    Ecological, Social and Economic Aspects of Italian Marine Spearfishing Tournaments (2009-2020)

    Marine recreational spearfishing is practiced by a small proportion of recreational fishers and ecological, social and economic aspects of such activities are poorly understood with respect to recreational angling. The Italian Federation of Sport Fishing and Underwater Activities (FIPSAS) started in 2009 the collection of catch data related to Italian spearfishing tournaments. Here, we provide the first snapshot of this catch data regarding 48 tournaments organized in 29 different Italian localities from 2009 to 2020. We also developed a survey to collect social and economic information that has been administrated to participants to the tournaments. We retrieved 8843 specimens caught by spearfishers, which accounted for 33 species and a total biomass of 5665 kg (mean individual mass ± standard error; 0.64 ± 0.01 kg). The five most frequently species represented 78% of the specimens caught (white seabream, Diplodus sargus 48%; brown wrasse, Labrus merula 12%; brown meagre, Sciaena umbra 9%, green wrasse, Labrus viridis 5%; salema, Sarpa salpa 4%). The overall catch per unit effort for all the tournaments was 0.47 ± 0.01 kg/spearfisher/h, and we showed interesting effects of type of tournaments and type of displacement on it. We also assessed the accuracy of participants to catch fish in accordance to tournaments-specific minimum weight limits. The five most caught species showed that spearfishers committed errors around 50 g below the weight limit. The survey showed that spearfishers participating to tournaments are trophy-fish oriented (i.e., they would rather catch one or two big fish than ten smaller fish), but not particularly catch oriented (i.e., they are just as happy if they do not shoot the fish they see and a fishing trip can be successful even if no fish are caught). Regarding the motives for fishing, they fish for experiencing adventure, excitement and new and different things as well as to be underwater. Participants spent from 2 to 26 days in the tournament locations (median equal to 7 days), which implies a total expenditure to solely participate to the tournaments ranging from 800 to 3500 Euros (mean ± standard deviation; 1800 ± 591 euros). This study represents the first ecological, social and economic baseline knowledge for an integrative management of contemporary spearfishing tournament

    Electrons on a sphere in disorder potential

    We investigate, both analytically and numerically, the behavior of the electron gas on a sphere in the presence of point-like impurities. We find a criterion when the disorder can be regarded as small one and the main effect is the broadening of rotational multiplets. In the latter regime the statistics of one impurity-induced band is studied numerically. The energy level spacing distribution function follows the law P(s) ~ s exp(-a s^b) with 1<b<2.     Ex vivo model predicted in vivo efficacy of CFTR modulator therapy in a child with rare genotype

    Background: New drugs that target the basic defect in cystic fibrosis (CF) patients may now be used in a large number of patients carrying responsive mutations. Nevertheless, further research is needed to extend the benefit of these treatments to patients with rare mutations that are still uncharacterized in vitro and that are not included in clinical trials. For this purpose, ex vivo models are necessary to preliminary assessing the effect of CFTR modulators in these cases. Method: We report the clinical effectiveness of lumacaftor/ivacaftor therapy prescribed to a CF child with a rare genetic profile (p.Phe508del/p.Gly970Asp) after testing the drug on nasal epithelial cells. We observed a significant drop of the sweat chloride value, as of the lung clearance index. A longer follow-up period is needed to define the effects of therapy on pancreatic status, although an increase of the fecal elastase values was found. Conclusion: Drug response obtained on nasal epithelial cells correlates with changes in vivo therapeutic endpoints and can be a predictor of clinical efficacy of novel drugs especially in pediatric patients

    Clinical and Genotypical Features of False-Negative Patients in 26 Years of Cystic Fibrosis Neonatal Screening in Tuscany, Italy.

