    Amylase release from streptolysin O-permeabilized pancreatic acinar cells. Effects of Ca2+, guanosine 5'-[gamma-thio]triphosphate, cyclic AMP, tetanus toxin and botulinum A toxin

    The molecular requirements for amylase release and the intracellular effects of botulinum A toxin and tetanus toxin on amylase release were investigated using rat pancreatic acinar cells permeabilized with streptolysin O. Micromolar concentrations of free Ca2+ evoked amylase release from these cells. Maximal release was observed in the presence of 30 microM free Ca2+. Ca(2+)-stimulated, but not basal, amylase release was enhanced by guanosine 5'-[gamma-thio]triphosphate (GTP[S]) (3-4 fold) or cyclic AMP (1.5-2 fold). Neither the two-chain forms of botulinum A toxin and tetanus toxin, under reducing conditions, nor the light chains of tetanus toxin, inhibited amylase release triggered by Ca2+, or combinations of Ca2+ + GTP[S] or Ca2+ + cAMP. The lack of inhibition was not due to inactivation of botulinum A toxin or tetanus toxin by pancreatic acinar cell proteolytic enzymes, as toxins previously incubated with permeabilized pancreatic acinar cells inhibited Ca(2+)-stimulated [3H]noradrenaline release from streptolysin O-permeabilized adrenal chromaffin cells. These data imply that clostridial neurotoxins inhibit a Ca(2+)-dependent mechanism which promotes exocytosis in neural and endocrine cells, but not in exocrine cells

    Kinetic and Spectroscopic Characterisation of Highly Reactive Methanesulfonates. Leaving Group Effects for Solvolyses and Comments on Geminal Electronic Effects Influencing SN_N1 Reactivity

    Highly reactive methanesulfonates (mesylates, ROMs) have been prepared from 1-phenylethanol. cyclohex-2-en-1-ol, diphenylmethanol and p-methoxybenzyl alcohol by treatment with methanesulfonyl chloride and triethylamine in dichloro- or trichloro-methane at - 20 to 0 °C. The mesylates. characterised in solution by 1^1H and 13^{13}C NMR at -20 °C, were obtained in satisfactory purity (ca. 95%) in cold solutions but they decomposed by reaction with chloride, triethylamine or the parent alcohol. Rate constants for solvolyses in aqueous acetone and aqueous ethanol have been determined by a fast response conductimetric method. Product selectivities for solvolyses of pmethoxybenzyl mesylate in aqueous ethanol and methanol at 0 °C have been determined by HPLC. From additional new or Iiterature kinetic data for solvolyses of corresponding bromides. chlorides and p-nitrobenzoates (OPNB). Br/CI. OMs/Br and OMs/OPNB rate ratios were calculated; the results are consistent with electronic effects stabilising the carbocationic transition states and increasing OMs/Br rate ratios for these SN 1 solvolyses; none of the evidence supports a geminal electronic effect on Br/CI rate ratios (e.g. caused by stabilisation of the initial state in pmethoxybenzyl chloride). Steric effects on ester /halide rate ratios for solvolyses of tertiary substrates are confirmed. Relative rates over a 1016^{16} range for ester and halide leaving groups are evaluated for solvolyses of 1-phenylethyl substrates in 80% ethanol-water. updating previous work by Noyce et al. (1972)

    The Duffin-Kemmer-Petiau formalism with vector and scalar potentials is used to point out a few misconceptions diffused in the literature. It is explicitly shown that the scalar coupling makes the DKP formalism not equivalent to the Klein-Gordon formalism or to the Proca formalism, and that the spin-1 sector of the DKP theory looks formally like the spin-0 sector. With proper boundary conditions, scattering of massive bosons in an arbitrary mixed vector-scalar square step potential is explored in a simple way and effects due to the scalar coupling on the particle-antiparticle production and localization of bosons are analyzed in some detail

