    Knots and Classical 3-Geometries

    It has been conjectured by Rovelli that there is a correspondence between the space of link classes of a Riemannian 3-manifold and the space of 3-geometries (on the same manifold).     Bubble transport by electro-magnetophoretic forces at anode bottom of aluminium cells

    Electrically conducting and nonconducting particles and bubbles experience additional forcing in a liquid which carries electric current. These so called electro-magnetophoretic forces are well known in metallurgical applications, like metal purification in vacuum-arc remelting, electro-slag processes, impurity removal or concentration change in special castings. However, the effect of electro-magnetophoretic forces has never been considered for aluminium cells where the gas bubbles evolving in the liquid electrolyte are surrounded by an electric current and significant magnetic fields. We present models to estimate the effect of electric current flow in the vicinity of the bubbles and the additional pressure distribution resulting from the magnetic forces in the surrounding liquid electrolyte. According to the estimates, this force becomes important for bubbles exceeding 2 mm in size, and could be sufficient to overcome the typical drag force associated with electrolyte flow thereby opposing motion of the bubble along the base of the anode when it is inclined at a slight angle. The effect could explain certain features of the anode effect onset. Mathematical models and numerical results are presented and a further implementation in the general MHD code for the aluminium cell design is discussed

    Role of Interleukin 17 in arthritis chronicity through survival of synoviocytes via regulation of synoviolin expression

    Background: The use of TNF inhibitors has been a major progress in the treatment of chronic inflammation. However, not all patients respond. In addition, response will be often lost when treatment is stopped. These clinical aspects indicate that other cytokines might be involved and we focus here on the role of IL-17. In addition, the chronic nature of joint inflammation may contribute to reduced response and enhanced chronicity. Therefore we studied the capacity of IL-17 to regulate synoviolin, an E3 ubiquitin ligase implicated in synovial hyperplasia in human rheumatoid arthritis (RA) FLS and in chronic reactivated streptococcal cell wall (SCW)-induced arthritis.<p></p> Methodology/Principal Findings: Chronic reactivated SCW-induced arthritis was examined in IL-17R deficient and wild-type mice. Synoviolin expression was analysed by real-time RT-PCR, Western Blot or immunostaining in RA FLS and tissue, and p53 assessed by Western Blot. Apoptosis was detected by annexin V/propidium iodide staining, SS DNA apoptosis ELISA kit or TUNEL staining and proliferation by PCNA staining. IL-17 receptor A (IL-17RA), IL-17 receptor C (IL-17-RC) or synoviolin inhibition were achieved by small interfering RNA (siRNA) or neutralizing antibodies. IL-17 induced sustained synoviolin expression in RA FLS. Sodium nitroprusside (SNP)-induced RA FLS apoptosis was associated with reduced synoviolin expression and was rescued by IL-17 treatment with a corresponding increase in synoviolin expression. IL-17RC or IL-17RA RNA interference increased SNP-induced apoptosis, and decreased IL-17-induced synoviolin. IL-17 rescued RA FLS from apoptosis induced by synoviolin knockdown. IL-17 and TNF had additive effects on synoviolin expression and protection against apoptosis induced by synoviolin knowndown. In IL-17R deficient mice, a decrease in arthritis severity was characterized by increased synovial apoptosis, reduced proliferation and a marked reduction in synoviolin expression. A distinct absence of synoviolin expressing germinal centres in IL-17R deficient mice contrasted with synoviolin positive B cells and Th17 cells in synovial germinal centre-like structures.<p></p> Conclusion/Significance: IL-17 induction of synoviolin may contribute at least in part to RA chronicity by prolonging the survival of RA FLS and immune cells in germinal centre reactions. These results extend the role of IL-17 to synovial hyperplasia.<p></p&gt

    Minimally invasive glaucoma surgery: comparison of iStent with iStent inject in primary open angle glaucoma

    Importance:Minimally invasive glaucoma surgery has gained significant traction in recent years. This study evaluates the first‐ and second‐generation trabecular micro‐bypass stents “iStent” and “iStent inject”.Background:To evaluate and compare the effect of a single iStent and double iStent inject in primary open angle glaucoma.Design:Prospective comparative case series.Participants:Primary open angle glaucoma patients undergoing trabecular micro‐bypass stent insertion combined with cataract surgery.Methods:Baseline demographic information, preoperative, intraoperative and postoperative outcomes including intraocular pressure (IOP), visual acuity, reliance on glaucoma medication and complications were collected and analysed.Main Outcome Measures:Primary, secondary and tertiary outcome measures were consecutively defined as an IOP of ≤18 mmHg with zero medications, an IOP of ≤18 mmHg with reduced medications or a 20% reduction in IOP with or without medication.Results:The study comprised 145 eyes in the iStent and 100 eyes in the iStent inject group. At 12 months, 56.0% of the iStent and 51.3% of the iStent inject eyes had achieved primary success and 63.1% and 57.7% secondary success. The mean postoperative IOP was 16.6 mmHg in iStent and 16.9 mmHg in iStent inject. Survival analysis demonstrated a greater incidence of failure in the iStent inject beyond 5 months.Conclusions and Relevance:Both trabecular micro‐bypass stents in this study were effective in reducing IOP and the burden of medication when combined with cataract surgery. There was no statistically significant difference between the two groups across our outcome measures although the iStent inject required earlier recommencement of medications for optimal IOP control

    Myc inhibition impairs autophagosome formation

    Autophagy, a major clearance route for most long-lived proteins and organelles, has long been implicated in cancer development. Myc is a proto-oncogene often found to be deregulated in many cancers, and thus presents as an attractive target for design of cancer therapy. Therefore, understanding the relationship between anti-Myc strategies and autophagy will be important for development of effective therapy. Here we show that Myc depletion inhibits autophagosome formation and impairs clearance of autophagy substrates. Myc suppression has an inhibitory effect on autophagy via reduction of JNK1 and Bcl2 phosphorylation. Additionally, the decrease in JNK1 phosphorylation observed with Myc knockdown is associated with a reduction in ROS production. Our data suggest that targeting Myc in cancer therapy might have the additional benefit of inhibiting autophagy in the case of therapy resistance associated with chemotherapy-induced autophagy

    Computer simulations of VANETs using realistic city topologies

    Researchers in vehicular ad hoc networks (VANETs) commonly use simulation to test new algorithms and techniques. This is the case because of the high cost and labor involved in deploying and testing vehicles in real outdoor scenarios. However, when determining the factors that should be taken into account in these simulations, some factors such as realistic road topologies and presence of obstacles are rarely addressed. In this paper, we first evaluate the packet error rate (PER) through actual measurements in an outdoor road scenario, and deduce a close model of the PER for VANETs. Secondly, we introduce a topology-based visibility scheme such that road dimension and geometry can be accounted for, in addition to line-of-sight. We then combine these factors to determine when warning messages (i.e., messages that warn drivers of danger and hazards) are successfully received in a VANET.
