reflectometry and ion beam analysis allowed the surface excess to be quantified above and below the melting point of the blended films. In these films, where the melting temperatures of the additive and matrix components are relatively similar (within 15 °C), the surface excess is almost independent of whether the blended film is semicrystalline or molten, suggesting that the additive undergoes cocrystallization with the matrix when the blended films are allowed to cool below the melting point

    Results from the adaptive optics coronagraph at the William Herschel Telescope

    Described here is the design and commissioning of a coronagraph facility for the 4.2-m William Herschel Telescope (WHT) and its Nasmyth Adaptive Optics for Multi-purpose Instrumentation (NAOMI). The use of the NAOMI system gives an improved image resolution of 0.15 arcsec at a wavelength of 2.2 μm. This enables the Optimised Stellar Coronagraph for Adaptive optics (OSCA) to suppress stellar light using smaller occulting masks and thus allows regions closer to bright astronomical objects to be imaged. OSCA provides a selection of 10 different occulting masks with sizes of 0.25–2.0 arcsec in diameter, including two with full grey-scale Gaussian profiles. There is also a choice of different sized and shaped Lyot stops (pupil plane masks). Computer simulations of the different coronagraphic options with the NAOMI segmented mirror have relevance for the next generation of highly segmented extremely large telescopes

    Biofilm formation on enteral feeding tubes by Cronobacter sakazakii, Salmonella serovars and other Enterobacteriaceae

    WHO (2007) recommended that to reduce microbial risks, powdered infant formula should be reconstituted with water at temperatures >70 °C, and that such feeds should be used within 2 h of preparation. However, this recommendation does not consider the use of enteral feeding tubes which can be in place for more than 48 h and can be loci for bacterial attachment. This study determined the extent to which 29 strains of Cronobacter sakazakii, Salmonella serovars, other Enterobacteriaceae and Acinetobacter spp. can adhere and grow on enteral feeding tubes composed of polyvinyl chloride and polyurethane. The study also included silver-impregnated tubing which was expected to have antibacterial activity. Bacterial biofilm formation by members of the Enterobacteriaceae was ca. 105-106 cfu/cm after 24 h. Negligible biofilm was detected for Acinetobacter gensp. 13; ca. 10 cfu/cm, whereas Cr. sakazakii strain ATCC 12868 had the highest biofilm cell density of 107 cfu/cm. Biofilm formation did not correlate with capsule production, and was not inhibited on silver-impregnated tubing. Bacteria grew in the tube lumen to cell densities of 107 cfu/ml within 8 h, and 109 cfu/ml within 24 h. It is plausible that in vivo the biofilm will both inoculate subsequent routine feeds and as the biofilm ages, clumps of cells will be shed which may survive passage through the neonate's stomach. Therefore biofilm formation on enteral feeding tubes constitutes a risk factor for susceptible neonates

    The New ‘Hidden Abode’: Reflections on Value and Labour in the New Economy

    In a pivotal section of Capital, volume 1, Marx (1976: 279) notes that, in order to understand the capitalist production of value, we must descend into the ‘hidden abode of production’: the site of the labour process conducted within an employment relationship. In this paper we argue that by remaining wedded to an analysis of labour that is confined to the employment relationship, Labour Process Theory (LPT) has missed a fundamental shift in the location of value production in contemporary capitalism. We examine this shift through the work of Autonomist Marxists like Hardt and Negri, Lazaratto and Arvidsson, who offer theoretical leverage to prize open a new ‘hidden abode’ outside employment, for example in the ‘production of organization’ and in consumption. Although they can open up this new ‘hidden abode’, without LPT's fine-grained analysis of control/resistance, indeterminacy and structured antagonism, these theorists risk succumbing to empirically naive claims about the ‘new economy’. Through developing an expanded conception of a ‘new hidden abode’ of production, the paper demarcates an analytical space in which both LPT and Autonomist Marxism can expand and develop their understanding of labour and value production in today's economy. </jats:p

    High-Energy Cosmology: gamma rays and neutrinos from beyond the galaxy

    Our knowledge of the high-energy universe is undergoing a period of rapid change as new astronomical detectors of high-energy radiation start to operate at their design sensitivities. Now is a boomtime for high-energy astrophysics, with new discoveries from Swift and HESS, results from MAGIC and VERITAS starting to be reported, the upcoming launches of the gamma-ray space telescopes GLAST and AGILE, and anticipated data releases from IceCube and Auger. A formalism for calculating statistical properties of cosmological gamma-ray sources is presented. Application is made to model calculations of the statistical distributions of gamma-ray and neutrino emission from (i) beamed sources, specifically, long-duration GRBs, blazars, and extagalactic microquasars, and (ii) unbeamed sources, including normal galaxies, starburst galaxies and clusters. Expressions for the integrated intensities of faint beamed and unbeamed high-energy radiation sources are also derived. A toy model for the background intensity of radiation from dark-matter annihilation taking place in the early universe is constructed. Estimates for the gamma-ray fluxes of local group galaxies, starburst, and infrared luminous galaxies are briefly reviewed. Because the brightest extragalactic gamma-ray sources are flaring sources, and these are the best targets for sources of PeV -- EeV neutrinos and ultra-high energy cosmic rays, rapidly slewing all-sky telescopes like MAGIC and an all-sky gamma-ray observatory beyond Milagro will be crucial for optimal science return in the multi-messenger age.Comment: 10 pages, 3 figs, accepted for publication in the Barcelona Conference on Multimessenger Astronomy; corrected eq. 27, revised Fig. 3, added 2 ref
