    The Utility of Cancer-Related Cultural Constructs to Understand Colorectal Cancer Screening Among African Americans

    Data suggest that colorectal cancer could be cut by approximately 60% if all people aged 50 years or older received regular screening. Studies have identified socio-cultural attitudes that might inform cancer education and screening promotion campaigns. This article applies item response theory (IRT) to a set of survey items selected to assess sociocultural attitudes in order to determine how current measures may affect what we know about how these attitudes affect colorectal cancer screening (CRCS). Design and Methods. A survey of colorectal cancer screening, screening attitudes and cultural beliefs was administered to 1021 African Americans - 683 women and 338 men, ages 50 to 75. Eligibility criteria for participation included being born in the United States, selfidentified African American male or female, age 50 to 75 years. The IRT analysis was performed on 655 individuals with complete data for the 43 observed variables. Results. Twenty-nine items comprise the Multi-construct African American Cultural Survey (MAACS) that addresses seven cultural constructs: mistrust/distrust, privacy, ethnic identity, collectivism, empowerment, and male gender roles. The items provide adequate information about the attitudes of the population across most levels of the constructs assessed. Among the sociocultural variables considered, empowerment (OR=1.078; 95% CI: 1.008, 1.151) had the strongest association with CRCS adherence and privacy showed promise. Conclusions. The MAACS provides a fixed length questionnaire to assess African American CRCS attitudes, two new constructs that might assist in CRCS promotion, and a suggested focus for identification of additional constructs of interest

    Cereal and Pulse Crops with Improved Resistance to Pratylenchus thornei Are Needed to Maximize Wheat Production and Expand Crop Sequence Options

    n the subtropical grain region of eastern Australia, two experiments were conducted, one initially with 2490 P. thornei/kg soil, the other with 8150 P. thornei/kg soil at 0–0.9 m soil depth. We determined the effect of P. thornei, residual from a weed-free fallow and pre-cropping with several cultivars each of barley (Hordeum vulgare), faba bean (Vicia faba), chickpea (Cicer arietinum), and wheat (Triticum aestivum) (Phase 1), on the growth of wheat cultivars with intolerance or tolerance to P. thornei (Phase 2). Pratylenchus thornei substantially increased after growing all cultivars of the Phase 1 faba bean, barley, and most cultivars of chickpea and wheat, and decreased after two moderately resistant wheat cultivars and the fallow treatment. The biomass of the Phase 2 tolerant cultivar ranged from 5070 to 6780 kg/ha and the intolerant cultivar 1020 to 4740 kg/ha. There was a negative linear relationship between P. thornei population densities and biomass of the Phase 2 intolerant cultivar but not of the tolerant cultivar. Growers are at risk of financial loss because they are restricted in their choice of crops to reduce damaging population densities of P. thornei. The development of resistant and tolerant crop genotypes can maximize production in P. thornei-affected farming systems

    Diverse and Variable Community Structure of Picophytoplankton across the Laurentian Great Lakes

    The Laurentian Great Lakes provide economic support to millions of people, drive biogeochemical cycling, and are an important natural laboratory for characterizing the fundamental components of aquatic ecosystems. Small phytoplankton are important contributors to the food web in much of the Laurentian Great Lakes. Here, for the first time, we reveal and quantify eight phenotypically distinct picophytoplankton populations across the Lakes using a multilaser flow cytometry approach, which distinguishes cells based on their pigment phenotype. The distributions and diversity of picophytoplankton flow populations varied across lakes and depths, with Lake Erie standing out with the highest diversity. By sequencing sorted cells, we identified several distinct lineages of Synechococcales spanning Subclusters 5.2 and 5.3. Distinct genotypic clusters mapped to phenotypically similar flow populations, suggesting that there may not be a clear one-to-one mapping between genotypes and phenotypes. This suggests genome-level differentiation between lakes but some degree of phenotypic convergence in pigment characteristics. Our results demonstrate that ecological selection for locally adapted populations may outpace homogenization by physical transport in this interconnected system. Given the reliance of the Lakes on in situ primary production as a source for organic carbon, this work sets the foundation to test how the community structure of small primary producers corresponds to biogeochemical and food web functions of the Great Lakes and other freshwater systems

    Transcriptional and Proteomic Analysis of a Ferric Uptake Regulator (Fur) Mutant of Shewanella oneidensis: Possible Involvement of Fur in Energy Metabolism, Transcriptional Regulation, and Oxidative Stress