    Cystic fibrosis (CF) is a life-threatening and common genetic disorder. Cystic fibrosis newborn screening (CF NBS) has been implemented in many countries over the last 30 years, becoming a widely accepted public health strategy in economically developed countries. False-negative (FN) cases can occur after CF NBS, with the number depending on the method. We evaluated the delayed diagnosis of CF, identifying the patients who had false-negative CF NBS results over 26 years (1992-2018) in Tuscany, Italy. The introduction of DNA analysis to the newborn screening protocol improved the sensitivity of the test and reduced the FNs. Our experience showed that, overall, at least 8.7% of cases of CF received FNs (18 cases) and were diagnosed later, with an average age of 6.6 years (range: 4 months to 22 years). Respiratory symptoms and salt-loss syndrome (metabolic hypochloremic alkalosis) are suggestive symptoms of CF and were commons events in FN patients. In Tuscany, a region with a high CFTR allelic heterogeneity, the salt-loss syndrome was a common event in FNs. Therefore, we provided evidence to support the claim that the FN patients had CFTR mutations rarer compared with the true-positive cases. We underline the importance of vigilance toward clinical manifestations suggestive of CF on the part of the primary care providers and hospital physicians in a region with an efficient newborn screening program

    Occurrence of grapevine virus A (GVA) and other closteroviruses in Tunisian grapevines affected by leafroll disease

    Vorkommen von Grapevine-Virus A (GVA) und anderen Closteroviren in blattrollkranken tunesischen RebenReben, die aus den Hauptweinbaugebieten Tunesiens stammten, wurden auf die Anwesenheit von Closteroviren hin überprüft. Während in keiner der symptomfreien Reben Viruspartikel entdeckt wurden, enthielten alle Reben mit Blattrollsymptomen - außer zweien - Closteroviruspartikel, die durch IEM (immune electron microscopy) in konzentrierten Blattextrakten oder unmittelbar in Rohsaft durch ISEM (immunosorbent electron microscopy) identifiziert wurden. Alle vier derzeit bekannten Closteroviren (GClV-1, GClV-2, GClV-3 und GVA) waren, meistens im Gemisch, in Reben mit Blattrollsymptomen vorhanden. GClV-3 und GVA wurden in 77 bzw. 50 % der geprüften Reben entdeckt. Ein tunesisches Isolat von GVA, das durch Planococcus citri auf krautige Testpflanzen übertragen wurde, unterschied sich in biologischer Hinsicht, aber nicht in den charakteristischen physikalisch-chemischen und serologischen Eigenschaften von zwei italienischen Isolaten desselben Virus

    Are we ready for scaling up restoration actions? An insight from Mediterranean macroalgal canopies

    Extensive loss of macroalgal forests advocates for large-scale restoration interventions, to compensate habitat degradation and recover the associated ecological functions and services. Yet, restoration attempts have generally been limited to small spatial extensions, with the principal aim of developing efficient restoration techniques. Here, the success of outplanting Cystoseira amentacea v. stricta germlings cultured in aquaria was experimentally explored at a scale of tens of kms, by means of a multifactorial experimental design. In the intertidal rocky shores of SE Italy, locations with a continuous distribution for hundreds of meters or with few thalli forming patches of few centimeters of C. amentacea canopy were selected. In each location, the effects of adult conspecifics and the exclusion of macrograzers (salema fish and sea urchins) on the survival of germlings were tested. We evaluated the most critical determinants of mortality for germlings, including the overlooked pressure of mesograzers (e.g. amphipods, small mollusks, polychaetes). Despite the high mortality observed during outplanting and early settlement stages, survival of C. amentacea germlings was consistently favored by the exclusion of macrograzers, while the presence of adult conspecifics had no effects. In addition, the cost analysis of the interventions showed the feasibility of the ex-situ method, representing an essential tool for preserving Cystoseira forests. Large scale restoration is possible but requires baseline information with an in-depth knowledge of the species ecology and of the areas to be restored, together with the development of specific cultivation protocols to make consistently efficient restoration interventions

    Cardiovascular Response to Intraneural Right Vagus Nerve Stimulation in Adult Minipig.

