Summary
Background: Medical imaging techniques can significantly aid correct diagnosis
and have become essential to advanced veterinary care. Infrared thermography
(IRT) is a novel and so far scarcely used tool in veterinary medicine in Hungary.
IRT records the emitted heat of the body in a thermal map. Abnormalities in the
thermal signature can indicate potential anomalies.
Objectives: By highlighting the capabilities of IRT in equine medicine, the authors
aim to popularise the method among practitioners. The advantages of IRT and its
potentials as a supplementary diagnostic tool are reviewed. Limitations of the
technique are also discussed, supplemented by guidance for how to overcome
them. Five case studies are presented to illustrate the versatility of IRT and pro vide examples of body parts that can be efficiently examined.
Materials and Methods: An ICI™ 7640 IR PAD 640P infrared camera was used to
record the thermal signature in areas of veterinary interest in five horses. Thermo grams were then evaluated to detect and localise potential pathologies.
Results and Discussion: IRT was successfully used to identify (1) an asym metrical thermal signature on the horse’s back due to an ill-fitted saddle; (2)
increased temperature in the area of left splenius cervicis; (3) a fractured molar in
the premaxilla-maxilla region; (4) a chip fracture in the proximal sesamoid bone
of the right forelimb; and (5) inflammation of the hoof on the right forelimb. IRT
can be used efficiently as a comparatively inexpensive and rapid pre-diagnostics
tool to localise or establish a strong suspicion of certain abnormalities before
directing the patient towards further imaging diagnostics (e.g. PET/CT, MRI). The
authors advise to include thermography in veterinary education and advocate the
establishment of a standardised framework for veterinary IRT concerning imaging
methods, regulations, and environmental conditions
Digital microscopy and image analysis techniques for pathogenicity of filamentous fungi
Az invazív gombák micéliális morfológiai változásainak vizsgálata
elengedhetetlen a fonalas gombák patogenitásának mélyebb megértésében. Egyrészt ez
hasznos ipari szempontból, mivel a micéliális folyamatok bizonyos típusai összefüggésben
vannak az ipari fermentációk termelékenységével, másrészt orvosbiológiai szempontból is
fontos, mivel az invazív gombás megbetegedések egyre gyakoribbak és halálos fertőzéseket
is okoznak immunhiányos betegekben. A morfológiai változások kvalitatív és kvantitatív
dinamikai paramétereinek meghatározása azonban továbbra is jelentős kihívást jelent a
tudomány jelenlegi eszközeivel. A spórák letapadási dinamikájának pontosabb megismerése
lehetőséget biztosíthat a fermentációs rendszerek termelékenységének növelésére, illetve az
invazív gombás megbetegedések elleni védekezésre.
A statikus és dinamikus képelemző technikák felhasználhatók olyan
gombapopulációk vizsgálatára is, melyekben az egyedi spórák jelentősége nő meg a
tenyésztési ciklus alatt és ezek kitapadási és/vagy spórázási dinamikája követhető.
Megfelelő időbeli felbontás mellett a hosszú távú videómikroszkópos vizsgálatok
segítségével definiált mikrobiológiai paraméterek valós időben és hosszú távon
vizsgálhatók. Ilyen paraméterek a spóranövekedés, letapadás, osztódás, biofilm képződés,
konfluencia, motilitás. Optimális esetben ezek a paraméterek a megfigyelés teljes
időtartama alatt követhetőek és ez által összefüggéseikben is vizsgálhatóak lesznek. A
videómikroszkópos rendszer által készített felvételek feldolgozása során nyert adatok
specifikusan adaptált dinamikus képanalízisét alkalmazva, egy vizsgált osztódási vagy
szaporodási szakasz pontosabban modellezhetővé válhat.
