    Evaluation of Microparticulate Ovarian Cancer Vaccine via Transdermal Route of Delivery

    Ovarian cancer is the fifth most commonly occurring malignancy in women, with the highest mortality rate among all the gynecological tumors. Microparticulate vaccine can serve as an immunotherapeutic approach with a promising antigenic delivery system without a need for conventional adjuvants. In this study, a microparticulate vaccine using whole cell lysate of a murine ovarian cancer cell line, ID8 was prepared by spray drying. Further, the effect of interleukins (ILs) such as IL-2 and IL-12 was evaluated in a separate study group by administering them with vaccine particles to enhance the immune response. The vaccine microparticles were administered to C57BL/6 female mice via transdermal alone and in combination with the oral route. The transdermal vaccine was delivered using a metallic microneedle device, AdminPen™. Orally administered microparticles also included an M-cell targeting ligand, Aleuria aurantia lectin, to enhance the targeted uptake from microfold cells (M-cells) in Peyer\u27s patches of small intestine. In case of combination of routes, mice were given 5 transdermal doses and 5 oral doses administered alternatively, beginning with transdermal dose. At the end of vaccination, mice were challenged with live tumor cells. Vaccine alone resulted in around 1.5 times tumor suppression in case of transdermal and combination of routes at the end of 15th week when compared to controls. Inclusion of interleukins resulted in 3 times tumor suppression when administered with transdermal vaccine and around 9 times tumor suppression for the combination route of delivery in comparison to controls. These results were further potentiated by serum IgG, IgG1 and IgG2a titers. Moreover, CD8+ T-cell, CD4+ T-cell and NK (natural killer) cell populations in splenocytes were elevated in case of vaccinated mice. Thus, vaccine microparticles could trigger humoral as well as cellular immune response when administered transdermally and via combination of route of delivery. However overall, vaccine administered with interleukins, via combination of route, was found to be the most efficacious to suppress the tumor growth and lead to a protective immune response

    Exploring Birthparent’s Experiences of Creating an Adoption Plan for Their Children with Special Needs

    Very little information is available regarding the experience and needs of families who create an adoption plan for their child with disability. The purpose of this study was to learn more about the experiences of birthparents who created an adoption plan after the diagnosis of Down syndrome so as to understand their needs during the process. Birthparents were invited to participate in the study through National Down Syndrome Adoption Network (NDSAN) by membership emails. Information about the study was made available on the organization’s website and social media pages. Potential participants also learned about the study from one of the co-authors of the study. Participant inclusion criteria included individuals who were 18 years of age and older, who had created an adoption plan for their baby with Down syndrome, and who were willing to participate in the telephone interview. Semi-structured telephone interviews were conducted with five mothers who had created adoption plans for their children with Down syndrome. Conventional content analysis methods were used for systematic coding and identification of emergent themes. All five participants were Caucasian, over 35 years of age, and all but one were married and had previous children. All except one chose to create an open adoption plan. The interviews focused on three major areas: (1) families’ reasons for creating an adoption plan, (2) informational and emotional needs of these families, and (3) messages to other birthparents and medical health professionals about special needs adoption. Participants reported various reasons for creating an adoption plan including unplanned pregnancy, being unprepared to raise a child with Down syndrome, and religious and personal beliefs about termination of pregnancy. Having support of their partners, gaining acceptance from others about their decision and talking to other birthparents who have gone through the same situation were the most significant emotional needs of these birthmothers. Finding information about adoption resources was one of the informational needs experienced. Not all birthmothers interviewed received information about the option of adoption from their genetic counselors or medical healthcare providers. The birthmothers also expressed concerns that people are unaware of the option of creating an adoption plan, and suggested that more exposure is necessary. Sharing results from this study with genetic counselors and other healthcare professionals may help future birthmothers approach this option with greater ease, and hopefully help healthcare professionals provide support and referrals to clients involved in special needs adoptions

    Lucem Ferre, a Master of Fine Arts thesis exhibition was influenced by an interpretation of bioluminescence, a chemical process in which a living organism catalyzes and releases light from within; this process is also effected by microorganisms living symbiotically within host animal and plant bodies. It is primarily observed in deep-sea marine as well as in terrestrial life. Bioluminescent life forms embody dualities; they are luminous and subdued, beautiful and repulsive, still and active. I use non-traditional materials, silicone, phosphorescent pigments and ultraviolet light, to create a jewelry collection of “wearable creatures” inhabiting an enigmatic and unworldly gallery environment. This 15-piece collection includes brooches, rings, armbands and a neckpiece; they are ambiguous, mysterious, luminous and whimsical jewelry pieces synthesizing visual and tactile elements to stimulate their simultaneous experience for viewer and wearer. As “wearable creatures” they exemplify the notion of portraying a blown-up world of microscopically investigated living organisms, simultaneously suggesting their internal growth and decay and their interactions with the body. Similar to bioluminescent life forms, these “wearable creatures” inhabit a life of dualities as they bear and bare their light