    The iron-directed, coordinate regulation of genes depends on the fur (ferric uptake regulator) gene product, which acts as an iron-responsive, transcriptional repressor protein. To investigate the biological function of a fur homolog in the dissimilatory metal-reducing bacterium Shewanella oneidensis MR-1, a fur knockout strain (FUR1) was generated by suicide plasmid integration into this gene and characterized using phenotype assays, DNA microarrays containing 691 arrayed genes, and two-dimensional polyacrylamide gel electrophoresis. Physiological studies indicated that FUR1 was similar to the wild-type strain when they were compared for anaerobic growth and reduction of various electron acceptors. Transcription profiling, however, revealed that genes with predicted functions in electron transport, energy metabolism, transcriptional regulation, and oxidative stress protection were either repressed (ccoNQ, etrA, cytochrome b and c maturation-encoding genes, qor, yiaY, sodB, rpoH, phoB, and chvI) or induced (yggW, pdhC, prpC, aceE, fdhD, and ppc) in the fur mutant. Disruption of fur also resulted in derepression of genes (hxuC, alcC, fhuA, hemR, irgA, and ompW) putatively involved in iron uptake. This agreed with the finding that the fur mutant produced threefold-higher levels of siderophore than the wild-type strain under conditions of sufficient iron. Analysis of a subset of the FUR1 proteome (i.e., primarily soluble cytoplasmic and periplasmic proteins) indicated that 11 major protein species reproducibly showed significant (P < 0.05) differences in abundance relative to the wild type. Protein identification using mass spectrometry indicated that the expression of two of these proteins (SodB and AlcC) correlated with the microarray data. These results suggest a possible regulatory role of S. oneidensis MR-1 Fur in energy metabolism that extends the traditional model of Fur as a negative regulator of iron acquisition systems

    NICMOS Imaging of a Damped Lyman-alpha Absorber at z=1.89 toward LBQS 1210+1731 : Constraints on Size and Star Formation Rate

    We report results of a high-resolution imaging search (in rest frame H-α\alpha and optical continuum) for the galaxy associated with the damped Lyman-α\alpha (DLA) absorber at z=1.892z=1.892 toward the zem=2.543z_{em}=2.543 quasar LBQS 1210+1731, using HST/NICMOS. After PSF subtraction, a feature is seen in both the broad-band and narrow-band images, at a projected separation of 0.25\arcsec from the quasar. If associated with the DLA, the object would be 23\approx 2-3 h701h_{70}^{-1} kpc in size with a flux of 9.8±2.49.8 \pm 2.4 μ\muJy in the F160W filter, implying a luminosity at λcentral=5500\lambda_{central}=5500 {\AA} in the rest frame of 1.5×10101.5 \times 10^{10} h702h_{70}^{-2} L_{\odot} at z=1.89z=1.89, for q0=0.5q_{0}=0.5. However, no significant H-α\alpha emission is seen, suggesting a low star formation rate (SFR) (3 σ\sigma upper limit of 4.0 h702h_{70}^{-2} M_{\odot} yr1^{-1}), or very high dust obscuration. Alternatively, the object may be associated with the host galaxy of the quasar. H-band images obtained with the NICMOS camera 2 coronagraph show a much fainter structure 45\approx 4-5 h701h_{70}^{-1} kpc in size and containing four knots of continuum emission, located 0.7\arcsec away from the quasar. We have probed regions far closer to the quasar sight-line than in most previous studies of high-redshift intervening DLAs. The two objects we report mark the closest detected high-redshift DLA candidates yet to any quasar sight line. If the features in our images are associated with the DLA, they suggest faint, compact, somewhat clumpy objects rather than large, well-formed proto-galactic disks or spheroids.Comment: 52 pages of text, 19 figures, To be published in Astrophysical Journal (accepted Dec. 8, 1999

    A Descriptive Profile of Physical Education Teachers and Related Program Characteristics in Alberta

