    Supporting Teachers in Times of Change: The Job Demands- Resources Model and Teacher Burnout During the COVID-19 Pandemic

    Burnout in teachers has been broadly investigated, but no studies have investigated burnout in teachers during a pandemic. The current study is based on a survey of 1278 Canadian teachers and examined whether the Job Demands-Resources model was a useful lens for examining teacher burnout in this unprecedented context. Results supported the model in general terms in that most demands were most strongly correlated with the initial exhaustion stage of burnout. However, not all resources were most strongly associated with the later stages of burnout, suggesting that the examination of specific resources in the context of a pandemic as opposed to examining resources together as a latent variable contributes to development of a more refined model. Suggestions for supporting teachers’ welfare are provided

    Does life satisfaction reduce risk of incident hypertension and stroke? Evidence from the Whitehall II cohort

    Background: Previous studies showed life satisfaction is related to reduced risk of coronary heart disease and diabetes, but its association with other cardiometabolic endpoints including hypertension and stroke remains unexplored. This study examined life satisfaction's prospective association with incident hypertension and stroke in middle-aged adults. Methods: At baseline (1985–1988), 6225 healthy British civil servants aged 35–55 from the Whitehall II cohort completed the validated Satisfaction with Life Scale and provided information regarding sociodemographics, a range of health-related factors, and psychological distress. Incident hypertension was ascertained according to clinic-derived measures of systolic or diastolic blood pressure of ≥140/90 mmHg, respectively, or self-reports of either physician-diagnosed hypertension or hypertensive medication use. Incident stroke and transient ischemic attack (TIA) were ascertained by self-reported physician diagnosis. Follow-up assessments occurred every 2–5 years through 2017. Cox proportional hazards regression models estimated hazard ratios (HR) and 95% confidence intervals (CI) of hypertension and stroke/TIA risk separately. Results: Over a 31-year follow-up, 2703 cases of hypertension and 370 cases of stroke/TIA occurred. Life satisfaction was not related to risk of developing hypertension but was associated with 12% decreased risk of stroke/TIA after controlling for sociodemographics, health status, and health behaviors (HRper 1-SD = 0.88; 95%CI = 0.79–0.98). However, the association was attenuated after adjustment for psychological distress. Conclusions: No robust associations were found between life satisfaction and incident hypertension and stroke/TIA, respectively, after accounting for well-established risk factors and psychological distress. More research is needed to understand why associations of life satisfaction with cardiometabolic health seem to vary across endpoints

    Pkd1 transgenic mice: Adult model of polycystic kidney disease with extrarenal and renal phenotypes

    While high levels of Pkd1 expression are detected in tissues of patients with autosomal dominant polycystic kidney disease (ADPKD), it is unclear whether enhanced expression could be a pathogenetic mechanism for this systemic disorder. Three transgenic mouse lines were generated from a Pkd1-BAC modified by introducing a silent tag via homologous recombination to target a sustained wild type genomic Pkd1 expression within the native tissue and temporal regulation. These mice specifically overexpressed the Pkd1 transgene in extrarenal and renal tissues from approximately 2- to 15-fold over Pkd1 endogenous levels in a copy-dependent manner. All transgenic mice reproducibly developed tubular and glomerular cysts leading to renal insufficiency. Interestingly, Pkd1(TAG) mice also exhibited renal fibrosis and calcium deposits in papilla reminiscent of nephrolithiasis as frequently observed in ADPKD. Similar to human ADPKD, these mice consistently displayed hepatic fibrosis and approximately 15% intrahepatic cysts of the bile ducts affecting females preferentially. Moreover, a significant proportion of mice developed cardiac anomalies with severe left ventricular hypertrophy, marked aortic arch distention and/or valvular stenosis and calcification that had profound functional impact. Of significance, Pkd1(TAG) mice displayed occasional cerebral lesions with evidence of ruptured and unruptured cerebral aneurysms. This Pkd1(TAG) mouse model demonstrates that overexpression of wildtype Pkd1 can trigger the typical adult renal and extrarenal phenotypes resembling human ADPKD.This work was supported by the Canadian Institutes of Health Research (CIHR) [MOP-81325 to MT] and a CIHR Frederick Banting and Charles Best studentship to AK and a Fonds de la Recherche en Santé du Québec (FRSQ) studentship to MC

    Brillouin light scattering study of Co2_{2}Cr0.6_{0.6}Fe0.4_{0.4}Al and Co2_{2}FeAl Heusler compounds

    The thermal magnonic spectra of Co2_{2}Cr0.6_{0.6}Fe0.4_{0.4}Al (CCFA) and Co2_2FeAl were investigated using Brillouin light scattering spectroscopy (BLS). For CCFA, the exchange constant A (exchange stiffness D) is found to be 0.48 μ\muerg/cm (203 meV A2^2), while for Co2_2FeAl the corresponding values of 1.55 μ\muerg/cm (370 meV A2^2) were found. The observed asymmetry in the BLS spectra between the Stokes and anti-Stokes frequencies was assigned to an interplay between the asymmetrical profiles of hybridized Damon-Esbach and perpendicular standing spin-wave modes, combined with the optical sensitivity of the BLS signal to the upper side of the CCFA or Co2_2FeAl film

