    Efficacy and safety of combination of ulinastatin and meglumine cyclic adenosine monophosphate in the treatment of acute myocardial infarction, and its effect on serum levels of hs-CRP, cTnI and CK

    Purpose: To determine the efficacy and safety of a combination of ulinastatin and meglumine cyclic adenosine monophosphate (cAMP) in the treatment of acute myocardial infarction (AMI), and its effect on serum levels of hypersensitive-c-reactive protein (hs-CRP), cardiac troponin I (cTnI), creatine kinase (CK).Methods: A total of 90 AMI patients admitted to The Second Affiliated Hospital of Qiqihar Medical College, Qiqihar City, Heilongjiang Province, China from January 2019 to January 2020 were selected and randomized (in a 1:1 ration) into control group and study group. Patients in the two groups received meglumine cAMP, while those in the study group were, in addition, treated with ulinastatin. The two groups were compared with regard to clinical efficacy, cardiac function indices, serum biochemical indices, incidence of drug-related side effects, duration and number of episodes of angina pectoris, and levels of neuroendocrine hormones.Results: The study group exhibited remarkably higher treatment effectiveness and cardiac function indices compared to the control group (p < 0.05). However, lower levels of serum biochemical indices, lower total incidence of drug toxicity, smaller number and shorter duration of angina pectoris, and lower levels of panel reactive antibodies (PRA) were observed in the study when compared to control group (p< 0.001).Conclusion: Treatment of AMI patients with the combination of ulinastatin and meglumine cAMP significantly reduces the clinical symptoms of the patients, with remarkable efficacy and high safety. Furthermore, it down-regulates serum levels of hs-CRP, cTnI and CK. Thus, the combination treatment seems superior to the conventional therapy

    DNMT3a in the hippocampal CA1 is crucial in the acquisition of morphine self‐administration in rats

    Drug‐reinforced excessive operant responding is one fundamental feature of long-lasting addiction‐like behaviors and relapse in animals. However, the transcriptional regulatory mechanisms responsible for the persistent drug‐specific (not natural rewards) operant behavior are not entirely clear. In this study, we demonstrate a key role for one of the de novo DNA methyltransferase, DNMT3a, in the acquisition of morphine self‐administration (SA) in rats. The expression of DNMT3a in the hippocampal CA1 region but not in the nucleus accumbens shell was significantly up‐regulated after 1‐ and 7‐day morphine SA (0.3 mg/kg/infusion) but not after the yoked morphine injection. On the other hand, saccharin SA did not affect the expression of DNMT3a or DNMT3b. DNMT inhibitor 5‐aza‐2‐deoxycytidine (5‐aza) microinjected into the hippocampal CA1 significantly attenuated the acquisition of morphine SA. Knockdown of DNMT3a also impaired the ability to acquire the morphine SA. Overall, these findings suggest that DNMT3a in the hippocampus plays an important role in the acquisition of morphine SA and may be a valid target to prevent the development of morphine addiction.     Self‐Assembly of Therapeutic Peptide into Stimuli‐Responsive Clustered Nanohybrids for Cancer‐Targeted Therapy

    Clinical translation of therapeutic peptides, particularly those targeting intracellular protein–protein interactions (PPIs), has been hampered by their inefficacious cellular internalization in diseased tissue. Therapeutic peptides engineered into nanostructures with stable spatial architectures and smart disease targeting ability may provide a viable strategy to overcome the pharmaceutical obstacles of peptides. This study describes a strategy to assemble therapeutic peptides into a stable peptide–Au nanohybrid, followed by further self‐assembling into higher‐order nanoclusters with responsiveness to tumor microenvironment. As a proof of concept, an anticancer peptide termed ÎČ‐catenin/Bcl9 inhibitors is copolymerized with gold ion and assembled into a cluster of nanohybrids (pCluster). Through a battery of in vitro and in vivo tests, it is demonstrated that pClusters potently inhibit tumor growth and metastasis in several animal models through the impairment of the Wnt/ÎČ‐catenin pathway, while maintaining a highly favorable biosafety profile. In addition, it is also found that pClusters synergize with the PD1/PD‐L1 checkpoint blockade immunotherapy. This new strategy of peptide delivery will likely have a broad impact on the development of peptide‐derived therapeutic nanomedicine and reinvigorate efforts to discover peptide drugs that target intracellular PPIs in a great variety of human diseases, including cancer.A strategy for clinical translation of therapeutic peptides by assembling them into a stable peptide–Au nanohybrid, followed by further self‐assembling into higher‐order nanoclusters with responsiveness to the tumor microenvironment, is presented.     First sodium laser guide star asterism launching platform in China on 1.8m telescope at Gaomeigu Observatory

    The application of sodium laser guide star is the key difference between modern adaptive optics system and traditional adaptive optics system. Especially in system like multi-conjugate adaptive optics, sodium laser guide star asterism which is formed by several laser guide stars in certain pattern is required to probe more atmospheric turbulence in different directions. To achieve this, a sodium laser guide star asterism launching platform is required. In this paper, we will introduce the sodium laser guide star asterism launching platform built and tested on the 1.8m telescope of the Gaomeigu Observatory. The platform has two functions: one is to compare the performance of sodium laser guide stars generated by different lasers at the same place; the other is to generate sodium laser guide star asterism with adjustable shape. The field test results at the beginning of 2021 verify the important role of the platform, which is also the first time to realize sodium laser guide star asterism in China

    An Updated Search of Steady TeV γ−\gamma-Ray Point Sources in Northern Hemisphere Using the Tibet Air Shower Array

