    A high proliferation rate measured by cyclin A predicts a favourable chemotherapy response in soft tissue sarcoma patients

    A small but not insignificant number of patients experience a prolonged survival after treatment of metastatic soft tissue sarcoma. This must be weighed against the majority of the patients who benefit little from the therapy, but nevertheless experience its side-effects. It would therefore be of utmost importance to be able to screen for those patients who respond to the treatment. Since proliferating cells are more sensitive to chemotherapy than non-proliferative cells, we measured the proliferation rate of the primary tumour of 55 soft tissue sarcoma patients with locally advanced or metastatic disease by determining the flow cytometric S phase fraction and immunohistochemical Ki-67 and cyclin A scores. S phase fraction or Ki-67 score did not predict chemotherapy response or progression-free survival. A high cyclin A score, however, correlated with a better chemotherapy response (P = 0.02) and longer progression-free survival time (P = 0.04). Our results suggest that a high cyclin A score predicts chemotherapy sensitivity. © 1999 Cancer Research Campaig

    Gender and line size factors modulate the deviations of the subjective visual vertical induced by head tilt

    <p>Abstract</p> <p>Background</p> <p>The subjective visual vertical (SVV, the visual estimation of gravitational direction) is commonly considered as an indicator of the sense of orientation. The present study examined the impact of two methodological factors (the angle size of the stimulus and the participant's gender) on deviations of the SVV caused by head tilt. Forty healthy participants (20 men and 20 women) were asked to make visual vertical adjustments of a light bar with their head held vertically or roll-tilted by 30° to the left or to the right. Line angle sizes of 0.95° and 18.92° were presented.</p> <p>Results</p> <p>The SVV tended to move in the direction of head tilt in women but away from the direction of head tilt in men. Moreover, the head-tilt effect was also modulated by the stimulus' angle size. The large angle size led to deviations in the direction of head-tilt, whereas the small angle size had the opposite effect.</p> <p>Conclusions</p> <p>Our results showed that gender and line angle size have an impact on the evaluation of the SVV. These findings must be taken into account in the growing body of research that uses the SVV paradigm in disease settings. Moreover, this methodological issue may explain (at least in part) the discrepancies found in the literature on the head-tilt effect.</p

    Comparison of the Ki-67 score and S-phase fraction as prognostic variables in soft-tissue sarcoma

    Immunohistochemically determined Ki-67 scores and flow cytometrically determined S-phase fractions were successfully evaluated from the primary tumours of 123 patients with soft-tissue sarcoma. All patients had either limb or superficial trunk tumours. Ki-67 score correlated strongly with ploidy, S-phase fraction and grade. Ki-67 did not correlate with the size of the primary tumour. When analysed as a continuous variable, Ki-67 was a stronger predictor of both metastasis-free survival and disease-specific overall survival (P= 0.003 and 0.04 respectively) than was the S-phase fraction (P= 0.06 and 0.07 respectively). We tested the relevance of different cut-point values by dividing the whole material into two parts at every 10% (e.g. 10% of patients vs. the remaining 90%, 20% vs. 80%, etc.). We counted the relative risk and confidence interval at all these cut-off points. Ki-67 had good prognostic discriminating power irrespective of the cut-point value, but S-phase fraction lost its prognostic power at higher cut-point values. In conclusion, we found that Ki-67 is a useful prognostic tool in the treatment of soft-tissue sarcoma patients irrespective of the cut-point value. S-phase fraction can be used at lower cut-point values. © 1999 Cancer Research Campaig

    Die Aktivitätsperiodik der weissen Maus im Kunsttag von 16–29 Stunden Länge

    Die Tagesperiodik der weißen Maus folgt dem langsam bis zu 21 Std verkürzten oder bis zu 27 Std verlängerten Kunsttag. Dabei verschiebt sich ihr Hauptmaximum im verkürzten Tag in die Dunkelzeit, im verlängerten Tag in die Lichtzeit. Die Verlagerung wird als Resultante zweier Kräfte angesehen: die endogene Komponente der Tierperiodik sucht ihre Eigenperiodik beizubehalten, der Zeitgeber zwingt dem Tier eine davon abweichende Frequenz auf. Dem auf mehr als 20 Std verkürzten oder auf 28 Std verlängerten Kunsttag vermag sich die Maus nicht anzupassen. Ebenso wie im plötzlich verkürzten oder verlängerten Tag stellt sie eine von der Zeitgeberfrequenz unabhängige Eigenperiodik ein. Die Eigenperiodik verhält sich wie im Dauerlicht oder Dauerdunkel: Fallen die Hauptaktivitätsschübe in die Lichtzeit, verlängert sich die Periodendauer auf etwa 26 Std, liegen sie in der Dunkelzeit, verkürzt sie sich auf etwa 23 Std. Dem 20- und 28-Std-Tag ist das Tier teilangepaßt. Die vorwiegend exogen durch den Lichtreiz gebildeten Morgenmaxima folgen noch dem Zeitgeber, die endogenen Hauptmaxima der Eigenperiodik des Tieres: es überlagern sich 2 verschiedene Frequenzen der Tierperiodik. Die Ergebnisse aller Untersuchungen weisen erneut auf die wesentliche Rolle einer endogenen Anlage der Tierperiodik hin
