    Autocatalytic amplification of Alzheimer-associated Aβ42 peptide aggregation in human cerebrospinal fluid

    Alzheimer’s disease is linked to amyloid β (Aβ) peptide aggregation in the brain, and a detailed understanding of the molecular mechanism of Aβ aggregation may lead to improved diagnostics and therapeutics. While previous studies have been performed in pure buffer, we approach the mechanism in vivo using cerebrospinal fluid (CSF). We investigated the aggregation mechanism of Aβ42 in human CSF through kinetic experiments at several Aβ42 monomer concentrations (0.8–10 µM). The data were subjected to global kinetic analysis and found consistent with an aggregation mechanism involving secondary nucleation of monomers on the fibril surface. A mechanism only including primary nucleation was ruled out. We find that the aggregation process is composed of the same microscopic steps in CSF as in pure buffer, but the rate constant of secondary nucleation is decreased. Most importantly, the autocatalytic amplification of aggregate number through catalysis on the fibril surface is prevalent also in CSF

    Standardization of Marine Meteorological Data from FINO Offshore Platforms

    KlimawandelDer Ausbau der Offshore-Windenergie gehört zu den erklärten Zielen der Bundesregierung. Um bessere Ken ntnisse der Bedingungen auf See zu erlangen, wurden drei Forschungsplattformen in der Nordsee (FINO 1 und 3) und Ostsee (FINO 2) errichtet. An diesen werden meteorologische und ozeanographische Größen in unterschiedlichen Höhen gemessen, um unter anderem Aussagen zu Vertikalprofilen der Windgeschwindigkeit treffen zu können. Da sich die Plattformen und Masten im Design unterscheiden und sich insbesondere bei den Windmessungen deutlich ein richtungsabhängiger Einfluss auf die Messungen zeigt, ist für die Vergleichbarkeit der Standorte eine standardisierte Auswertung der Messergebnisse erforderlich. Ziel des Projektes FINO-Wind ist es, die Vergleichbarkeit der Daten der drei Standorte zu verbessern und den Datennutzern nach standardisierten Methoden qualitätsgeprüfte Daten zur Verfügung zu stellen. Es sollen deshalb Standardisierungsverfahren zur Auswertung der Winddaten entwickelt werden. Insbesondere die verschiedenen Masteffekte werden eingehend untersucht und durch Windkanalmessungen, Vergleich mit LiDAR-Daten, CFD (Computational Fluid Dynamics)-Modellierungen und Anwendung der UAM (Uniform ambient flow mast correction)-Methode abgeschätzt. Daraus werden Korrekturfaktoren für Windmessungen abgeleitet, die später angewendet werden sollen. Die Messdaten, die als 10-Minuten-Werte vorliegen, werden des Weiteren einer umfassenden und automatisierten Qualitätsprüfung unterzogen. Dabei durchlaufen die Daten in aufeinanderfolgenden Schritten formale, klimatologische, zeitliche, Wiederholungs- und Konsistenzprüfungen und werden nach erfolgreichem Abschluss jeder Sequenz mit spezifischen Qualitätsflags gekennzeichnet. Aus der Analyse und dem Vergleich der Instrumentierung in unterschiedlichen Höhen, der Installation und Ausrichtung sowie die Mastkonstruktionen sollen Empfehlungen herausgearbeitet werden, wie zukünftige Anwendungen im Bereich der Offshore-Windmessungen verbessert werden können

    Social Gerontology- Integrative and Territorial Aspects: A Citation Analysis of Subject Scatter and Database Coverage

    To determine the mix of resources used in social gerontology research, a citation analysis was conducted. A representative sample of citations was selected from three prominent gerontology journals and information was added to determine subject scatter and database coverage for the cited materials. Results indicate that a significant portion of gerontology research, even from a social science perspective, relies roughly equally on medical resources as it does social science resources. Furthermore, there is a small but defined core of literature constituting scholarly “territory” unique to gerontology. Analysis of database indexing indicated that broad, interdisciplinary databases provide more comprehensive coverage of the cited materials than do subject-specific databases

    STAT3 gain-of-function mutations connect leukemia with autoimmune disease by pathological NKG2Dhi CD8+T cell dysregulation and accumulation