    Spin 1 fields in Riemann-Cartan space-times "via" Duffin-Kemmer-Petiau theory

    We consider massive spin 1 fields, in Riemann-Cartan space-times, described by Duffin-Kemmer-Petiau theory. We show that this approach induces a coupling between the spin 1 field and the space-time torsion which breaks the usual equivalence with the Proca theory, but that such equivalence is preserved in the context of the Teleparallel Equivalent of General Relativity.Comment: 8 pages, no figures, revtex.     Relativistic Aharonov-Casher Phase in Spin One

    The Aharonov-Casher (AC) phase is calculated in relativistic wave equations of spin one. The AC phase has previously been calculated from the Dirac-Pauli equation using a gauge-like technique \cite{MK1,MK2}. In the spin-one case, we use Kemmer theory (a Dirac-like particle theory) to calculate the phase in a similar manner. However the vector formalism, the Proca theory, is more widely known and used. In the presence of an electromagnetic field, the two theories are `equivalent' and may be transformed into one another. We adapt these transformations to show that the Kemmer theory results apply to the Proca theory. Then we calculate the Aharonov-Casher phase for spin-one particles directly in the Proca formalism.Comment: 12 page

    Quantum Mechanics of Proca Fields

    We construct the most general physically admissible positive-definite inner product on the space of Proca fields. Up to a trivial scaling this defines a five-parameter family of Lorentz invariant inner products that we use to construct a genuine Hilbert space for the quantum mechanics of Proca fields. If we identify the generator of time-translations with the Hamiltonian, we obtain a unitary quantum system that describes first-quantized Proca fields and does not involve the conventional restriction to the positive-frequency fields. We provide a rather comprehensive analysis of this system. In particular, we examine the conserved current density responsible for the conservation of the probabilities, explore the global gauge symmetry underlying the conservation of the probabilities, obtain a probability current density, construct position, momentum, helicity, spin, and angular momentum operators, and determine the localized Proca fields. We also compute the generalized parity (\cP), generalized time-reversal (\cT), and generalized charge or chirality (\cC) operators for this system and offer a physical interpretation for its \cP\cT-, \cC-, and \cC\cP\cT-symmetries.Comment: Published version, typos fixed, a change in symbol, 1 fi

    On Equivalence of Duffin-Kemmer-Petiau and Klein-Gordon Equations

    A strict proof of equivalence between Duffin-Kemmer-Petiau (DKP) and Klein-Gordon (KG) theories is presented for physical S-matrix elements in the case of charged scalar particles interacting in minimal way with an external or quantized electromagnetic field. First, Hamiltonian canonical approach to DKP theory is developed in both component and matrix form. The theory is then quantized through the construction of the generating functional for Green functions (GF) and the physical matrix elements of S-matrix are proved to be relativistic invariants.     The usual approximation scheme is used to study the solution of the Duffin-Kemmer-Petiau (DKP) equation for a vector Yukawa potential in the framework of the parametric Nikiforov-Uvarov (NU) method. The approximate energy eigenvalue equation and the corresponding wave function spinor components are calculated for arbitrary total angular momentum in closed form. Further, the approximate energy equation and wave function spinor components are also given for case.     Building information modelling to cut disruption in housing retrofit

    There is a large stock of solid-wall homes in the UK with poor thermal insulation and low energy performance. Although the UK government has supported efforts to improve these buildings, the identification of appropriate technical solutions that effectively improve the existing stock remains challenging. This research investigates how four dimensional building information modelling (4D BIM) could improve the retrofit of social housing, specifically that of ‘no-fines’ solid-wall homes, through the development of what-if scenarios that enable the analysis of alternative solutions considering costs, energy performance and disruption to users. This paper focuses on the use of 4D building information models to evaluate disruption to end users. The results indicate that the development of such models supports a better understanding of the retrofit process on site. It also supports the definition of production plans with as minimal disruption as possible to users while delivering energy-oriented and cost-effective solutions
