    How well do CMIP5 Earth System Models simulate present climate conditions in Europe and Africa?

    This study provides a comprehensive evaluation of seven Earth System Models (ESMs) from the Coupled Model Intercomparison Project Phase 5 in present climate conditions from a downscaling perspective, taking into account the requirements of both statistical and dynamical approaches. ECMWF's ERA-Interim reanalysis is used as reference for an evaluation of circulation, temperature and humidity variables on daily timescale, which is based on distributional similarity scores. To additionally obtain an estimate of reanalysis uncertainty, ERA-Interim's deviation from the Japanese Meteorological Agency JRA-25 reanalysis is calculated. Areas with considerable differences between both reanalyses do not allow for a proper assessment, since ESM performance is sensitive to the choice of reanalysis. For use in statistical downscaling studies, ESM performance is computed on the grid-box scale and mapped over a large spatial domain covering Europe and Africa, additionally highlighting those regions where significant distributional differences remain even for the centered/zero-mean time series. For use in dynamical downscaling studies, performance is specifically assessed along the lateral boundaries of the three CORDEX domains defined for Europe, the Mediterranean Basin and Africa.S.B. would like to thank the CSIC JAE-PREDOC programme for financial support.     Semi-automated level design via auto-playtesting for handheld casual game creation

    We provide a proof of principle that novel and engaging mobile casual games with new aesthetics, game mechanics and player interactions can be designed and tested directly on the device for which they are intended. We describe the Gamika iOS application which includes generative art assets; a design interface enabling the making of physics-based casual games containing multiple levels with aspects ranging from Frogger-like to Asteroids-like and beyond; a configurable automated playtester which can give feedback on the playability of levels; and an automated fine-tuning engine which searches for level parameterisations that enable the game to pass a battery of tests, as evaluated by the auto-playtester. Each aspect of the implementation represents a baseline with much room for improvement, and we present some experimental results and describe how these will guide the future directions for Gamika

    Certification Labels for Trustworthy AI: Insights From an Empirical Mixed-Method Study

    Auditing plays a pivotal role in the development of trustworthy AI. However, current research primarily focuses on creating auditable AI documentation, which is intended for regulators and experts rather than end-users affected by AI decisions. How to communicate to members of the public that an AI has been audited and considered trustworthy remains an open challenge. This study empirically investigated certification labels as a promising solution. Through interviews (N = 12) and a census-representative survey (N = 302), we investigated end-users' attitudes toward certification labels and their effectiveness in communicating trustworthiness in low- and high-stakes AI scenarios. Based on the survey results, we demonstrate that labels can significantly increase end-users' trust and willingness to use AI in both low- and high-stakes scenarios. However, end-users' preferences for certification labels and their effect on trust and willingness to use AI were more pronounced in high-stake scenarios. Qualitative content analysis of the interviews revealed opportunities and limitations of certification labels, as well as facilitators and inhibitors for the effective use of labels in the context of AI. For example, while certification labels can mitigate data-related concerns expressed by end-users (e.g., privacy and data protection), other concerns (e.g., model performance) are more challenging to address. Our study provides valuable insights and recommendations for designing and implementing certification labels as a promising constituent within the trustworthy AI ecosystem

    Introduction: Most countries want to make the transition to increased or even exclusive use of renewable energy. To achieve this goal, how can individuals be persuaded to use more renewable electricity? For example, does the way energy companies communicate so-called consumer subsidies matter in this regard, and if so, which communication strategy is best? For example, is a monetary promotion (e.g., cashback) better than a non-monetary one (e.g., gift)? Methods: In a total of four studies (with a total of more than 1700 participants), we investigated what type of promotion most influenced the choice of a renewable energy product, varying, for example, the environmental friendliness of the renewable energy product. Results: The monetary promotion (e.g., get $35 back through subsidies) appeared to be the most successful; it significantly increased the choice of the renewable electricity product (i.e., between 12–22%). However, this result was only evident when the subsidized renewable product was not the product already preferred by most individuals. Other measures, such as the willingness to pay (WTP), showed no differential effects. Discussion: Overall, the observed pattern suggests that promoting renewable energy choices, is similar to promoting donations to a charity. Accordingly, the description of the consumer subsidy as a monetary promotion (i.e., cashback or negative labeling) is most effective in terms of promotion. However, the effect of monetary promotions seems to diminish if the subsidized product is already the product preferred by most consumers. Nevertheless, the use of monetary promotions can encourage the transition to renewable energy

    Pharmacokinetic profile of irinotecan in patients with chronic kidney disease:Two cases and literature review