    The association between cancer and autoimmune disease is unexplained, exemplified by T cell large granular lymphocytic leukemia (T-LGL) where gain-of-function (GOF) somatic STAT3 mutations correlate with co -exist-ing autoimmunity. To investigate whether these mutations are the cause or consequence of CD8+ T cell clonal expansions and autoimmunity, we analyzed patients and mice with germline STAT3 GOF mutations. STAT3 GOF mutations drove the accumulation of effector CD8+ T cell clones highly expressing NKG2D, the receptor for stress-induced MHC-class-I-related molecules. This subset also expressed genes for granzymes, perforin, interferon-y, and Ccl5/Rantes and required NKG2D and the IL-15/IL-2 receptor IL2RB for maximal accumula-tion. Leukocyte-restricted STAT3 GOF was sufficient and CD8+ T cells were essential for lethal pathology in mice. These results demonstrate that STAT3 GOF mutations cause effector CD8+ T cell oligoclonal accumu-lation and that these rogue cells contribute to autoimmune pathology, supporting the hypothesis that somatic mutations in leukemia/lymphoma driver genes contribute to autoimmune disease.Peer reviewe

    Bunyavirus requirement for endosomal K+ reveals new roles of cellular ion channels during infection

    In order to multiply and cause disease a virus must transport its genome from outside the cell into the cytosol, most commonly achieved through the endocytic network. Endosomes transport virus particles to specific cellular destinations and viruses exploit the changing environment of maturing endocytic vesicles as triggers to mediate genome release. Previously we demonstrated that several bunyaviruses, which comprise the largest family of negative sense RNA viruses, require the activity of cellular potassium (K+) channels to cause productive infection. Specifically, we demonstrated a surprising role for K+ channels during virus endosomal trafficking. In this study, we have used the prototype bunyavirus, Bunyamwera virus (BUNV), as a tool to understand why K+ channels are required for progression of these viruses through the endocytic network. We report three major findings: First, the production of a dual fluorescently labelled bunyavirus to visualize virus trafficking in live cells. Second, we show that BUNV traffics through endosomes containing high [K+] and that these K+ ions influence the infectivity of virions. Third, we show that K+ channel inhibition can alter the distribution of K+ across the endosomal system and arrest virus trafficking in endosomes. These data suggest high endosomal [K+] is a critical cue that is required for virus infection, and is controlled by cellular K+ channels resident within the endosome network. This highlights cellular K+ channels as druggable targets to impede virus entry, infection and disease

    Liposomes for cryopreservation of bovine sperm

    In this study, the effect of various unilamellar liposomes on cryopreservation of bovine spermatozoa has been investigated. Liposomes were composed of saturated lipids with various acyl chain lengths: DSPC (18:0), DPPC (16:0), DMPC (14:0), or DLPC (12:0). Alternatively, liposomes were prepared using unsaturated egg phosphatidylcholine (EPC) or DOPC (18:1, neutral), alone or in combination with lipids with various head groups: DOPS (negatively charged), DOPG (negatively charged), and DOPE (neutral). Fourier transform infrared spectroscopy studies showed that bovine sperm membranes display a gradual phase transition from 10 to 24 (o)C. Phase transition temperatures of the liposomes varied from -20 to +53 (o)C. Sperm was incubated in the presence of liposomes for either 6 or 24 h at 4 °C prior to freezing. Postfreeze survival rates were determined based on the percentage of progressively motile cells as well as the percentage of acrosome- and plasma membrane-intact cells. With DOPC liposomes a postthaw progressive motility of 43% was obtained compared with 59% using standard egg yolk freezing extender. Postthaw progressive motility increased up to 52% using DOPC:DOPG (9:1) liposomes, whereas DOPC:DOPS or DOPC:DOPE liposomes did not increase survival compared with DOPC liposomes. Among the saturated lipids, only DMPC was found to increase cryosurvival, up to 44% based on progressive motility. DLPC liposomes caused a complete loss in cell viability, already prior to freezing, whereas DPPC and DSPC liposomes neither positively nor negatively affected cryosurvival. Taken together, the higher postthaw survival obtained with DOPC:DOPG liposomes as compared with DOPC liposomes can likely be attributed to increased liposome-sperm interactions between the charged phosphatidylglycerol groups and charged regions in the sperm membranes.
