Kora újkori látomásszövegek feltárása és kiadása: megjelent egy 18. századi látomás-szövegeket tartalmazó kötet. 2. Kutatás folyt a következő témákban: A boszorkányüldözés historiográfiája, statisztikája, jogi háttere, társadalmi kontextusa, teológiai vonatkozásai, kezdetei és megszűnése; a mágia dekriminalizációja a magyar joggyakorlatban; a boszorkányság hiedelemrendszere; a varázskönyvek használata és használói; a prédikációk, mint az elit és népi kultúra kommunikációs csatornái; a prédikációk ördög-képzetei. E témákkal részt vettünk több fontos magyar és nemzetközi konferencián, szerveztünk is ilyenket. 12 kutató közel 100 tanulmánya illetve forráskiadványa készült el. | Mentality, world-view and belief-systems of the early modern communities: 1. Exploration, identifying and publishing sources: Exploring documents of witchcraft cases in cities in Transylvania, Slovakia and in Hungary. Altogether we have acquired the material of approximately 400 trials amounting to 1500 pages. We have continued transcribing the case material collected during the previous cycle and preparing it for print. We have published a volume containing trial material in the counties and one on that of the towns. Exploring handwritten and printed sources of benedictions and exorcisms in Hungary: a volume is in the press. Exploring Calvinist preachings of the early modern age: a volume of preachings against superstitions and another volume against smoking are published. Exploring and publishing visions-literature: we have published a book containing 18th century visions. 2. Research was carried out on the following subjects: The historiography and comprehensive statistics of witch-hunt; the legal background and social context of witch-hunt, the theological aspects, religious and clerical history of witch-hunts; the belief system of witchcraft; the use and the users of magic books; preachings as channels of communication between the elite and popular culture, concepts of the Satan in the Calvinist preachings. We have participated at several important Hungarian and international conferences, and we also organised two major and several minor conferences on these areas of research. In this area 12 researchers accomplished approximately 100 papers resp. books

    Temporal response to harmonic driving in electroconvection

    The temporal evolution of the spatially periodic electroconvection (EC) patterns has been studied within the period of the driving ac voltage by monitoring the light intensity diffracted from the pattern. Measurements have been carried out on a variety of nematic systems, including those with negative dielectric and positive conductivity anisotropy, exhibiting "standard EC" (s-EC), those with both anisotropies negative exhibiting "non-standard EC" (ns-EC), as well as those with the two anisotropies positive. Theoretical predictions have been confirmed for stationary s-EC and ns-EC patterns. Transitions with Hopf bifurcation have also been studied. While traveling had no effect on the temporal evolution of dielectric s-EC, traveling conductive s-EC and ns-EC patterns exhibited a substantially altered temporal behavior with a dependence on the Hopf frequency. It has also been shown that in nematics with both anisotropies positive, the pattern develops and decays within an interval much shorter than the period, even at relatively large driving frequencies.Comment: 19 pages, 5 figure

    Saxagliptin Cardiotoxicity in Chronic Heart Failure: The Role of DPP4 in the Regulation of Neuropeptide Tone

    Dipeptidyl-peptidase-4 (DPP4) inhibitors are novel medicines for diabetes. The SAVORTIMI-53 clinical trial revealed increased heart-failure-associated hospitalization in saxagliptin-treated patients. Although this side effect could limit therapeutic use, the mechanism of this potential cardiotoxicity is unclear. We aimed to establish a cellular platform to investigate DPP4 inhibition and the role of its neuropeptide substrates substance P (SP) and neuropeptide Y (NPY), and to determine the expression of DDP4 and its neuropeptide substrates in the human heart. Western blot, radio-, enzyme-linked immuno-, and RNA scope assays were performed to investigate the expression of DPP4 and its substrates in human hearts. Calcein-based viability measurements and scratch assays were used to test the potential toxicity of DPP4 inhibitors. Cardiac expression of DPP4 and NPY decreased in heart failure patients. In human hearts, DPP4 mRNA is detectable mainly in cardiomyocytes and endothelium. Treatment with DPP4 inhibitors alone/in combination with neuropeptides did not affect viability but in scratch assays neuropeptides decreased, while saxagliptin co-administration increased fibroblast migration in isolated neonatal rat cardiomyocyte-fibroblast co-culture. Decreased DPP4 activity takes part in the pathophysiology of end-stage heart failure. DPP4 compensates against the elevated sympathetic activity and altered neuropeptide tone. Its inhibition decreases this adaptive mechanism, thereby exacerbating myocardial damage

    Prevalence of inflammatory bowel disease among coeliac disease patients in a Hungarian coeliac centre

