    Ontogeny of synaptophysin and synaptoporin in the central nervous system

    The expression of the synaptic vesicle antigens synaptophysin (SY) and synaptoporin (SO) was studied in the rat striatum, which contains a nearly homogeneous population of GABAergic neurons. In situ hybridization revealed high levels of SY transcripts in the striatal anlage from embryonic day (E) 14 until birth. In contrast. SO hybridization signals were low, and no immunoreactive cell bodies were detected at these stages of development. At E 14, SY-immunoreactivity was restricted to perikarya. In later prenatal stages of development SY-immunoreactivity appeared in puncta (identified as terminals containing immunostained synaptic vesicles), fibers, thick fiber bundles and ‘patches’. In postnatal and adult animals, perikarya of striatal neurons exhibited immunoreaction for SO; ultrastructurally SO antigen was found in the Golgi apparatus and in multivesicular bodies. SO-positive boutons were rare in the striatum. In the neuropil, numerous presynaptic terminals positive for SY were observed. Our data indicate that the expression of synaptic vesicle proteins in GABAergic neurons of the striatum is developmentally regulated. Whereas SY is prevalent during embryonic development, SO is the major synaptic vesicle antigen expressed postnatally by striatal neurons which project to the globus pallidus and the substantia nigra.     <p>Abstract</p> <p>Background</p> <p>Arsenic is a toxic and highly abundant metalloid that endangers human health through drinking water and the food chain. The most common forms of arsenic in the environment are arsenate (As(V)) and arsenite (As(III)). As(V) is a non-functional phosphate analog that enters the food chain via plant phosphate transporters. Inside cells, As(V) becomes reduced to As(III) for subsequent extrusion or compartmentation. Although much is known about As(III) transport and handling in microbes and mammals, the transport systems for As(III) have not yet been characterized in plants.</p> <p>Results</p> <p>Here we show that the Nodulin26-like Intrinsic Proteins (NIPs) AtNIP5;1 and AtNIP6;1 from <it>Arabidopsis thaliana</it>, OsNIP2;1 and OsNIP3;2 from <it>Oryza sativa</it>, and LjNIP5;1 and LjNIP6;1 from <it>Lotus japonicus </it>are bi-directional As(III) channels. Expression of these NIPs sensitized yeast cells to As(III) and antimonite (Sb(III)), and direct transport assays confirmed their ability to facilitate As(III) transport across cell membranes. On medium containing As(V), expression of the same NIPs improved yeast growth, probably due to increased As(III) efflux. Our data furthermore provide evidence that NIPs can discriminate between highly similar substrates and that they may have differential preferences in the direction of transport. A subgroup of As(III) permeable channels that group together in a phylogenetic tree required N-terminal truncation for functional expression in yeast.</p> <p>Conclusion</p> <p>This is the first molecular identification of plant As(III) transport systems and we propose that metalloid transport through NIPs is a conserved and ancient feature. Our observations are potentially of great importance for improved remediation and tolerance of plants, and may provide a key to the development of low arsenic crops for food production.</p

    Differential expression of synaptophysin and synaptoporin during pre- and postnatal development of the hippocampal network

    The closely related synaptic vesicle membrane proteins synaptophysin and synaptoporin are abundant in the hippocampal formation of the adult rat. But the prenatal hippocampal formation contains only synaptophysin, which is first detected at embryonic day 17 (E17) in perikarya and axons of the pyramidal neurons. At E21 synaptophysin immunoreactivity extends into the apical dendrites of these cells and in newly formed terminals contacting these dendrites. The transient presence of synaptophysin in axons and dendrites suggests a functional involvement of synaptophysin in fibre outgrowth of developing pyramidal neurons. Synaptoporin expression parallels the formation of dentate granule cell synaptic contacts with pyramidal neurons: the amount of hippocampal synaptoporin, determined in immunoblots and by synaptoporin immunostaining of developing mossy fibre terminals, increases during the first postnatal week. Moreover, in the adult, synaptoporin is found exclusively in the mossy fibre terminals present in the hilar region of the dentate gyrus and the regio inferior of the cornu ammonis. In contrast, synaptophysin is present in all synaptic fields of the hippocampal formation, including the mossy fibre terminals, where it colocalizes with synaptoporin in the same boutons. Our data indicate that granule neuron terminals differ from all other terminals of the hippocampal formation by the presence of both synaptoporin and synaptophysin. This difference, observed in the earliest synaptic contacts in the postnatal hippocampus and persisting into adult life, suggests distinct functions of synaptoporin in these nerve terminals