A képanalízis során kvalitatív és kvantitatív szempontok szerint is
tanulmányozhatjuk az egyes folyamatokat. A videómikroszkópia egy olyan, hosszú ideig
(hetekig) folytatható monitoring-módszer, amelyet a célszerűen kiválasztott digitális
képelemzéssel és adatfeldolgozással kombinálva olyan kísérleti elrendezést kapunk
melynek fő előnye, hogy a változások időbeli lefutása dinamikusan vizsgálhatóvá, illetve
modellezhetővé válik, így hűen tükrözi az időben lezajló folyamatokat. In order to get a deeper understanding of the pathogenicity of filamentous fungi it is
necessary to examine the mycelial morphological changes of invasive fungal species. It is,
on the one hand, important from an industrial point of view, as certain types of mycelial
processes may affect the productivity of industrial fermentation, while on the other hand it
is also important from a medical point of view, because the number of patients with
invasive fungal diseases is on the rise, and some fungi can even cause lethal infections in
immunocompromised patients. However, the determination of qualitative and quantitative
dynamic parameters of morphological changes still represents a major challenge due to the
limitations of our tools. A deeper understanding of the dynamics of spore adherence may
help us increase the productivity of fermentation systems and fight against invasive fungal
diseases.
Static and dynamic image analysis techniques can also be used for studying fungal
populations in which the significance of individual spores grows during the growth phase,
and their adherence and/or germination dynamics can be observed.
With an appropriate temporal resolution, microbiological parameters defined with the help
of long-term video microscopy can be studied real time and over a longer period. Such
parameters include spore growth, adherence, division, appearance of biofilm, confluence,
and motility. Optimally, these parameters can be monitored throughout the entire
observation, which allows us to study their interrelations as well. The specially adapted
dynamic image analysis of data collected through processing images captured by the video
microscopy system may allow us to model a specific division or reproduction phase more
accurately.
In the course of image analysis we can study individual processes based on qualitative and
quantitative aspects as well. Video microscopy is a long-term (up to weeks) monitoring
system that, when combined with carefully chosen digital image analysis and data
processing, results in an experimental setup, the main advantage of which is that it allows us
to study and model the dynamics of changes in time, providing us with a real or actual
picture of temporal changes.N
Képalkotó és képelemző technikák prezentálása Aspergillus nidulanson, mint modellszervezeten
Az elmúlt évtizedekben a mikroszkopikus gombák a biotechnológiai és ipari jellegű kutatások közszereplőjévé váltak. Ezen fejlesztések miatt vált fontossá a gombák élettani folyamatainak alaposabb megismerése. Ezek a kutatások lehetnek az alapjai a gombák különböző gyógyszeripari és mezőgazdasági alkalmazásának.
A gombák akár a természetben, akár a fermentációs iparban gyakran szembesülnek az éhezés jelenségével, ami különböző mértékű autolízishez vezet. Ez utóbbi, egy olyan nagy hidroláz aktivitással kísért sejtpusztulási folyamat, amely magában foglalja a sejtfal lebomlását illetve különböző mértékű degradációját. Ezen éhezési folyamat változását vizsgáltuk pásztázó elektronmikroszkóppal, és ezeket a képeket elemeztük.
Képelemzés alatt a számunkra értékes adatok kinyerését értjük a kép(ek)ből, főleg a digitális képfeldolgozási technikák segítségével. A nagy mennyiségű adat miatt a képelemzésekhez nélkülözhetetlen a számítógép használata, mely segítségével elvégezhetjük a különböző kvalitatív és kvantitatív méréseket a képeken. A scanning elektronmikroszkóppal készített képeknek képelemző módszerekkel való feldolgozása adatokat biztosít az autolizált gomba hifa összehasonlító vizsgálatához.
Az adatokat feldolgozva grafikonos és táblázatos formában reprezentálhatjuk a kapott eredményeket. A kapott adatokat kiértékelve kapunk információt az ágens, vizsgált anyag hatásáról különböző nagyításokkal készített képeken.MSc/MAbiológusg
