    Methamphetamine and Viagra Use: Relationship to Sexual Risk Behaviors

    Recent studies show that Viagra and methamphetamine use are associated with unprotected anal intercourse among men who have sex with men (MSM). In Long Beach, California, we have reported on an association between Viagra use and the use of amphetamines during sex. The current research investigated the use of both Viagra and amphetamine in men in Long Beach, California. Data on 1,839 men recruited into HIV prevention and testing programs were collected using the Risk Behavior Assessment. A generalized logit model was constructed comparing ever having used both amphetamine and Viagra together and separately, as compared to never having used either (referent). Men who used both methamphetamine and Viagra showed a significantly higher prevalence of hepatitis B, syphilis, and HIV compared to those who used only one or neither drug. Of the 1,794 complete cases, 11.1% (199/1794) had used both amphetamine and Viagra. Of 20 potential risk and protective factors for use of amphetamine and Viagra, 12 were significant predictors: ever used gamma-hydroxybutyrate (GHB), ever used cocaine, ever used ecstasy, being infected with HIV, race = White compared to other, ever having hepatitis B, ever using crack, ever given money to have sex, living in a hotel, ever been in drug treatment, and ever using heroin. The protective factor was being heterosexual. Viagra use was associated with insertive, and methamphetamine was associated with receptive, anal intercourse. GHB use appears to play a more important role than previously thought

    Risk factors for methamphetamine use in youth: a systematic review

    <p>Abstract</p> <p>Background</p> <p>Methamphetamine (MA) is a potent stimulant that is readily available. Its effects are similar to cocaine, but the drug has a profile associated with increased acute and chronic toxicities. The objective of this systematic review was to identify and synthesize literature on risk factors that are associated with MA use among youth.</p> <p>More than 40 electronic databases, websites, and key journals/meeting abstracts were searched. We included studies that compared children and adolescents (≤ 18 years) who used MA to those who did not. One reviewer extracted the data and a second checked for completeness and accuracy. For discrete risk factors, odds ratios (OR) were calculated and when appropriate, a pooled OR with 95% confidence intervals (95% CI) was calculated. For continuous risk factors, mean difference and 95% CI were calculated and when appropriate, a weighted mean difference (WMD) and 95% CI was calculated. Results were presented separately by comparison group: low-risk (no previous drug abuse) and high-risk children (reported previous drug abuse or were recruited from a juvenile detention center).</p> <p>Results</p> <p>Twelve studies were included. Among low-risk youth, factors associated with MA use were: history of heroin/opiate use (OR = 29.3; 95% CI: 9.8–87.8), family history of drug use (OR = 4.7; 95% CI: 2.8–7.9), risky sexual behavior (OR = 2.79; 95% CI: 2.25, 3.46) and some psychiatric disorders. History of alcohol use and smoking were also significantly associated with MA use. Among high-risk youth, factors associated with MA use were: family history of crime (OR = 2.0; 95% CI: 1.2–3.3), family history of drug use (OR = 4.7; 95% CI: 2.8–7.9), family history of alcohol abuse (OR = 3.2; 95% CI: 1.8–5.6), and psychiatric treatment (OR = 6.8; 95% CI: 3.6–12.9). Female sex was also significantly associated with MA use.</p> <p>Conclusion</p> <p>Among low-risk youth, a history of engaging in a variety of risky behaviors was significantly associated with MA use. A history of a psychiatric disorder was a risk factor for MA for both low- and high-risk youth. Family environment was also associated with MA use. Many of the included studies were cross-sectional making it difficult to assess causation. Future research should utilize prospective study designs so that temporal relationships between risk factors and MA use can be established.</p

    Cooling athletes with a spinal cord injury

    Cooling strategies that help prevent a reduction in exercise capacity whilst exercising in the heat have received considerable research interest over the past 3 decades, especially in the lead up to a relatively hot Olympic and Paralympic Games. Progressing into the next Olympic/Paralympic cycle, the host, Rio de Janeiro, could again present an environmental challenge for competing athletes. Despite the interest and vast array of research into cooling strategies for the able-bodied athlete, less is known regarding the application of these cooling strategies in the thermoregulatory impaired spinal cord injured (SCI) athletic population. Individuals with a spinal cord injury (SCI) have a reduced afferent input to the thermoregulatory centre and a loss of both sweating capacity and vasomotor control below the level of the spinal cord lesion. The magnitude of this thermoregulatory impairment is proportional to the level of the lesion. For instance, individuals with high-level lesions (tetraplegia) are at a greater risk of heat illness than individuals with lower-level lesions (paraplegia) at a given exercise intensity. Therefore, cooling strategies may be highly beneficial in this population group, even in moderate ambient conditions (~21 °C). This review was undertaken to examine the scientific literature that addresses the application of cooling strategies in individuals with an SCI. Each method is discussed in regards to the practical issues associated with the method and the potential underlying mechanism. For instance, site-specific cooling would be more suitable for an athlete with an SCI than whole body water immersion, due to the practical difficulties of administering this method in this population group. From the studies reviewed, wearing an ice vest during intermittent sprint exercise has been shown to decrease thermal strain and improve performance. These garments have also been shown to be effective during exercise in the able-bodied. Drawing on additional findings from the able-bodied literature, the combination of methods used prior to and during exercise and/or during rest periods/half-time may increase the effectiveness of a strategy. However, due to the paucity of research involving athletes with an SCI, it is difficult to establish an optimal cooling strategy. Future studies are needed to ensure that research outcomes can be translated into meaningful performance enhancements by investigating cooling strategies under the constraints of actual competition. Cooling strategies that meet the demands of intermittent wheelchair sports need to be identified, with particular attention to the logistics of the sport