    BACKGROUND: Celiac disease, Crohn disease and ulcerative colitis are inflammatory disorders of the gastrointestinal tract with some common genetic, immunological and environmental factors involved in their pathogenesis. Several research shown that patients with celiac disease have increased risk of developing inflammatory bowel disease when compared with that of the general population. The aim of this study is to determine the prevalence of inflammatory bowel disease in our celiac patient cohort over a 15-year-long study period. METHODS: To diagnose celiac disease, serological tests were used, and duodenal biopsy samples were taken to determine the degree of mucosal injury. To set up the diagnosis of inflammatory bowel disease, clinical parameters, imaging techniques, colonoscopy histology were applied. DEXA for measuring bone mineral density was performed on every patient. RESULTS: In our material, 8/245 (3,2 %) coeliac disease patients presented inflammatory bowel disease (four males, mean age 37, range 22-67), 6/8 Crohn's disease, and 2/8 ulcerative colitis. In 7/8 patients the diagnosis of coeliac disease was made first and inflammatory bowel disease was identified during follow-up. The average time period during the set-up of the two diagnosis was 10,7 years. Coeliac disease serology was positive in all cases. The distribution of histology results according to Marsh classification: 1/8 M1, 2/8 M2, 3/8 M3a, 2/8 M3b. The distribution according to the Montreal classification: 4/6 Crohn's disease patients are B1, 2/6 Crohn's disease patients are B2, 2/2 ulcerative colitis patients are S2. Normal bone mineral density was detected in 2/8 case, osteopenia in 4/8 and osteoporosis in 2/8 patients. CONCLUSIONS: Within our cohort of patients with coeliac disease, inflammatory bowel disease was significantly more common (3,2 %) than in the general population

    Cell-to-cell variability in troponin I phosphorylation in a porcine model of pacing-induced heart failure

    We tested the hypothesis that myocardial contractile protein phosphorylation and the Ca2+ sensitivity of force production are dysregulated in a porcine model of pacing-induced heart failure (HF). The level of protein kinase A (PKA)-dependent cardiac troponin I (TnI) phosphorylation was lower in the myocardium surrounding the pacing electrode (pacing site) of the failing left ventricle (LV) than in the controls. Immunohistochemical assays of the LV pacing site pointed to isolated clusters of cardiomyocytes exhibiting a reduced level of phosphorylated TnI. Flow cytometry on isolated and permeabilized cardiomyocytes revealed a significantly larger cell-to-cell variation in the level of TnI phosphorylation of the LV pacing site than in the opposite region in HF or in either region in the controls: the interquartile range (IQR) on the distribution histogram of relative TnI phosphorylation was wider at the pacing site (IQR = 0.53) than that at the remote site of HF (IQR = 0.42; P = 0.0047) or that of the free wall of the control animals (IQR = 0.36; P = 0.0093). Additionally, the Ca2+ sensitivities of isometric force production were higher and appeared to be more variable in single permeabilized cardiomyocytes from the HF pacing site than in the healthy myocardium. In conclusion, the level of PKA-dependent TnI phosphorylation and the Ca2+ sensitivity of force production exhibited a high cell-to-cell variability at the LV pacing site, possibly explaining the abnormalities of the regional myocardial contractile function in a porcine model of pacing-induced HF

    The Importance of Aquaporin 1 in Pancreatitis and Its Relation to the CFTR Cl- Channel

    Aquaporins (AQPs) facilitate the transepithelial water flow involved in epithelial fluid secretion in numerous tissues;however, their function in the pancreas is less characterized. Acute pancreatitis (AP) is a serious disorder in which specific treatment is still not possible. Accumulating evidence indicate that decreased pancreatic ductal fluid secretion plays an essential role in AP;therefore, the aim of this study was to investigate the physiological and pathophysiological role of AQPs in the pancreas. Expression and localization of AQPs were investigated by real-time PCR and immunocytochemistry, whereas osmotic transmembrane water permeability was estimated by the dye dilution technique, in Capan-1 cells. The presence of AQP1 and CFTR in the mice and human pancreas were investigated by immunohistochemistry. Pancreatic ductal HCO3- and fluid secretion were studied on pancreatic ducts isolated from wild-type (WT) and AQP1 knock out (KO) mice using microfluorometry and videomicroscopy, respectively. In vivo pancreatic fluid secretion was estimated by magnetic resonance imaging. AP was induced by intraperitoneal injection of cerulein and disease severity was assessed by measuring biochemical and histological parameters. In the mice, the presence of AQP1 was detected throughout the whole plasma membrane of the ductal cells and its expression highly depends on the presence of CFTR Cl- channel. In contrast, the expression of AQP1 is mainly localized to the apical membrane of ductal cells in the human pancreas. Bile acid treatment dose- and time-dependently decreased mRNA and protein expression of AQP1 and reduced expression of this channel was also demonstrated in patients suffering from acute and chronic pancreatitis. HCO3- and fluid secretion significantly decreased in AQP1 KO versus WT mice and the absence of AQP1 also worsened the severity of pancreatitis. Our results suggest that AQP1 plays an essential role in pancreatic ductal fluid and HCO3- secretion and decreased expression of the channel alters fluid secretion which probably contribute to increased susceptibility of the pancreas to inflammation