    Purification of the food-borne carcinogens 2-amino-3-methylimidazo [4,5-f]quinoline and 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline in heated meat products by immunoaffinity chromatography

    A rapid and simple scheme has been developed for the isolation and purification of two of the major mutagenic heterocyclic amines formed in heated beef products by affinity chromatography using monoclonal antibodies which recognize 2-amino-3-methylimidazo(4,5-f]quinoline (IQ). Two cell lines producing IgG antibodies were established following fusion of Sp2 or P3x.63 myeloma cells with spleen cells of immunized BALB/cby mice. The antigen was bovine gamma globulin haptenized with 2-(3-carboxypropylthio)-3-methylimidazo[4,5-f]quinoline. The antibodies were immobilized on CNBr-activated Sepharose 4B. IQ and MeIQx formed in heated beef products were partially purified by XAD-2 chromatography and then applied to the affinity columns. Purification by affinity chromatography was adequate for subsequent quantitative analysis by HPLC with UV detection. With this purification scheme as little as 1 g of beef extract or 15 g of fried beef could be assayed for IQ and MeIQx at the part per billion level. Both antibodies had similar affinity constants for IQ (9.3 × 106 and 6.7 × 106 M−1) and for MeIQx (7.1 × 105 and 2.7 × 105 M−1) and both were suitable for immunoaffinity purification of IQ from complex mixtures. MAb2 could be used as well to selectively remove MeIQx from meat products after partial purification by XAD-2. MAb1, despite having a 3-fold higher affinity than MAb2 for MeIQx, could not be used for affinity chromatography for this mutage

    AsaGEI2b: a new variant of a genomic island identified in the Aeromonas salmonicida subsp. salmonicida JF3224 strain isolated from a wild fish in Switzerland

    Aeromonas salmonicida subsp. salmonicida is the causal agent of furunculosis in salmonids. We recently identified a group of genomic islands (AsaGEI) in this bacterium. AsaGEI2a, one of these genomic islands, has almost exclusively been identified in isolates from North America. To date, Aeromonas salmonicida subsp. salmonicida JF3224, a strain isolated from a wild brown trout (Salmo trutta) caught in Switzerland, was the only European isolate that appeared to bear AsaGEI2a. We analyzed the genome of JF3224 and showed that the genomic island in JF3224 is a new variant of AsaGEI, which we have called AsaGEI2b. While AsaGEI2b shares the same integrase gene and insertion site as AsaGEI2a, it is very different in terms of many other features. Additional genomic investigations combined with PCR genotyping revealed that JF3224 is sensitive to growth at 25°C, leading to insertion sequence-dependent rearrangement of the locus on the pAsa5 plasmid that encodes a type three secretion system, which is essential for the virulence of the bacterium. The analysis of the JF3224 genome confirmed that AsaGEIs are accurate indicators of the geographic origins of A. salmonicida subsp. salmonicida isolates and is another example of the susceptibility of the pAsa5 plasmid to DNA rearrangement

    Sulfur isotopes in otoliths allow discrimination of anadromous and non-anadromous ecotypes of sockeye salmon (Oncorhynchus nerka)

    Oncorhynchus nerka occur both as anadromous sockeye salmon that spend most of their life in the ocean, and as non-anadromous kokanee salmon that remain in fresh water their entire lives. We assessed whether stable isotopes of sulfur (δ34S) in otoliths could be used to distinguish sockeye salmon and kokanee ecotypes that are otherwise difficult to identify when they share a common freshwater rearing environment. We also investigated the chemical link between salmon and their diet by measuring δ34S in various fish tissues (eggs, muscle, scales) and zooplankton. δ34S (mean±SE) in sockeye salmon eggs (18.7 ± 0.4‰) and marine zooplankton (20.5 ± 0.1‰) were enriched by 10–14‰ compared with kokanee eggs and freshwater zooplankton. δ34S in the otolith cores of sockeye salmon (19.2 ± 0.7‰) and kokanee salmon (5.3 ± 1.1‰) were similar to δ34S in marine and freshwater zooplankton, respectively, indicating that the core is derived from maternal yolk tissue and reflects the maternal diet. δ34S in the freshwater growth zone of otoliths did not differ significantly between sockeye (5.9 ± 1.1‰) and kokanee salmon (4.4 ± 1.2‰), and was similar to freshwater zooplankton. The mean difference between δ34S in the otolith core and first year of growth was 13.3 ± 1.4‰ for sockeye and 0.65 ± 1.3‰ for kokanee salmon. A quadratic discriminant function developed from measurements of δ34S in otoliths of known maternal origin provided perfect classification rates in cross-validation tests. Thus, sulfur isotope ratios in otoliths are effective in discriminating between anadromous and non-anadromous ecotypes of O. nerka
