    Profession loss crisis at an old age: Specific features, factors, and mechanisms of coping

    This article discusses the specific characteristics of profession loss crisis at an old age. Profession loss crisis is the last normative crisis of personal professional development that is caused by the completion of one's professional biography after reaching a certain age. The research employs a psychobiographic method and a critical events method. These methods are based on the use of a formalized biographical questionnaire worked out by Norakidze V.G. and reconstructed by Zeer E.F. The authors have identified and provided a detailed description of the main factors that cause profession loss crisis: Random events, adverse circumstances while implementing professional plans, etc. The article outlines the main strategies for coping with this crisis: Changing jobs, re-training, the assistance of colleagues and administration, etc. The authors suggest technologies to minimize the effects of these factors and overcome profession loss crisis effectively. © 2019 by the authors

    Alanine 501 Mutations in Penicillin-Binding Protein 2 from Neisseria gonorrhoeae : Structure, Mechanism, and Effects on Cephalosporin Resistance and Biological Fitness

    Resistance of Neisseria gonorrhoeae to expanded-spectrum cephalosporins such as ceftriaxone and cefixime has increased markedly in the past decade. The primary cephalosporin resistance determinant is a mutated penA gene, which encodes the essential peptidoglycan transpeptidase, penicillin-binding protein 2 (PBP2). Decreased susceptibility and resistance can be conferred by mosaic penA alleles containing upward of 60 amino acid changes relative to wild-type PBP2, or by nonmosaic alleles with relatively few mutations, the most important of which occurs at Ala501 located near the active site of PBP2. Recently, fully cefixime- and ceftriaxone-resistant clinical isolates that harbored a mosaic penA allele with an A501P mutation were identified. To examine the potential of mutations at Ala501 to increase resistance to expanded-spectrum cephalosporins, we randomized codon 501 in a mosaic penA allele and transformed N. gonorrhoeae to increased cefixime resistance. Interestingly, only five substitutions of Ala501 (A501V, A501T, A501P, A501R, and A501S) that increased resistance and preserved essential transpeptidase function were isolated. To understand their structural implications, these mutations were introduced into the nonmosaic PBP2-6140CT, which contains four C-terminal mutations present in PBP2 from the penicillin-resistant strain FA6140. The crystal structure of PBP2-6140CT-A501T was determined and revealed ordering of a loop near the active site and a new hydrogen bond involving Thr501 that connects the loop and the SxxK conserved active site motif. The structure suggests that increased rigidity in the active site region is a mechanism for cephalosporin resistance mediated by Ala501 mutations in PBP2

    Entropic Fluctuations in Statistical Mechanics I. Classical Dynamical Systems

    Within the abstract framework of dynamical system theory we describe a general approach to the Transient (or Evans-Searles) and Steady State (or Gallavotti-Cohen) Fluctuation Theorems of non-equilibrium statistical mechanics. Our main objective is to display the minimal, model independent mathematical structure at work behind fluctuation theorems. Besides its conceptual simplicity, another advantage of our approach is its natural extension to quantum statistical mechanics which will be presented in a companion paper. We shall discuss several examples including thermostated systems, open Hamiltonian systems, chaotic homeomorphisms of compact metric spaces and Anosov diffeomorphisms.Comment: 72 pages, revised version 12/10/2010, to be published in Nonlinearit

Исследование проводилось в Екатеринбурге (Россия) с участием 291 учителя средней школы. Учителя писали психобиографические эссе, которые обрабатывались методом контент-анализа. В результате были определены два типа копинг-стратегий: реактивный и проактивный. Реактивные стратегии предполагают проявление инициативы, а не просто приспособление к ситуации; в проактивных стратегиях преобладают механизмы защиты и адаптации. В ходе исследования выявлено, что в процессе повышения квалификации учителя переключаются с реактивных стратегий на проактивные. Реактивные стратегии преобладают в кризисах профессиональной и образовательной ориентации и выбора профессии, а проактивные — в кризисах выбора карьеры, профессионального роста, профессиональных ожиданий.This article discusses professional crises and strategies of coping with them. The study was conducted in Yekaterinburg (Russia) and involved 291 secondary school teachers. The teachers wrote psychobiographical essays, which were processed through content analysis methods. As a result, two types of coping strategies were identified: reactive and proactive. Reactive strategies imply taking the initiative rather than merely adjusting to the situation while in proactive strategies defence and adaptation mechanisms prevail. We found that in their professional development teachers switch from reactive to proactive strategies. Reactive strategies predominate in professional and educational orientation and profession choice crises while proactive ones, in crises of career choice, professional growth, professional expectations and non-normative crises

    Rapid ecological change in the coastal zone of Lake Baikal (East Siberia): Is the site of the world\u27s greatest freshwater biodiversity in danger?