However, how the BMP effector phospho-Mother against decapentaplegic (pMad) is processed following receptor activation remains poorly understood. Here we show that Drosophila Epsin1/Liquid facets (Lqf) positively regulates synaptic growth through post-endocytotic processing of pMad signaling complex. Lqf and the BMP receptor Wishful thinking (Wit) interact genetically and biochemically. lqf loss of function (LOF) reduces bouton number whereas overexpression of lqf stimulates bouton growth. Lqf-stimulated synaptic overgrowth is suppressed by genetic reduction of wit. Further, synaptic pMad fails to accumulate inside the motoneuron nuclei in lqf mutants and lqf suppresses synaptic overgrowth in spinster (spin) mutants with enhanced BMP signaling by reducing accumulation of nuclear pMad. Interestingly, lqf mutations reduce nuclear pMad levels without causing an apparent blockage of axonal transport itself. Finally, overexpression of Lqf significantly increases the number of multivesicular bodies (MVBs) in the synapse whereas lqf LOF reduces MVB formation, indicating that Lqf may function in signaling endosome recycling or maturation. Based on these observations, we propose that Lqf plays a novel endosomal role to ensure efficient retrograde transport of BMP signaling endosomes into motoneuron nuclei.Yeshttp://www.plosone.org/static/editorial#pee

    A hierarchical Bayesian quantitative microbiological risk assessment model for Salmonella in the sheep meat food chain

    Multiple foodborne routes of Salmonella infection have been observed; however, the majority of the literature to date has been dominated by research into the most frequently observed reservoirs, such as chicken, beef, and pork. While less commonly observed, outbreaks of Salmonella within sheep meat still occur, requiring extensive investigation by food safety inspectors. Risk assessment models inform policy makers and investigators of the risks posed by pathogens at each stage of the food chain, and help suggest at which stages in the food chain outbreaks are likely induced. This work is the first risk assessment into the prevalence of Salmonella throughout the sheep meat food chain, from farm to fork. A Bayesian evidence-synthesis model is used, informed by data gathered from 27 individual studies - an exhaustive search of the existing literature, to express and enumerate the current understanding of Salmonella prevalence in the sheep meat food chain in the form of probabilities of colonisation throughout the food chain. The resulting posterior estimate projects that 9 (0–29 95% HDI) UK individuals are likely to fall ill with salmonellosis due to sheep meat every year. A variance-based sensitivity analysis reveals that the abattoir module is the stage of greatest bacterial proliferation, highlighting it as the most probable source of outbreaks, though not to the exclusion of other factors

    Mathematical modelling approaches to understanding the spread of Campylobacter in broiler flocks

    Campylobacter spp. are one of the leading causes of human gastroenteritis globally, with the primary route of transmission being poultry meat. Flocks of broilers, chickens grown for their meat, will rapidly become colonised by Campylobacter on the farm, and this infection persists throughout the food chain. Recent attempts to prevent the spread of Campylobacter in broiler flocks have focused on biosecurity measures on farms to reduce the chance of initial infection. Such measures have proved ineffective, resulting in calls for more fundamental research into the ecology of Campylobacter in chicken flocks, and greater investigation of the bacteria-host relationship. This thesis presents a multitude of mathematical frameworks to explore these dynamics, driven by on-farm data from the last several decades. Firstly, I present a mechanistic model system of stochastic differential equations to demonstrate the population dynamics of multiple strains of Campylobacter at both in-host and flock level. I find that in-host dynamics are different from those observed in the flock at large, with single strains of Campylobacter seen to dominate the gut of a host-bird at any given time, compared to the more stable patterns of co-occurrence seen at the wider flock level. I show that Campylobacter outbreaks are most influenced by the demographic capabilities of other native gut microflora within host-birds, raising the hypothesis that further attention to bird gut health may be key to limiting the spread of Campylobacter. Building on this, I investigate data detailing the occurrence of multiple Campylobacter strains within a flock of chickens across a year. By using Bayesian transition models, competition matrix models, and patch-occupancy models, I demonstrate the existence of variation in the response to bacterial challenge amongst birds in the flock, and that this variation may drive the proliferation of Campylobacter. This is further supported by models I present showing that Campylobacter strains exist in a hierarchical system of competition, and that the diversity of strains found cannot be supported without the inclusion of host-bird factors. Lastly, I investigated the causal relationships between a variety of management and welfare factors for multiple commercial flocks by building and comparing logistic regression models with Bayesian network models. I show that measures of poor bird health and Campylobacter status are conditionally dependent upon one another, revealing that decisions in the rearing of these birds simultaneously jeopardise their health, and increase the risk of Campylobacter colonisation.</p