    Objective: This study explored intraneural stimulation of the right thoracic vagus nerve (VN) in sexually mature male minipigs to modulate safe heart rate and blood pressure response. Material and methods: We employed an intraneural electrode designed for the VN of pigs to perform VN stimulation (VNS). This was delivered using different numbers of contacts on the electrode and different stimulation parameters (amplitude, frequency, and pulse width), identifying the most suitable stimulation configuration. All the parameter ranges had been selected from a computational cardiovascular system model. Results: Clinically relevant responses were observed when stimulating with low current intensities and relatively low frequencies delivered with a single contact. Selecting a biphasic, charge-balanced square wave for VNS with a current amplitude of 500 μA, frequency of 10 Hz, and pulse width of 200 μs, we obtained heart rate reduction of 7.67 ± 5.19 beats per minute, systolic pressure reduction of 5.75 ± 2.59 mmHg, and diastolic pressure reduction of 3.39 ± 1.44 mmHg. Conclusion: Heart rate modulation was obtained without inducing any observable adverse effects, underlining the high selectivity of the intraneural approach

    Influence of Heart Rate on Left and Right Ventricular Longitudinal Strain in Patients with Chronic Heart Failure

    Over the past years, a number of studies have demonstrated the relevance of strain assessed by two-dimensional speckle tracking echocardiography (STE) in evaluating ventricular function. The aim of this study was to analyze changes in left (LV) and right ventricular (RV) longitudinal strain associated with variations of heart rate (HR) in participants with and without chronic heart failure (CHF). We enrolled 45 patients, 38 of these diagnosed with CHF and carrying an implantable cardioverter defibrillator, and seven patients with pacemakers and without CHF. The frequency of atrial stimulation was increased to 90 beats/min and an echocardiogram was performed at each increase of 10 beats/min. Global LV and RV longitudinal strain (LVGLS and RVGLS, respectively) and RV free wall longitudinal strain (RVfwLS) were calculated at each HR. When analyzed as continuous variables, significant reductions in LVGLS were detected at higher HRs, whereas improvements in both RVGLS and RVfwLS were observed. Patients with a worsening of LVGLS (76% overall) were more likely to present lower baseline LV function. Only a few patients (18% for RVGLS and 16% for RVfwLS) exhibited HR-related deteriorations of RV strain measures, which was associated with lower levels of baseline RV function and higher pulmonary systolic pressures. Finally, 21 (47%) and 25 (56%) participants responded with improvements in RVGLS and RVfwLS, respectively. Our findings revealed heterogeneous RV and LV responses to increases in HR. These findings might ultimately be used to optimize cardiac functionality in patients diagnosed with CHF

    Pharmacological And Genetic Reversal Of Age-Dependent Cognitive Deficits Attributable To Decreased Presenilin Function

    Alzheimer\u27s disease (AD) is the leading cause of cognitive loss and neurodegeneration in the developed world. Although its genetic and environmental causes are not generally known, familial forms of the disease (FAD) are attributable to mutations in a single copy of the Presenilin (PS) and amyloid precursor protein genes. The dominant inheritance pattern of FAD indicates that it may be attributable to gain or change of function mutations. Studies of FAD-linked forms of presenilin (psn) in model organisms, however, indicate that they are loss of function, leading to the possibility that a reduction in PS activity might contribute to FAD and that proper psn levels are important for maintaining normal cognition throughout life. To explore this issue further, we have tested the effect of reducing psn activity during aging in Drosophila melanogaster males. We have found that flies in which the dosage of psn function is reduced by 50% display age-onset impairments in learning and memory. Treatment with metabotropic glutamate receptor (mGluR) antagonists or lithium during the aging process prevented the onset of these deficits, and treatment of aged flies reversed the age-dependent deficits. Genetic reduction of Drosophila metabotropic glutamate receptor (DmGluRA), the inositol trisphosphate receptor (InsP(3)R), or inositol polyphosphate 1-phosphatase also prevented these age-onset cognitive deficits. These findings suggest that reduced psn activity may contribute to the age-onset cognitive loss observed with FAD. They also indicate that enhanced mGluR signaling and calcium release regulated by InsP(3)R as underlying causes of the age-dependent cognitive phenotypes observed when psn activity is reduced
