    Comparison of epifluorescent viable bacterial count methods

    Two methods, the 2-(4-Iodophenyl) 3-(4-nitrophenyl) 5-phenyltetrazolium chloride (INT) method and the direct viable count (DVC), were tested and compared for their efficiency for the determination of the viability of bacterial populations. Use of the INT method results in the formation of a dark spot within each respiring cell. The DVC method results in elongation or swelling of growing cells that are rendered incapable of cell division. Although both methods are subjective and can result in false positive results, the DVC method is best suited to analysis of waters in which the number of different types of organisms present in the same sample is assumed to be small, such as processed waters.     Microbial biofilm studies of the environmental control and life support system water recovery test for Space Station Freedom

    NASA is developing a water recovery system (WRS) for Space Station Freedom to reclaim human waste water for reuse by astronauts as hygiene or potable water. A water recovery test (WRT) currently in progress investigates the performance of a prototype of the WRS. Analysis of biofilm accumulation, the potential for microbially influenced corrosion (MIC) in the WRT, and studies of iodine disinfection of biofilm are reported. Analysis of WRT components indicated the presence of organic deposits and biofilms in selected tubing. Water samples for the WRT contained acid-producing and sulfate-reducing organisms implicated in corrosion processes. Corrosion of an aluminum alloy was accelerated in the presence of these water samples; however, stainless steel corrosion rates were not accelerated. Biofilm iodine sensitivity tests using an experimental laboratory scale recycled water system containing a microbial check valve (MCV) demonstrated that an iodine concentration of 1 to 2 mg/L was ineffective in eliminating microbial biofilm. For complete disinfection, an initial concentration of 16 mg/L was required, which was gradually reduced by the MCV over 4 to 8 hours to 1 to 2 mg/L. This treatment may be useful in controlling biofilm formation

    Analysis of debris from Spacelab Space Life Sciences-1

    Airborne microbiological and particulate contamination generated aboard Spacelab modules is a potential safety hazard. In order to shed light on the characteristics of these contaminants, microbial and chemical/particulate analyses were performed on debris vacuumed from cabin and avionics air filters in the Space Life Sciences-1 (SLS-1) module of the Space Transportation System 40 (STS-40) mission 1 month after landing. The debris was sorted into categories (e.g., metal, nonmetal, hair/fur, synthetic fibers, food particles, insect fragments, etc.). Elemental analysis of particles was done by energy dispersive analysis of x rays (metals) and Fourier transform infrared spectroscopy (nonmetals). Scanning electron micrographs were done of most particles. Microbiological samples were grown on R2A culture medium and identified. Clothing fibers dominated the debris by volume. Other particles, all attributed to the crew, resulted from abrasions and impacts during missions operations (e.g., paint chips, plastic, electronic scraps and clothing fibers). All bacterial species identified are commonly found in the atmosphere or on the human body. Bacillus sp. was the most frequently seen bacterium. One of the bacterial species, Enterobacter agglomerans, could cause illness in crew members with depressed immune systems

    To develop a lunar outpost, we must understand the blowing of soil during launch and landing of the new Altair Lander. For example, the Apollo 12 Lunar Module landed approximately 165 meters from the deactivated Surveyor Ill spacecraft, scouring its surfaces and creating numerous tiny pits. Based on simulations and video analysis from the Apollo missions, blowing lunar soil particles have velocities up to 2000 m/s at low ejection angles relative to the horizon, reach an apogee higher than the orbiting Command and Service Module, and travel nearly the circumference of the Moon [1-3].     Muonium Decay

    Modifications of the mu+ lifetime in matter due to muonium (M = mu+ e-) formation and other medium effects are examined. Muonium and free mu+ decay spectra are found to differ at O(alpha m_e/m_mu) from Doppler broadening and O(alpha^2 m_e/m_mu) from the Coulomb bound state potential. However, both types of corrections are shown to cancel in the total decay rate due to Lorentz and gauge invariance respectively, leaving a very small time dilation lifetime difference, (tau_M - tau_mu+)/tau_mu+ = alpha^2 m_e^2/ 2m_mu^2 \simeq 6\times 10^-10, as the dominant bound state effect.     Genetic and Antigenic Analysis of the First A/New Caledonia/20/99-like H1N1 Influenza Isolates Reported in the Americas

    From February through May of 1999, 13 cases of Influenza A virus (FLUAV), type H1N1 were reported at a Department of Defense influenza surveillance sentinel site in Lima, Peru. Genetic and antigenic analysis by hemagglutination inhibition and direct nucleotide sequencing of the HA1 region of the hemagglutinin gene were performed on two isolates, A/Peru/1641/99 and A/Peru/1798/99. Both isolates were distinct from the Bayern/7/95-like viruses circulating in the Americas and closely related to a Beijing/262/95-like variant, A/New Caledonia/20/99. With the exception of travel-related cases, the detection of these isolates represents the first appearance of New Caledonia/20/99-like viruses in the Americas. Since the characterization of these Peru isolates, a number of New Caledonia/20/99-like viruses have been reported worldwide. For the 2000/01 and 2001/02 influenza seasons, the World Health Organization (WHO) has recommended the inclusion of A/New Caledonia/20/99 as the H1N1 vaccine component for both the southern and northern hemispheres