    A survey of teachers and principals in Alberta was conducted to gain a descriptive profile of who is leaching physical education (PE) and to assess the relationship between PE specialists and variables associated with program delivery. A probability-sampling procedure was used to obtain a representative sample of schools. In these schools nonprobability procedures were used to recruit teachers. A total of'480 teachers' and 162 principals' questionnaires were returned. Although 50% (n=236) of PE teachers in the sample were classified as PE specialists (i.e., had either a degree, major or minor, in PE or a closely related area), there was a significant gap in the number of PE classes being taught by division. Of the 1,219 PE classes surveyed in this study, PE specialists taught 49% and 55% of classes at the elementary levels (Divisions I & 11) compared with 91% of junior high (Division III) and 90% of secondary (Division TV) PE classes. Significant differences were found between PE specialists and non-PE specialists on a number of items including perceptions of preparedness, teaching enjoyment and competence to teach PE, the number of PE specialists across grade levels, and the percentage of time devoted to PE in the timetable. Implications with respect to implementing PE specialists across all grades and the need for future pedagogical research to investigate the effect of PE specialists are also discussed.Une enquête a été entreprise auprès d'enseignants et de directeurs d'école en Alberta dans le but d'établir un profil descriptif des enseignants d'éducation physique (EP) et d'évaluer le rapport entre les spécialistes en EP et les variables associées à l'exécution de programmes. On a eu recours aune méthode d'échantillonnage au hasard pour obtenir un échantillon représentatif des écoles. Par la suite, on y a employé des procédures non probabilistes pour recruter des enseignants. En tout, 642 questionnaires (480 provenant d'enseignants et 162 de directeurs) nous ont été renvoyés. Alors que 50% (n=236) des enseignants de EP de l'échantillon se classaient comme spécialistes en EP (c'est-à-dire qu'ils avaient soit un diplôme, une majeure ou une mineure en EP ou dans un domaine connexe), un écart notable existait dans le nombre de cours de EP enseignés par division. Des 1 219 cours de EP inclus dans l'enquête, à l'élémentaire, 49% (Division I) et 55% (Division II) d'entre eux étaient enseignés par des spécialistes en EP. Au secondaire premier cycle (Division III), 91% des cours étaient enseignés par des spécialistes en EP; au secondaire (Division IV), 90% des cours l'étaient. Des différences importantes distinguaient les spécialistes en EP des non spécialistes, y compris leurs perceptions quant à leur état de préparation, le plaisir qu'il retirait de l'enseignement de l'EP, leur compétence à le faire, le nombre de spécialistes en EP à tous les niveaux scolaires et le nombre d'heures consacrées aux cours de EP.     A systematic review of biomarkers for disease progression in Parkinson's disease

    This article presents independent research funded by the National Institute for Health Research (NIHR) under its Programme Grants for Applied Research Programme (Grant Reference Number RP-PG-0707-10124).BACKGROUND: Using surrogate biomarkers for disease progression as endpoints in neuroprotective clinical trials may help differentiate symptomatic effects of potential neuroprotective agents from true disease-modifying effects. A systematic review was undertaken to determine what biomarkers for disease progression in Parkinson's disease (PD) exist. METHODS: MEDLINE and EMBASE (1950-2010) were searched using five search strategies. Abstracts were assessed to identify papers meriting review in full. Studies of participants with idiopathic PD diagnosed by formal criteria or clearly described clinical means were included. We made no restriction on age, disease duration, drug treatment, or study design. We included studies which attempted to draw associations between any tests used to investigate disease progression and any clinical measures of disease progression. The electronic search was validated by hand-searching the two journals from which most included articles came. RESULTS: 183 studies were included: 163 (89%) cross-sectional, 20 (11%) longitudinal. The electronic search strategy had a sensitivity of 71.4% (95% CI 51.1-86.0) and a specificity of 97.1% (95% CI 96.5-97.7). In longitudinal studies median follow-up was 2.0 years (IQR 1.1-3.5). Included studies were generally poor quality--cross-sectional with small numbers of participants, applying excessive inclusion/exclusion criteria, with flawed methodologies and simplistic statistical analyses. CONCLUSION: We found insufficient evidence to recommend the use of any biomarker for disease progression in PD clinical trials, which may simply reflect the poor quality of research in this area. We therefore present a provisional 'roadmap' for conducting future disease progression biomarker studies, and recommend new quality criteria by which future studies may be judged.Publisher PDFPeer reviewe

    Potassium Fertilizer Rate Recommendations: Does Accounting for Soil Stock of Potassium Matter?

    Profitability, yield, and fertilizer use are compared across three different potassium (K) fertilizer rate recommendation ideologies. Existing agronomic, “build and maintain” rate recommendations (KE) are compared to profit-maximizing rates with and without taking long-run soil-test K (STK) implications into account. Regardless of starting STK, K use equilibrated over the course of 3 years irrespective of ideology. Since taking long-run STK into account did not alter ending STK and only led to a miniscule yield effect, we encourage producers to use annual profit-maximizing K rates that were 3–11% lower than KE rates and generated more profit with minimal yield loss
