    Comparative Interactomes of VRK1 and VRK3 with Their Distinct Roles in the Cell Cycle of Liver Cancer

    Vaccinia-related kinase 1 (VRK1) and VRK3 are members of the VRK family of serine/threonine kinases and are principally localized in the nucleus. Despite the crucial roles of VRK1/VRK3 in physiology and disease, the molecular and functional interactions of VRK1/VRK3 are poorly understood. Here, we identified over 200 unreported VRK1/VRK3-interacting candidate proteins by affinity purification and LC-MS/MS. The networks of VRK1 and VRK3 interactomes were found to be associated with important biological processes such as the cell cycle, DNA repair, chromatin assembly, and RNA processing. Interactions of interacting proteins with VRK1/VRK3 were confirmed by biochemical assays. We also found that phosphorylations of XRCC5 were regulated by both VRK1/VRK3, and that of CCNB1 was regulated by VRK3. In liver cancer cells and tissues, VRK1/VRK3 were highly upregulated and its depletion affected cell cycle progression in the different phases. VRK3 seemed to affect S phase progression and G2 or M phase entry and exit, whereas VRK1 affects G1/S transition in the liver cancer, which could be explained by different interacting candidate proteins. Thus, this study not only provides a resource for investigating the unidentified functions of VRK1/VRK3, but also an insight into the regulatory roles of VRK1/VRK3 in biological processes.11Ysciescopuskc

    Chlorophyll and Total Suspended Materials Concentrations and Remote Sensing Reflectance Data measured at the Red Tide Area of Jinhae, Geoje, and East Sea during August from 1998 to 2003 and August 2013

    The chlorophyll and total suspended materials concentrations and remote sensing reflectance data were observed for red tides occurring every summer in waters around the Korean Peninsula. In observation area and date, the field survey were performed (1) in the Jinhae and Geoje coasts during August 1998, August 1999, August 2001, and August 2003, (2) in East Sea coast during August 2013. The remote sensing reflectance data were obtained from portable spectroradiometer. The chlorophyll concentration data were obtained from spectrophotometric method and the total suspended materials concentration data were obtained from filter-weight difference method. The remote sensing reflectance data were validated using Moon et al.(2012). The chlorophyll concentration data were validated using baseline correction and subtraction of 750 nm value, and the total suspended materials concentration data were validated using variation of humidity

    Little Response of Cerebral Metastasis from Hepatocellular Carcinoma to Any Treatments

    Objective : We retrospectively evaluated the survival outcome of patients with brain metastasis from hepatocellular carcinoma (HCC). Methods : Between 1991 and 2007, a total of 20 patients were diagnosed as having brain metastasis from HCC. The mean age of the patients was 55 +/- 13 years, and 17(85.0%) were men. Seventeen (85.0%) patients had already extracranial metastases. The median time from diagnosis of HCC to brain metastasis was 18.5 months. Fourteen (70.0%) patients had stroke-like presentation due to intracerebral hemorrhage (ICH). Ten (50.0%) patients had single or solitary brain metastasis. Among a total of 34 brain lesions, 31 (91.2%) lesions had the hemorrhagic components. Results : The median survival time was 8 weeks (95% Cl, 5.08-10.92), and the actuarial survival rates were 85.0%, 45.0%, 22.5%, and 8.4% at 4, 12, 24, and 54 weeks. Age < 60 years, treatment of the primary and/or extracranial lesions, and recurrent ICH were the possible prognostic factors (p = 0.044, p < 0.001, and p = 0.111, respectively). The median progression-free survival (PFS) time was 3 months (95% Cl, 0.95-5.05). Conclusion : The overall survival of the patients with brain metastasis from HCC was very poor with median survival time being only 8 weeks.     Comparison of the Effectiveness of Interventional Endoscopy in Bleeding Peptic Ulcer Disease according to the Timing of Endoscopy

    Background/Aims: The optimal timing for interventional endoscopy in bleeding peptic ulcer disease is controversial. This study compared the outcomes between early endoscopy and delayed endoscopy in patients with bleeding peptic ulcer disease. Methods: We conducted a prospective analysis of data from 90 patients with bleeding peptic ulcer disease who visited the emergency room between May 2006 and September 2007. Patients were categorized into two groups: the early-endoscopy group (admitted during the daytime or at night with prompt endoscopic management) and the delayed-endoscopy group (admitted at night or during weekends, with endoscopic management delayed until the next day). We compared the clinical outcomes of endoscopy between the two groups. Results: There were 49 patients in the early-endoscopy group and 41 patients in the delayed-endoscopy group. Patient demographics, clinical characteristics, bleeding control modality, and Rockall score did not differ between the two groups. There were also no significant differences between the early- and delayed-encloscopy groups 'in the re-bleeding rate (3/49 vs 5/41, p=0.313), the duration of hospital stay (10.7 vs 9.3 days, p=0.437), and the total amount of blood transfused (3.4 vs 2.7 units, p=0.240). Conclusions: The effectiveness of interventional endoscopy for patients with bleeding peptic ulcer disease is not significantly affected by the timing of endoscopy.     Inflammatory Processes in Kawasaki Disease Reach their Peak at the Sixth Day of Fever Onset: Laboratory Profiles According to Duration of Fever