    Traces of explosive volcanic eruptions in the Upper Ordovician of the Siberian Platform

    © 2014, Estonian Academy Publishers. All rights reserved. Ordovician K-bentonite beds have a long history of investigation all around the world. They have been reported from Gondwana, the Argentine Precordillera, the Yangtze Platform, Laurentia, Baltica, and numerous terrains between Gondwana and Baltica, which now constitute a part of Europe. In recent years several K-bentonite beds have also been discovered in the Upper Ordovician of the Siberian Platform. This discovery is significant not only for their value in local and regional chronostratigraphic correlation but also for global geochronology, paleogeography, paleotectonic and paleoclimatic reconstructions. All in all, eight individual K-bentonite beds have been identified in the Baksian, Dolborian and Burian regional stages, which correspond roughly to the Upper Sandbian–Katian Global Stages. Zircon crystals from the uppermost K-bentonite bed within the Baksian regional stage provide a 206Pb/238U age of 450.58 ± 0.27 Ma. We will present preliminary results of the study of the three lowermost beds from the Baksian Regional Stage and suggest that the Taconic–Enisej (also spelled Yenisei or Yenisey) volcanic arc was continuous along the western margin of Siberia

Reproduced with permission from Astronomy & Astrophysics. © 2018 ESO.Radio continuum (RC) emission in galaxies allows us to measure star formation rates (SFRs) unaffected by extinction due to dust, of which the low-frequency part is uncontaminated from thermal (free-free) emission. We calibrate the conversion from the spatially resolved 140 MHz RC emission to the SFR surface density (ΣSFR\Sigma_{\rm SFR}) at 1 kpc scale. We used recent observations of three galaxies (NGC 3184, 4736, and 5055) from the LOFAR Two-metre Sky Survey (LoTSS), and archival LOw-Frequency ARray (LOFAR) data of NGC 5194. Maps were created with the facet calibration technique and converted to radio ΣSFR\Sigma_{\rm SFR} maps using the Condon relation. We compared these maps with hybrid ΣSFR\Sigma_{\rm SFR} maps from a combination of GALEX far-ultraviolet and Spitzer 24 μm\mu\rm m data using plots tracing the relation at 1.2×1.21.2\times 1.2-kpc2^2 resolution. The RC emission is smoothed with respect to the hybrid ΣSFR\Sigma_{\rm SFR} owing to the transport of cosmic-ray electrons (CREs). This results in a sublinear relation (ΣSFR)RC[(ΣSFR)hyb]a(\Sigma_{\rm SFR})_{\rm RC} \propto [(\Sigma_{\rm SFR})_{\rm hyb}]^{a}, where a=0.59±0.13a=0.59\pm 0.13 (140 MHz) and a=0.75±0.10a=0.75\pm 0.10 (1365 MHz). Both relations have a scatter of σ=0.3 dex\sigma = 0.3~\rm dex. If we restrict ourselves to areas of young CREs (α>0.65\alpha > -0.65; IνναI_\nu \propto \nu^\alpha), the relation becomes almost linear at both frequencies with a0.9a\approx 0.9 and a reduced scatter of σ=0.2 dex\sigma = 0.2~\rm dex. We then simulate the effect of CRE transport by convolving the hybrid ΣSFR\Sigma_{\rm SFR} maps with a Gaussian kernel until the RC-SFR relation is linearised; CRE transport lengths are l=1l=1-5 kpc. Solving the CRE diffusion equation, we find diffusion coefficients of D=(0.13D=(0.13-1.5)×1028cm2s11.5) \times 10^{28} \rm cm^2\,s^{-1} at 1 GeV. A RC-SFR relation at 1.41.4 GHz can be exploited to measure SFRs at redshift z10z \approx 10 using 140140 MHz observations.Peer reviewe

    Stochastic properties of coastal flooding events – Part 1: convolutional-neural-network-based semantic segmentation for water detection

    The frequency and intensity of coastal flooding is expected to accelerate in low-elevation coastal areas due to sea level rise. Coastal flooding due to wave overtopping affects coastal communities and infrastructure; however, it can be difficult to monitor in remote and vulnerable areas. Here we use a camera-based system to measure beach and back-beach flooding as part of the after-storm recovery of an eroded beach on the Texas coast. We analyze high-temporal resolution images of the beach using convolutional neural network (CNN)-based semantic segmentation to study the stochastic properties of flooding events. In the first part of this work, we focus on the application of semantic segmentation to identify water and overtopping events. We train and validate a CNN with over 500 manually classified images and introduce a post-processing method to reduce false positives. We find that the accuracy of CNN predictions of water pixels is around 90 % and strongly depends on the number and diversity of images used for training.</p