    Quasi-free Compton Scattering and the Polarizabilities of the Neutron

    Differential cross sections for quasi-free Compton scattering from the proton and neutron bound in the deuteron have been measured using the Glasgow/Mainz tagging spectrometer at the Mainz MAMI accelerator together with the Mainz 48 cm ⊘\oslash ×\times 64 cm NaI(Tl) photon detector and the G\"ottingen SENECA recoil detector. The data cover photon energies ranging from 200 MeV to 400 MeV at θγLAB=136.2∘\theta^{LAB}_\gamma=136.2^\circ. Liquid deuterium and hydrogen targets allowed direct comparison of free and quasi-free scattering from the proton. The neutron detection efficiency of the SENECA detector was measured via the reaction p(γ,π+n)p(\gamma,\pi^+ n). The "free" proton Compton scattering cross sections extracted from the bound proton data are in reasonable agreement with those for the free proton which gives confidence in the method to extract the differential cross section for free scattering from quasi-free data. Differential cross sections on the free neutron have been extracted and the difference of the electromagnetic polarizabilities of the neutron have been obtained to be α−β=9.8±3.6(stat)12.1.1(syst)±2.2(model)\alpha-\beta= 9.8\pm 3.6(stat){}^{2.1}_1.1(syst)\pm 2.2(model) in units 10−4fm310^{-4}fm^3. In combination with the polarizability sum α+β=15.2±0.5\alpha +\beta=15.2\pm 0.5 deduced from photoabsorption data, the neutron electric and magnetic polarizabilities, αn=12.5±1.8(stat)−0.6+1.1±1.1(model)\alpha_n=12.5\pm 1.8(stat){}^{+1.1}_{-0.6}\pm 1.1(model) and βn=2.7∓1.8(stat)−1.1+0.6(syst)∓1.1(model)\beta_n=2.7\mp 1.8(stat){}^{+0.6}_{-1.1}(syst)\mp 1.1(model) are obtained. The backward spin polarizability of the neutron was determined to be γπ(n)=(58.6±4.0)×10−4fm4\gamma^{(n)}_\pi=(58.6\pm 4.0)\times 10^{-4}fm^4

    Quasi-free π0\pi^0 Photoproduction from the Bound Nucleon

    Differential cross-sections for quasi-free π0\pi^0 photoproduction from the proton and neutron bound in the deuteron have been measured for Eγ=200−400E_\gamma= 200 - 400 MeV at θγlab=136.2∘\theta^{\rm lab}_\gamma = 136.2^\circ usind the Glasgow photon tagger at MAMI, the Mainz 48 cm ∅\varnothing ×\times 64 cm NaI(Tl) photon detector and the G\"ottingen SENECA recoil detector. For the proton measurements made with both liquid deuterium and liquid hydrogen targets allow direct comparison of "free" π0\pi^0 photoproduction cross-sections as extracted from the bound proton data with experimental free cross sections which are found to be in reasonable agreement below 320 MeV. At higher energies the "free" cross sections extracted from quasifree data are significantly smaller than the experimental free cross sections and theoretical predictions based on multipole analysis. For the first time, "free" neutron cross sections have been extracted in the Δ\Delta-region. They are also in agreement with the predictions from multipole analysis up to 320 MeV and significantly smaller at higher photon energies