    Aims: There are limited pharmacokinetic data on the use of irinotecan in patients with reduced glomerular filtration rate (GFR) and no haemodialysis. In this case report, we present 2 cases and review the current literature. Methods: The dose of irinotecan in both patients was reduced pre-emptively due to reduced GFR. The first patient had her irinotecan dose reduced to 50%, but was nevertheless admitted to hospital because of irinotecan-induced toxicity, including gastrointestinal toxicity and neutropenic fever. The dose was reduced further to 40% for the second cycle; however, the patient was again admitted to the hospital, and irinotecan was stopped indefinitely. The second patient also had his irinotecan dose reduced to 50% and was admitted to the emergency department for gastrointestinal toxicity after the first cycle. However, irinotecan could be administered in the same dose in later cycles. Results: The area under the curve to infinity of irinotecan and SN-38 in the first patient were comparable to those of an individual receiving 100% dose intensity. The area under the curve to infinity of irinotecan and SN-38 in patient 2 in both cycles were slightly less than reference values. Furthermore, clearance values of irinotecan and SN-38 in our patients were comparable to those without renal impairment. Conclusion: Our case report suggests that reduced GFR may not significantly affect the clearance of irinotecan and SN-38, but can still result in clinical toxicity. Reduced initial dosing seems indicated in this patient population. Further research is needed to fully understand the relationship between reduced GFR, pharmacokinetics, and toxicity of irinotecan and SN-38.</p

    A healthier beverage choice is based on a subjective assessment of sweet taste

    Despite promising interventions to lower people’s daily sugar consumption, such as health- or taste-focused labels, the consumption of sugar-sweetened beverages (SSBs) continues to rise. To improve the effectiveness of existing labels, the way people process sugar amounts in grams (g) as displayed on beverages seems to merit elucidation. For example, do people perceive the difference in the amount of sugar, and thus in the subjective sweet taste, between two beverages according to Weber’s law? Additionally, is that perceived difference the cause of their beverage choice? In order to investigate these questions, participants in this online experiment first had to estimate the sugar difference between two beverages based on grams and then decide whether they would switch to a lower-sugar beverage. We found that participants’ different estimates followed Weber’s law. The choice of the lower-sugar beverage, however, depended on how large they personally perceived that difference. In other words, the choice was independent of the ratio. These results show that future labels, rather than indicating the total amount of sugar, should indicate whether the reduction, for example in the amount of sugar compared to another beverage, was perceived as significant by others

    Variable rate spraying in varied micro-meteorological conditions

    This study evaluated effects of crosswind on the variable rate sprayer application treatments spray coverage and deposition on different citrus canopy sizes.  The axial-fan airblast sprayer retrofitted with variable liquid- and air-assist rates was field-tested with different crosswind conditions on small (about 2 m tall and < 1.5 m wide) and medium-sized (about 3 m tall and < 2.5 m wide) canopies.  Crosswinds of 1.3, 2.7, and 4.0 ms-1 on the canopies being sprayed were generated using the stationary conical air shaker as the air blower unit.  Water sensitive papers (WSPs) were used to collect droplet deposits and image processing software was used to analyze the WSPs scanned at 600 dpi.  Percent spray coverage on the WSPs was found to be one of the most suited parameters to evaluate the effectiveness of spray application treatments.  Overall, the variable rate spray application treatments had comparable spray coverage on respective canopies (front, middle, and across WSP locations in the canopy) during all crosswind conditions.  For both types of canopies, spray coverage was higher on the canopy front and decreased as the spray penetrated inside (i.e. canopy middle) and across.  Due to coalescing, larger droplets (Dv,0.5 [volume median diameter] = 838 to 2,624 µm) were formed on the WSPs located on canopy front, whereas coalescing reduced as the spray penetrated inside (Dv,0.5 = 391 to 1,625 µm on canopy middle) and across the canopy (Dv,0.5 = 307 to 508 µm).    Keywords: airblast sprayer, adjustable air-assistance, crosswind, spray coverage, citru

    Feasibility of pharmacy-initiated pharmacogenetic screening for CYP2D6 and CYP2C19

    PURPOSE: Our purpose was to investigate the feasibility of pharmacy-initiated pharmacogenetic (PGt) screening in primary care with respect to patient willingness to participate, quality of DNA collection with saliva kits, genotyping, and dispensing data retrieved from the pharmacy. METHODS: Polypharmacy patients aged >60 years who used at least one drug with Anatomical Therapeutic Chemical (ATC) code N06AA01-N06AX19 (antidepressants), A02BC01-A02BC05 (proton-pump inhibitors), N05AA01-N05AH04 (antipsychotics), or C07AB02 (metoprolol) in the preceding 2 years were randomly selected. DNA was collected with saliva kits and genotyped for CYP2D6 and CYP2C19 with the AmpliChip. Pharmacy dispensing records were retrieved and screened for drugs interacting with the patient's CYP2D6 and CYP2C19 genotype by using the evidence-based PGt guidelines from the Dutch Pharmacogenetics Working Group. RESULTS: Out of the 93 invited patients, 54 (58.1%) provided informed consent. Nine saliva samples (16.7%) contained too little DNA. Call rates for CYP2D6 and CYP2C19 were 93.3% and 100%, respectively. Frequencies of genotype-predicted phenotype were 2.4%, 38.1%, 54.8%, and 4.8% for CYP2D6 poor metabolizers (PM), intermediate metabolizers (IM), extensive metabolizers (EM), and ultrarapid metabolizers (UM) respectively. For CYP2C19 genotype-predicted phenotype, frequencies were 2.2%, 15.6%, and 82.2% for PM, IM, and EM, respectively. CONCLUSIONS: This study shows that pharmacy-initiated PGt screening is feasible for a primary care setting
