    Translation of mRNA for human lymphotoxin in microinjected Xenopus oocytes

    AbstractSynthesis and secretion of biologically active human lymphotoxin (LT) can be detected in Xenopus laevis oocytes following their inoculation with poly(A+) RNA from human stimulated peripheral blood lymphocytes, but not in oocytes inoculated with RNA from unstimulated lymphocytes or from fibroblastoid cells. In size-fractionating mRNA of stimulated lymphocytes most LT activity is found to be coded for by RNA with an approximate sedimentation value of 19 S

    Topic Modeling and Text Analysis for Qualitative Policy Research

    This paper contributes to a critical methodological discussion that has direct ramifications for policy studies: how computational methods can be concretely incorporated into existing processes of textual analysis and interpretation without compromising scientific integrity. We focus on the computational method of topic modeling and investigate how it interacts with two larger families of qualitative methods: content and classification methods characterized by interest in words as communication units and discourse and representation methods characterized by interest in the meaning of communicative acts. Based on analysis of recent academic publications that have used topic modeling for textual analysis, our findings show that different mixed‐method research designs are appropriate when combining topic modeling with the two groups of methods. Our main concluding argument is that topic modeling enables scholars to apply policy theories and concepts to much larger sets of data. That said, the use of computational methods requires genuine understanding of these techniques to obtain substantially meaningful results. We encourage policy scholars to reflect carefully on methodological issues, and offer a simple heuristic to help identify and address critical points when designing a study using topic modeling.Peer reviewe

    A randomised controlled trial of calcium channel blockade (CCB) with Amlodipine For the treatment oF subcortical ischaEmic vasCular demenTia (AFFECT):study protocol

    Background Vascular dementia is the second most common cause of dementia affecting over seven million people worldwide, yet there are no licensed treatments. There is an urgent need for a clinical trial in this patient group. Subcortical ischaemic vascular dementia is the most common variant of vascular dementia. This randomised trial will investigate whether use of calcium channel blockade with amlodipine, a commonly used agent, can provide the first evidence-based pharmacological treatment for subcortical ischaemic vascular dementia. Methods/Design This is a randomised controlled trial of calcium channel blockade with Amlodipine For the treatment oF subcortical ischaEmic vasCular demenTia (AFFECT) to test the hypothesis that treatment with amlodipine can improve outcomes for these patients in a phase IIb, multi-centre, double-blind, placebo-controlled randomised trial. The primary outcome is the change from baseline to 12 months in the Vascular Dementia Assessment Scale cognitive subscale (VADAS-cog). Secondary outcomes include cognitive function, executive function, clinical global impression of change, change in blood pressure, quantitative evaluation of lesion accrual based on magnetic resonance imaging (MRI), health-related quality of life, activities of daily living, non-cognitive dementia symptoms, care-giver burden and care-giver health-related quality of life, cost-effectiveness and institutionalisation. A total of 588 patients will be randomised in a 1:1 ratio to either amlodipine or placebo, recruited from sites across the UK and enrolled in the trial for 104 weeks. Discussion There are no treatments licensed for vascular dementia. The most common subtype is subcortical ischaemic vascular dementia (SIVD). This study is designed to investigate whether amlodipine can produce benefits compared to placebo in established SIVD. It is estimated that the numbers of people with VaD and SIVD will increase globally in the future and the results of this study should inform important treatment decisions.     O objetivo deste artigo é fazer um balanço da literatura sobre os Estudos de Comunidade (EC) no Brasil, destacando o contexto intelectual em que foram produzidos e os debates que suscitaram, especialmente entre as décadas de 1940 e 1960. Método de pesquisa em comunidades, oriundo da Antropologia, em investigações sobre o processo de mudança social, os EC estiveram em voga nos Estados Unidos entre os anos 1920 e 1950, tendo papel fundamental na institucionalização das Ciências Sociais no Brasil. Destacamos três aspectos na produção desses estudos no país, iniciando com uma abordagem conceitual: definições, origem e sua relação com temáticas marcantes nos anos 1950. Em seguida, privilegiamos os aspectos relacionados ao papel dos EC na história das Ciências Sociais no Brasil, com destaque para o processo de institucionalização das Ciências Sociais. Por fim, revisitamos os debates que tais estudos geraram entre os cientistas sociais quanto ao padrão de trabalho sociológico a ser desenvolvido naquele momento.<br>The purpose of this article is to examine the literature about Community Studies(CS) in Brazil, highlighting their intellectual context of production and the debates evoked by them, especially between the 1940's and 1960's. As a research method arising from Anthropology on communities in social change process, CS was used in United States between 1920's and 1950's, and played a fundamental role in the institutionalization of Social Sciences in Brazil. Three aspects are highlighted on CS production: first, in a conceptual approach, we discuss their definitions, origins and relation to main topics in the 1950's. The second aspect is related to the role of CS in the history of Social Sciences, especially their institutionalization process. By the end, we revisit the debates between social scientists about the sociological work standard to be developed at that time
