Synergistische Effekte von NO und Progesteron auf die Differenzierung des Endometriums zum Zeitpunkt der Implantation
Nach der Fertilisation ist die Implantation der Blastocyste in das Endometrium
eine notwendige Voraussetzung fĂĽr eine erfolgreiche Schwangerschaft.
Progesteron, das schwangerschaftserhaltende maternale Steroidhormon,
kontrolliert nicht nur die Transformation des Endometriums in die rezeptive
Phase und ermöglicht damit der Blastocyste zu implantieren, sondern ist auch
verantwortlich fĂĽr die Transformation der endometrialen Fibroblasten in
Deziduazellen. Das kompakte Deziduagewebe ist auch an der Kontrolle der
Invasion der Trophoblastzellen beteiligt, die letztendlich fĂĽr die Entstehung eines
feto-maternalen Austauschorgans der Plazenta verantwortlich sind.
Unter vielen anderen Faktoren induziert der erhöhte Progesteronspiegel während
und nach der Implantation eine erhöhte Expression von induzierter Nitric Oxide
Synthase (iNOS) im Endometrium und führt damit zu einer Erhöhung des NO
Spiegels. Welche Rolle NO vor allem während und nach der Implantation der
Blastocyste zukommt ist unbekannt. Um die zellbiologischen Grundlagen der
Bedeutung von iNOS zu erarbeiten und seine mögliche Interaktion mit
Progesteron zu evaluieren wurden im Rahmen der Promotionsarbeit gravide
Ratten in der Präimplantationsphase (d1-d4 p.c.), Periimplantationsphase (d5-d8)
und in der Postimplantationsphase (d7-d10 p.c.) mit jeweils dem iNOS Inhibitor
(Aminoguanidin) und dem Antigestagen (Onapriston) sowohl in der
Einzelapplikation, als auch in der Kombination behandelt.
Die Applikation der Einzeldosierungen in der Präimplantationsphase führte bei
Aminoguanidin (iNOS Inhibitor) zu keinem und bei Onapriston (Antigestagen)
zu einem geringen Effekt auf die Implantationsrate. Bei allen Tieren hingegen,
die in der Kombination behandelt wurden, zeigte sich eine drastische Erhöhung
des Onapriston Effektes durch den iNOS Inhibitor der zu einer vollständigen
Hemmung der Gravidität führte. Diese drastische Auswirkung auf die Gravidität
ist auf einen Effekt auf die Präimplantationsembryonen zurückzuführen. Im
Synergismus der beiden Substanzen entwickeln sich nur vereinzelt Blastocysten
die zudem morphologisch verändert aussehen.
In der Peri- als auch in der Postimplantationsphase zeigte ebenfalls der iNOS
Inhibitor keinen und das Antigestagen nur einen marginalen Effekt auf den Erhalt
der Gravidität.. Die Kombination beider Substanzen führte sowohl in der Peri-als
auch in der Postimplantationsphase zu einem Synergismus, der allerdings nur
nach Applikation in der Postimplantationsphase von d7-d9 p.c. zu einer
vollständigen Hemmung der Gravidität führte. Diese Hemmung resultiert in einer
Störung des Dezidualisierungsprozesses, bei dem sowohl die
Differenzierungsvorgänge betroffen waren, als auch Markergene von
Signalkaskaden der Deziduazellen verändert wurden.
So zeigte sich ein gestörtes Expressionmuster des Differenzierungsmarkers
Desmin, der nicht mehr in allen Deziduazellen vorhanden war. Zusätzlich wurde
eine Beeinträchtigung der Sinusiodausbildung in der "junctionalen zone" und in
der mesometralen Seite nachgewiesen, die Sinusoide bleiben englumig. Zu einem
späteren Zeitpunkt lässt sich in der "junctionalen zone" zusätzlich ein
Proliferationsstop, und apoptotische Zellen nachweisen. Dies deutet darauf hin,
dass dieser Bereich der Dezidua besonders sensibel auf die Kombination beider
Substanzen reagiert.
Die Applikation des iNOS Inhibitors in Kombination mit dem Antigestagen fĂĽhrt
während der Dezidualisierung zu einer veränderten Expression von COX II und
NFkappa B als auch der aktivierten MAPkinase ERK1&2. Die
Einzeldosierungen zeigen hingegen keine Auswirkungen auf diese
Signaltransduktionsmarker.
Der Verlust von COX II nach Applikation beider Substanzen im Bereich der
"junctionalen zone" deutet auf eine Beteiligung der Prostaglandine bei der
Dilatation der dezidualen Gefäße hin. Unter der synergistischen Wirkung der
Substanzen kommt es in den Deziduazellen zu einer Translokation des
Transkriptionsfaktor NFkappaB in den Kern mit folgendem Abbau. Die
aktivierte MAPkinase ERK1&2 konnte nach Gabe der Substanzen in der
gesamten Dezidua nicht mehr nachgewiesen werden, wird aber bei abnehmender
Wirkkonzentration beider Substanzen auf der mesometralen Seite reexprimiert.