    Ecological degradation of the benthic littoral zone is an emerging, urgent problem at Lake Baikal (East Siberia), the most species-rich lake on Earth. Within the last five years, multiple changes have occurred in the nearshore benthos where most of the lake\u27s endemic species reside. These changes include proliferation of benthic algae, deaths of snails and endemic sponges, large coastal wash-ups of dead benthic algae and macrophytes, blooms of toxin-producing benthic cyanobacteria, and inputs of industrial contaminants into parts of the lake. Some changes, such as massive coastal accumulations of benthic algae, are currently shared with the Laurentian Great Lakes (LGLs); however, the drivers of these changes differ between Lake Baikal and the LGLs. Coastal eutrophication from inputs of untreated sewage is causing problems at multiple sites in Lake Baikal, whereas in the LGLs, invasive dreissenid mussels redirect pelagic nutrients to the littoral substrate. At other locations in Lake Baikal, ecological degradation may have different causes including water level fluctuations and the input of toxic industrial contaminants. Importantly, the recent deterioration of the benthic littoral zone in both Lake Baikal and the LGLs has occurred while little change has occurred offshore. This highlights the necessity of monitoring both the littoral and pelagic zones of large lakes for assessing ecosystem health, change and conservation

    Bi-allelic variants in TSPOAP1, encoding the active zone protein RIMBP1, cause autosomal recessive dystonia

    Dystonia is a debilitating hyperkinetic movement disorder, which can be transmitted as a monogenic trait. Here, we describe homozygous frameshift, nonsense and missense variants in TSPOAP1, encoding the active zone RIM-binding protein 1 (RIMBP1), as a novel genetic cause of autosomal recessive dystonia in seven subjects from three unrelated families. Subjects carrying loss-of-function variants presented with juvenile-onset progressive generalized dystonia, associated with intellectual disability and cerebellar atrophy. Conversely, subjects carrying a pathogenic missense variant (p.Gly1808Ser) presented with isolated adult-onset focal dystonia. In mice, complete loss of RIMBP1, known to reduce neurotransmission, led to motor abnormalities reminiscent of dystonia, decreased Purkinje cell dendritic arborization, and reduced numbers of cerebellar synapses. In vitro analysis of the p.Gly1808Ser variant showed larger spike-evoked calcium transients and enhanced neurotransmission, suggesting that RIMBP1-linked dystonia can be caused by either reduced or enhanced rates of spike-evoked release in relevant neural networks. Our findings establish a direct link between dysfunction of the presynaptic active zone and dystonia and highlight the critical role played by well-balanced neurotransmission in motor control and disease pathogenesis

    HYPEST study: profile of hypertensive patients in Estonia

    <p>Abstract</p> <p>Background</p> <p>More than one third of adult population in Estonia has problems with elevated blood pressure (BP). The <it>Hypertension in Estonia </it>(HYPEST) study represents the country's first hypertension-targeted sample collection aiming to examine the epidemiological and genetic determinants for hypertension (HTN) and related cardiovascular diseases (CVD) in Estonian population. The HYPEST subjects (n = 1,966) were recruited across Estonia between 2004-2007 including clinically diagnosed HTN cases and population-based controls. The present report is focused on the clinical and epidemiological profile of HYPEST cases, and gender-specific effects on the pathophysiology of hypertension.</p> <p>Methods</p> <p>Current analysis was performed on 1,007 clinically diagnosed HTN patients (617 women and 390 men) aged 18-85 years. The hypertensives were recruited to the study by BP specialists at the North Estonia Medical Center, Centre of Cardiology, Tallinn or at the Cardiology Clinic, Tartu University Hospital, Estonia. Longitudinal BP data was extracted retrospectively from clinical records. Current and retrospective data of patient's medical history, medication intake and lifestyle habits were derived from self-administrated questionnaire and each variable was examined separately for men and women. Eleven biochemical parameters were measured from fasting serum samples of 756 patients.</p> <p>Results</p> <p>The distribution of recruited men and women was 39% and 61% respectively. Majority of Estonian HTN patients (85%) were overweight (BMI ≥ 25 kg/m<sup>2</sup>) and a total of 79% of patients had additional complications with cardiovascular system. In men, the hypertension started almost 5 years earlier than in women (40.5 ± 14.5 vs 46.1 ± 12.7 years), which led to earlier age of first myocardial infarction (MI) and overall higher incidence rate of MI among male patients (men 21.2%, women 8.9%, <it>P </it>< 0.0001). Heart arrhythmia, thyroid diseases, renal tubulo-intestinal diseases and hyperlipidemia were more prevalent in hypertensive women compared to men (<it>P </it>< 0.0001). An earlier age of HTN onset was significantly associated with smoking (<it>P </it>= 0.00007), obesity (BMI ≥ 30 kg/m<sup>2</sup>; <it>P </it>= 0.0003), increased stress (<it>P </it>= 0.0003) and alcohol consumption (<it>P </it>= 0.004).</p> <p>Conclusion</p> <p>Understanding the clinical profile of HTN patients contributes to CVD management. Estonian hypertension patients exhibited different disease and risk profiles of male and female patients. This well-characterized sample set provides a good resource for studying hypertension and other cardiovascular phenotypes.</p
