Functional characterization and discovery of modulators of SbMATE, the agronomically important aluminium tolerance transporter from Sorghum bicolor.
About 50% of the world's arable land is strongly acidic (pHââ€â5). The low pH solubilizes root-toxic ionic aluminium (Al3+) species from clay minerals, driving the evolution of counteractive adaptations in cultivated crops. The food crop Sorghum bicolor upregulates the membrane-embedded transporter protein SbMATE in its roots. SbMATE mediates efflux of the anionic form of the organic acid, citrate, into the soil rhizosphere, chelating Al3+ ions and thereby imparting Al-resistance based on excluding Al+3 from the growing root tip. Here, we use electrophysiological, radiolabeled, and fluorescence-based transport assays in two heterologous expression systems to establish a broad substrate recognition profile of SbMATE, showing the proton and/or sodium-driven transport of 14C-citrate anion, as well as the organic monovalent cation, ethidium, but not its divalent analog, propidium. We further complement our transport assays by measuring substrate binding to detergent-purified SbMATE protein. Finally, we use the purified membrane protein as an antigen to discover native conformation-binding and transport function-altering nanobodies using an animal-free, mRNA/cDNA display technology. Our results demonstrate the utility of using Pichia pastoris as an efficient eukaryotic host to express large quantities of functional plant transporter proteins. The nanobody discovery approach is applicable to other non-immunogenic plant proteins
A CCD Search for Variable Stars in the Open Cluster NGC 6611
We present the results of the variability survey in the young open
cluster NGC 6611 based on observations obtained during 34 nights spanning one
year. In total, we found 95 variable stars. Most of these stars are classified
as periodic and irregular pre-main sequence (PMS) stars. The analysis of the
2MASS photometry and four-colour IRAC photometry revealed 165 Class II
young stellar sources, 20 of which are irregular variables and one is an
eclipsing binary. These classifications, complemented by UKIDSS
photometry and VPHAS photometry, were used to identify 24
candidates for classical T Tauri stars and 30 weak-lined T Tauri stars. In
addition to the PMS variables, we discovered eight Scuti candidates.
None of these were previously known. Furthermore, we detected 17 eclipsing
binaries where two were previously known. Based on the proper motions provided
by the Gaia EDR3 catalogue, we calculated the cluster membership probabilities
for 91 variable stars. For 61 variables, a probability higher than 80% was
determined, which makes them cluster members. Only 25 variables with a
probability less than 20% were regarded to be non-members.Comment: 30 pages, 31 figure
Microlensing of Relativistic Knots in the Quasar HE1104-1805
We present 3 years of photometry of the ``Double Hamburger'' lensed quasar,
HE1104-1805, obtained on 102 separate nights using the OGLE 1.3-m telescope.
Both the A and B images show variations, but with substantial differences in
the lighcurves at all time delays. At the 310 day delay reported by Wisotzki
and collaborators the difference lightcurve has an rms amplitude of 0.060 mag.
The structure functions for the A and B images are quite different, with image
A more than twice as variable as image B (a factor of 4 in structure function)
on timescales of less than a month. Adopting microlensing as a working
hypothesis for the uncorrelated variability, the short timescale argues for the
relativistic motion of one or more components of the source. We argue that the
small amplitude of the fluctuations is due to the finite size of the source

Preliminary results for RR Lyrae stars and Classical Cepheids from the Vista Magellanic Cloud (VMC) Survey
The Vista Magellanic Cloud (VMC, PI M.R. Cioni) survey is collecting
-band time series photometry of the system formed by the two Magellanic
Clouds (MC) and the "bridge" that connects them. These data are used to build
-band light curves of the MC RR Lyrae stars and Classical Cepheids and
determine absolute distances and the 3D geometry of the whole system using the
-band period luminosity (), the period - luminosity - color ()
and the Wesenhiet relations applicable to these types of variables. As an
example of the survey potential we present results from the VMC observations of
two fields centered respectively on the South Ecliptic Pole and the 30 Doradus
star forming region of the Large Magellanic Cloud. The VMC -band light
curves of the RR Lyrae stars in these two regions have very good photometric
quality with typical errors for the individual data points in the range of
0.02 to 0.05 mag. The Cepheids have excellent light curves (typical
errors of 0.01 mag). The average magnitudes derived for both types
of variables were used to derive relations that are in general good
agreement within the errors with the literature data, and show a smaller

Suppression of phase transitions and glass phase signatures in mixed cation halide perovskites
Cation engineering provides a route to control the structure and properties of hybrid halide perovskites, which has resulted in the highest performance solar cells based on mixtures of Cs, methylammonium, and formamidinium. Here, we present a multi-technique experimental and theoretical study of structural phase transitions, structural phases and dipolar dynamics in the mixed methylammonium/dimethylammonium MA1-xDMAxPbBr3 hybrid perovskites (0ââ€âxââ€â1). Our results demonstrate a significant suppression of the structural phase transitions, enhanced disorder and stabilization of the cubic phase even for a small amount of dimethylammonium cations. As the dimethylammonium concentration approaches the solubility limit in MAPbBr3, we observe the disappearance of the structural phase transitions and indications of a glassy dipolar phase. We also reveal a significant tunability of the dielectric permittivity upon mixing of the molecular cations that arises from frustrated electric dipoles
The OGLE View of Microlensing towards the Magellanic Clouds. II. OGLE-II SMC data
The primary goal of this paper is to provide the evidence that can either
prove or falsify the hypothesis that dark matter in the Galactic halo can clump
into stellar-mass compact objects. If such objects existed, they would act as
lenses to external sources in the Magellanic Clouds, giving rise to an
observable effect of microlensing. We present the results of our search for
such events, based on the data from the second phase of the OGLE survey
(1996-2000) towards the SMC. The data set we used is comprised of 2.1 million
monitored sources distributed over an area of 2.4 square degrees. We found only
one microlensing event candidate, however its poor quality light curve limited
our discussion on the exact distance to the lensing object.
Given a single event, taking the blending (crowding of stars) into account
for the detection efficiency simulations, and deriving the HST-corrected number
of monitored stars, the microlensing optical depth is tau=(1.55+-1.55)10e-7.
This result is consistent with the expected SMC self-lensing signal, with no
need of introducing dark matter microlenses. Rejecting the unconvincing event
leads to the upper limit on the fraction of dark matter in the form of MACHOs
to f<20 per cent for deflectors' masses around 0.4 Msun and f<11 per cent for
masses between 0.003 and 0.2 Msun (95 per cent confidence limit). Our result
indicates that the Milky Way's dark matter is unlikely to be clumpy and form