Die Reexpression des aktivierten diphosphorylierten ERK1&2 weiĂźt auf eine
direkte Beteiligung von NO und Progesterons an der Aktivierung des MAPkinase
Signalweges in Abhängigkeit von der Konzentration der Substanzen im Blut hin.
Wie der Einsatz des iNOS-Inhibitors und des Antigestagens aufgezeigt haben,
liegt der synergistische Effekt von NO und Progesteron in der Induzierung und
Aufrecherhaltung von verstärkten Phosphorylierungsvorgängen, die für die
Differenzierungskaskaden der Fibroblasten in Deziduazellen erforderlich sind
Runoff treatment with aspen wood
Runoff from roadways and urban areas is often contaminated with heavy metals such as zinc and copper. The use of aspen wood (Populus tremula) was investigated as a potential sorbentfor these metal contaminated runoff waters. The objective was to design an alternative runoff treatment technology based on a renewable resource, i.e. wood. Laboratory isotherms and column experiments were conducted in order to investigate the sorption and desorption of dissolved copper and zinc to and from aspen wood fibers. Sorption ofCu(II) and Zn(II) followed non-linear Freundlich isotherms. The aspen wood-water partition coefficient values revealed that Cu(II) had a greater tendency to sorb to the aspen wood than Zn(II). The column experiments indicated that, at least initially, both metals were being rapidly sorbed. During later stages of the sorption experiments, slower, but steady transport of the metal ions into the wood fibers appeared to be the dominating removal mechanism. Desorption of copper with E-pure water was faster than zinc, but the total amount of metal mass released from the wood was small in both cases, i.e., only 2% to 4% of the amount sorbed. Fractionation of milk proteins on pilot scale with particular focus on β-casein
The aim of this study was to increase the yield and purity of casein fractions at pilot scale and determine the main process parameters influencing the isolation of β-casein, such as cold extraction time, separation speed. The fractions were obtained from micellar casein by means of selective precipitation following separation by a nozzle centrifuge. The purities, which were calculated based on the total casein content of the β-casein fraction, and the yields achieved for β-casein ranged from 68.7 to 89.6% and from 10 to 32%, respectively. For the other two fractions, αS- and κ-casein, the obtained purities were 61% and 43%, respectively. Using the proposed isolation method, high purities for the β-casein fraction were achievable; however, the other two fractions, αS- and κ-casein, need further improvement

Electrophysiological, cognitive and clinical profiles of at -risk mental state : the longitudinal Minds in Transition (MinT) study
The onset of schizophrenia is typically preceded by a prodromal period lasting several years during which sub-threshold symptoms may be identified retrospectively. Clinical interviews are currently used to identify individuals who have an ultra-high risk (UHR) of developing a psychotic illness with a view to provision of interventions that prevent, delay or reduce severity of future mental health issues. The utility of bio-markers as an adjunct in the identification of UHR individuals is not yet established. Several event-related potential measures, especially mismatch-negativity (MMN), have been identified as potential biomarkers for schizophrenia. In this 12-month longitudinal study, demographic, clinical and neurocognitive data were acquired from 103 UHR and 65 healthy controls, of whom 81 UHR and 61 controls provided valid EEG data during passive and active auditory tasks at baseline. Despite widespread differences between UHR and controls on other measures [JT1], ERPs did not differ between these groups. MMN amplitude was modulated by age and the effects of cannabis, which is recognized as a risk factor for development of psychosis. Of 67 UHR at the 12-month follow-up, 7 (10%) had transitioned to a psychotic illness. The statistical power to detect differences between those who did or did not transition was limited by the lower than expected transition rate. ERPs did not predict transition, with trends in the opposite direction to that predicted. In exploratory analysis, the strongest predictors of transition were measures of verbal memory and subjective emotional disturbance. This study’s findings suggest caution is required when interpreting recent reports of reduced MMN and P3b in UHR groups. These findings are consistent with indications that abnormalities in these components are indicators of biological changes associated with illness onset as opposed to general vulnerability factors.1 page(s
Mismatch negativity and cognitive and functional impairment in the schizophrenia prodrome. Preliminary results from the Minds in Transition (MinT) study
This project aims to gain a greater understanding of the changes in the brains of young people when they are developing a severe mental illness and to investigate the utility of an electrophysiological index of auditory sensory memory (mismatch negativity) as a predictor of transition to a schizophrenia spectrum disorder. Young people attending youth mental health centres in Newcastle, Orange and Sydney will be recruited over a 3.5-year period. Those meeting ultra-high risk (UHR) criteria as determined according to the Comprehensive Assessment of At Risk Mental States, will be followed up over a 12-month period. At baseline clinical, electrophysiological recordings, high-resolution structural and DTI magnetic resonance brain images, neurocognition and social cognition data are collected, with follow-up data collected after 12-months. This preliminary report describes correlations between baseline MMN amplitude data and symptoms and cognitive functioning. To date, baseline data have been obtained on 40 UHR participants. These preliminary results indicate associations between duration MMN and functional status, duration MMN and auditory verbal memory and frequency MMN and cognitive flexibility. Results suggest duration MMN may be tapping into underlying temporal lobe mechanisms, while frequency MMN may be tapping into underlying frontal lobe mechanisms and such deficits are occurring during the prodrome phase.2 page(s