    Using the data taken from Tibet II High Density (HD) Array (1997 February-1999 September) and Tibet-III array (1999 November-2005 November), our previous northern sky survey for TeV γ−\gamma-ray point sources has now been updated by a factor of 2.8 improved statistics. From 0.0∘0.0^{\circ} to 60.0∘60.0^{\circ} in declination (Dec) range, no new TeV γ−\gamma-ray point sources with sufficiently high significance were identified while the well-known Crab Nebula and Mrk421 remain to be the brightest TeV γ−\gamma-ray sources within the field of view of the Tibet air shower array. Based on the currently available data and at the 90% confidence level (C.L.), the flux upper limits for different power law index assumption are re-derived, which are approximately improved by 1.7 times as compared with our previous reported limits.Comment: This paper has been accepted by hepn

    Synthesis and Growth Mechanism of Ni Nanotubes and Nanowires

    Highly ordered Ni nanotube and nanowire arrays were fabricated via electrodeposition. The Ni microstructures and the process of the formation were investigated using conventional and high-resolution transmission electron microscope. Herein, we demonstrated the systematic fabrication of Ni nanotube and nanowire arrays and proposed an original growth mechanism. With the different deposition time, nanotubes or nanowires can be obtained. Tubular nanostructures can be obtained at short time, while nanowires take longer time to form. This formation mechanism is applicable to design and synthesize other metal nanostructures and even compound nanostuctures via template-based electrodeposition

    Attitudes on the donation of human embryos for stem cell research among Chinese IVF patients and students

    Bioethical debates on the use of human embryos and oocytes for stem cell research have often been criticized for the lack of empirical insights into the perceptions and experiences of the women and couples who are asked to donate these tissues in the IVF clinic. Empirical studies that have investigated the attitudes of IVF patients and citizens on the (potential) donation of their embryos and oocytes have been scarce and have focused predominantly on the situation in Europe and Australia. This article examines the viewpoints on the donation of embryos for stem cell research among IVF patients and students in China. Research into the perceptions of patients is based on in-depth interviews with IVF patients and IVF clinicians. Research into the attitudes of students is based on a quantitative survey study (n=427). The empirical findings in this paper indicate that perceptions of the donation of human embryos for stem cell research in China are far more diverse and complex than has commonly been suggested. Claims that ethical concerns regarding the donation and use of embryos and oocytes for stem cell research are typical for Western societies but absent in China cannot be upheld. The article shows that research into the situated perceptions and cultural specificities of human tissue donation can play a crucial role in the deconstruction of politicized bioethical argumentation and the (often ill-informed) assumptions about “others” that underlie socio-ethical debates on the moral dilemmas of technology developments in the life sciences

    Observation of ηc→ωω\eta_c\to\omega\omega in J/Ïˆâ†’ÎłÏ‰Ï‰J/\psi\to\gamma\omega\omega

    Using a sample of (1310.6±7.0)×106(1310.6\pm7.0)\times10^6 J/ψJ/\psi events recorded with the BESIII detector at the symmetric electron positron collider BEPCII, we report the observation of the decay of the (11S0)(1^1 S_0) charmonium state ηc\eta_c into a pair of ω\omega mesons in the process J/Ïˆâ†’ÎłÏ‰Ï‰J/\psi\to\gamma\omega\omega. The branching fraction is measured for the first time to be B(ηc→ωω)=(2.88±0.10±0.46±0.68)×10−3\mathcal{B}(\eta_c\to\omega\omega)= (2.88\pm0.10\pm0.46\pm0.68)\times10^{-3}, where the first uncertainty is statistical, the second systematic and the third is from the uncertainty of B(J/Ïˆâ†’ÎłÎ·c)\mathcal{B}(J/\psi\to\gamma\eta_c). The mass and width of the ηc\eta_c are determined as M=(2985.9±0.7±2.1) M=(2985.9\pm0.7\pm2.1)\,MeV/c2c^2 and Γ=(33.8±1.6±4.1) \Gamma=(33.8\pm1.6\pm4.1)\,MeV.Comment: 13 pages, 6 figure

    Measurement of proton electromagnetic form factors in e+e−→ppˉe^+e^- \to p\bar{p} in the energy region 2.00-3.08 GeV

    The process of e+e−→ppˉe^+e^- \rightarrow p\bar{p} is studied at 22 center-of-mass energy points (s\sqrt{s}) from 2.00 to 3.08 GeV, exploiting 688.5~pb−1^{-1} of data collected with the BESIII detector operating at the BEPCII collider. The Born cross section~(σppˉ\sigma_{p\bar{p}}) of e+e−→ppˉe^+e^- \rightarrow p\bar{p} is measured with the energy-scan technique and it is found to be consistent with previously published data, but with much improved accuracy. In addition, the electromagnetic form-factor ratio (∣GE/GM∣|G_{E}/G_{M}|) and the value of the effective (∣Geff∣|G_{\rm{eff}}|), electric (∣GE∣|G_E|) and magnetic (∣GM∣|G_M|) form factors are measured by studying the helicity angle of the proton at 16 center-of-mass energy points. ∣GE/GM∣|G_{E}/G_{M}| and ∣GM∣|G_M| are determined with high accuracy, providing uncertainties comparable to data in the space-like region, and ∣GE∣|G_E| is measured for the first time. We reach unprecedented accuracy, and precision results in the time-like region provide information to improve our understanding of the proton inner structure and to test theoretical models which depend on non-perturbative Quantum Chromodynamics