    We evaluated the inflammatory indices according to the fever duration in children with Kawasaki disease (KD), and determined duration when the inflammatory processes in KD reach their peak. Children with KD (n=152) were classified into 7 groups according to fever duration: at the third day or earlier (n=20), fourth (n=33), fifth (n=46), sixth (n=15), seventh (n=15), eighth (n=9), and at the ninth day or later after fever onset (n= 14). The levels of various laboratory indices were determined 3 times: before, 24 hr and 7 days after intravenous immunoglobulin administration (2 g/kg). WBC and neutrophil counts, and C-reactive protein level were the highest at the sixth day. Levels of hemoglobin, albumin, and high density lipoprotein cholestrol were the lowest at the sixth day. Although these indices were not significant statistically between groups, the indices showed either bell-shaped or U-shaped distribution of which peak or trench were at the sixth day. These findiugs showed that the inflammatory processes in KD reach peak on the sixth day of fever onset. This finding is important because a higher single-dose intravenous immunoglobulin treatment before the peak day may help reduce the coronary artery lesions in KD

    Effects of stimulating interleukin -2/anti- interleukin -2 antibody complexes on renal cell carcinoma

    Abstract Background Current therapies for advanced renal cell carcinoma (RCC) have low cure rates or significant side effects. It has been reported that complexes composed of interleukin (IL)-2 and stimulating anti-IL-2 antibody (IL-2C) suppress malignant melanoma growth. We investigated whether it could have similar effects on RCC. Methods A syngeneic RCC model was established by subcutaneously injecting RENCA cells into BALB/c mice, which were administered IL-2C or phosphate-buffered saline every other day for 4 weeks. RCC size was measured serially, and its weight was assessed 4 weeks after RENCA injection. Immune cell infiltration into RCC lesions and spleen was assessed by flow cytometry and immunohistochemistry. Results IL-2C treatment increased the numbers of CD8+ memory T and natural killer (NK) cells in healthy BALB/c mice (P < 0.01). In the spleen of RCC mice, IL-2C treatment also increased the number of CD8+ memory T, NK cells, and macrophages as compared to PBS-treated controls (P < 0.01). The number of interferon-γ- and IL-10-producing splenocytes increased and decreased, respectively after 4 weeks in the IL-2C-treated mice (P < 0.01). Tumor-infiltrating immune cells including CD4+ T, CD8+ T, NK cells as well as macrophages were increased in IL-2C-treated mice than controls (P < 0.05). Pulmonary edema, the most serious side effect of IL-2 therapy, was not exacerbated by IL-2C treatment. However, IL-2C had insignificant inhibitory effect on RCC growth (P = 0.1756). Conclusions IL-2C enhanced immune response without significant side effects; however, this activity was not sufficient to inhibit RCC growth in a syngeneic, murine model

    Robust algorithm for estimating total suspended solids (TSS) in inland and nearshore coastal waters

    One of the challenging tasks in modern aquatic remote sensing is the retrieval of near-surface concentrations of Total Suspended Solids (TSS). This study aims to present a Statistical, inherent Optical property (IOP) -based, and muLti-conditional Inversion proceDure (SOLID) for enhanced retrievals of satellite-derived TSS under a wide range of in-water bio-optical conditions in rivers, lakes, estuaries, and coastal waters. In this study, using a large in situ database (N \u3e 3500), the SOLID model is devised using a three-step procedure: (a) water-type classification of the input remote sensing reflectance (Rrs), (b) retrieval of particulate backscattering (bbp) in the red or near-infrared (NIR) regions using semi-analytical, machine-learning, and empirical models, and (c) estimation of TSS from bbp via water-type-specific empirical models. Using an independent subset of our in situ data (N = 2729) with TSS ranging from 0.1 to 2626.8 [g/m3], the SOLID model is thoroughly examined and compared against several state-of-the-art algorithms (Miller and McKee, 2004; Nechad et al., 2010; Novoa et al., 2017; Ondrusek et al., 2012; Petus et al., 2010). We show that SOLID outperforms all the other models to varying degrees, i.e.,from 10 to \u3e100%, depending on the statistical attributes (e.g., global versus water-type-specific metrics). For demonstration purposes, the model is implemented for images acquired by the MultiSpectral Imager aboard Sentinel-2A/B over the Chesapeake Bay, San-Francisco-Bay-Delta Estuary, Lake Okeechobee, and Lake Taihu. To enable generating consistent, multimission TSS products, its performance is further extended to, and evaluated for, other missions, such as the Ocean and Land Color Instrument (OLCI), Moderate Resolution Imaging Spectroradiometer (MODIS), Visible Infrared Imaging Radiometer Suite (VIIRS), and Operational Land Imager (OLI). Sensitivity analyses on uncertainties induced by the atmospheric correction indicate that 10% uncertainty in Rrs leads to \u3c20% uncertainty in TSS retrievals from SOLID. While this study suggests that SOLID has a potential for producing TSS products in global coastal and inland waters, our statistical analysis certainly verifies that there is still a need for improving retrievals across a wide spectrum of particle loads