    Neutron skin of 208^{208}Pb from Coherent Pion Photoproduction

    Information on the size and shape of the neutron skin on 208^{208}Pb has been extracted from coherent pion photoproduction cross sections measured using the Crystal Ball together with the Glasgow tagger at the MAMI electron beam facility. On exploitation of an interpolated fit of a theoretical model to the measured cross sections the half-height radius and diffuseness of the neutron distribution are found to be 6.70±0.03(stat)\pm 0.03(stat) fm and 0.55±0.01(stat)\pm 0.01(stat)−0.03+0.02(sys)^{+0.02}_{-0.03}(sys) fm respectively, corresponding to a neutron skin thickness Δrnp\Delta r_{np}=0.15±0.03(stat)\pm 0.03(stat)−0.03+0.01(sys)^{+0.01}_{-0.03}(sys) fm. The results give the first successful extraction of a neutron skin with an electromagnetic probe and indicate the skin of 208^{208}Pb has a halo character. The measurement provides valuable new constraints on both the structure of nuclei and the equation of state for neutron-rich matter.Comment: 4 figures 5 pages. Version submitted to journal.     Measuring Compton scattered photons and recoil neutrons in coincidence, quasi-free Compton scattering by the neutron has been investigated at MAMI (Mainz) at thetaγlab=136otheta^{lab}_\gamma=136^o in an energy range from 200 to 400 MeV. From the data a polarizability difference of αn−βn=9.8±3.6(stat)−1.1+2.1(syst)±2.2(model)\alpha_n - \beta_n = 9.8 \pm 3.6(stat)^{+2.1}_{-1.1}(syst)\pm 2.2(model) in units of 10−4fm310^{-4}fm^3 has been determined. In combination with the polarizability sum αn+βn=15.2±0.5\alpha_n+\beta_n= 15.2\pm 0.5 deduced from photo absorption data, the neutron electric and magnetic polarizabilities, αn=12.5±1.8(stat)−0.6+1.1(syst)±1.1(model)\alpha_n=12.5\pm 1.8(stat)^{+1.1}_{-0.6}(syst)\pm 1.1(model) and βn=2.7∓1.8(stat)−1.1+0.6(syst)∓1.1(model)\beta_n = 2.7\mp 1.8(stat)^{+0.6}_{-1.1}(syst)\mp 1.1(model), are obtained

    The isospin structure of photoproduction of pi-eta pairs from the nucleon in the threshold region

    Photoproduction of πη\pi\eta-pairs from nucleons has been investigated from threshold up to incident photon energies of ≈\approx~1.4~GeV. The quasi-free reactions γp→pπ0η\gamma p\rightarrow p\pi^0\eta, γn→nπ0η\gamma n\rightarrow n\pi^0\eta, γp→nπ+η\gamma p\rightarrow n\pi^+\eta, and γn→pπ−η\gamma n\rightarrow p\pi^-\eta were for the first time measured from nucleons bound in the deuteron. The corresponding reactions from a free-proton target were also studied to investigate final-state interaction effects (for neutral pions the free-proton results could be compared to previous measurements; the γp→nπ+η\gamma p\rightarrow n\pi^+\eta reaction was measured for the first time). For the π0η\pi^0\eta final state coherent production via the γd→dπ0η\gamma d\rightarrow d\pi^0\eta reaction was also investigated. The experiments were performed at the tagged photon beam of the Mainz MAMI accelerator using an almost 4π4\pi coverage electromagnetic calorimeter composed of the Crystal Ball and TAPS detectors. The total cross sections for the four different final states obey the relation σ(pπ0η)\sigma(p\pi^0\eta) ≈\approx σ(nπ0η)\sigma(n\pi^0\eta) ≈\approx 2σ(pπ−η)2\sigma(p\pi^-\eta) ≈\approx 2σ(nπ+η)2\sigma(n\pi^+\eta) as expected for a dominant contribution from a Δ⋆→ηΔ(1232)→πηN\Delta^{\star}\rightarrow\eta\Delta(1232)\rightarrow\pi\eta N reaction chain, which is also supported by the shapes of the invariant-mass distributions of nucleon-meson and π\pi-η\eta pairs. The experimental results are compared to the predictions from an isobar reaction model.Comment: accepted for publication in Phys. Lett.
