    Detection of aerolysin and hemolysin genes in Aeromonas spp. isolated from environmental and shellfish sources by polymerase chain reaction

    Polymerase chain reaction (PCR) technique was used to assay for the detection of specific genes in the genomes of the Aeromonas spp. isolated from environmental and shellfish sources, particularly aero and hlyA genes, responsible for aerolysin and hemolysin toxins production in this genus. The results showed that: (i) the 1500 bp amplicon of the hlyA gene was detected in 20/38 of the Aeromonas hydrophila, 13/38 of the A. caviae and 6/9 of the A. veronii biovar sobria isolates; (ii) the 690 bp amplicon of the aero gene was detected in 20/38 of A. hydrophila, 17/38 of A. caviae and 6/9 of A. veronii biovar sobria isolates; (iii) the nucleotide blast results of aerolysin gene sequences of the representative strains of A. hydrophila, A. caviae and A. veronii biovar sobria revealed a high homology of 94%, 95% and 95% with published sequences, respectively and; (iv) the protein blast showed 97%, 94% and 96% homology when compared to the published sequences, respectively. The finding of A. hydrophila virulence genes in other members of the genus Aeromonas, may give a new perspective to the significance of these results. The method described here may be a useful detection tool to assist in further investigation of aero and hlyA genes in the genus Aeromonas, especially for food microbiologist

    Wavelets operational methods for fractional differential equations and systems of fractional differential equations

    In this thesis, new and effective operational methods based on polynomials and wavelets for the solutions of FDEs and systems of FDEs are developed. In particular we study one of the important polynomial that belongs to the Appell family of polynomials, namely, Genocchi polynomial. This polynomial has certain great advantages based on which an effective and simple operational matrix of derivative was first derived and applied together with collocation method to solve some singular second order differential equations of Emden-Fowler type, a class of generalized Pantograph equations and Delay differential systems. A new operational matrix of fractional order derivative and integration based on this polynomial was also developed and used together with collocation method to solve FDEs, systems of FDEs and fractional order delay differential equations. Error bound for some of the considered problems is also shown and proved. Further, a wavelet bases based on Genocchi polynomials is also constructed, its operational matrix of fractional order derivative is derived and used for the solutions of FDEs and systems of FDEs. A novel approach for obtaining operational matrices of fractional derivative based on Legendre and Chebyshev wavelets is developed, where, the wavelets are first transformed into corresponding shifted polynomials and the transformation matrices are formed and used together with the polynomials operational matrices of fractional derivatives to obtain the wavelets operational matrix. These new operational matrices are used together with spectral Tau and collocation methods to solve FDEs and systems of FDEs

However, it is categorized as difficult to machine material due to its unique combination of properties which include high strength, toughness, wear resistance and low thermal conductivity.     The stepper motor movement process produced different frequency and pulses. This research explained about the frequency and pulses for the stepper motor movement by using Programmable Logic Controller (PLC) as research method. The study was done to find the suitable frequency and pulses for stepper motor movement by developing a prototype stepper motor controller system. The pulse frequency used did not affected the distance of moving load in the stepper motor operations. The increasing number of pulse frequency only will affect the time taken for the stepper motor to complete its operations. The result showed that number of pulse frequency at high operation was 5000 Hz. Pulse number reacted as a manipulated variable that affected both factor which is time taken of stepper motor operation and the distance of moving load

    Alternative fuel sources such as biofuels are needed in order to overcome environmental problem caused by fossil fuel consumption. Currently, most biofuel are produced from land based crops and there is a possibility that marine biomass such as macroalgae can be an alternative source for biofuel production. The carbohydrate in macroalgae can be broken down into simple sugar through thermo-chemical hydrolysis and enzymatic hydrolysis. Dilute-acid hydrolysis was believed to be the most available and affordable method. However, the process may release inhibitors which would affect alcohol yield from fermentation. Thus, this work was aimed at investigating if it is possible to avoid this critical pre-treatment step in macroalgae fermentation process by using Sasad, a local Sabahan fermentation agent and to compare the yield with rice wine fermentation. This work hoped to determine and compare the alcohol content from Gracilaria sp. and rice fermentation with Sasad. Rice fermentation was found containing ethanol and 2 - methyl - 1 - propanol. Fermentation of Gracilaria sp. had shown the positive presence of 3 - methyl - 1 - butanol. It was found that Sasad can be used as a fermentation agent for bioalcohol production from Gracilaria sp. without the need for a pretreatment step. However further investigations are needed to determine if pre-treatment would increase the yield of alcohol

    Contribution of forest floor fractions to carbon storage and abundance patterns of arbuscular mycorrhizal fungal colonisation in a tropical montane forest

    Forest floor carbon stocks, which include different components of litter, hemic and sapric materials, have not been empirically quantified in tropical montane forest, although they influence soil carbon (C) pools. To date, the contribution of arbuscular mycorrhizae in C sequestration potentials in tropical montane forests have not been clearly investigated. This study determined the amount of C stocks in the different decomposing layers of forest floor, mainly litter, hemic and sapric materials. The abundance of arbuscular mycorrhizal root colonisation differed among forest floor fractions. Forest floor was measured for depth, area density, dry mass and carbon fraction separately in Sungai Kial Forest Reserve, Pahang, Malaysia to calculate C stocks. Percentages of root colonisation in the hemic and sapric materials were investigated. The results showed that forest floor C stocks were significantly higher in hemic (5 Mg C ha−1) and sapric (7.7 Mg C ha−1) compared with the litter fragments (1.5 Mg C ha−1). Mycorrhizal root colonisation was significantly higher (75%) in the toeslope compared with the summit area in the hemic materials.     Histomorphometric changes in the perirenal adipocytes of adrenalectomized rats treated with dexamethasone

    INTRODUCTION: Prolonged steroid treatment administered to any patient can cause visceral obesity, which is associated with metabolic disease and Cushing's syndrome. Glucocorticoids have a profound negative effect on adipose tissue mass, giving rise to obesity, which in turn is regulated by the 11&#946;-hydroxysteroid dehydrogenase type 1 enzyme. Adrenalectomized rats treated with dexamethasone exhibited an increase in visceral fat deposition but not in body weight. OBJECTIVES: The main aim of this study was to determine the effect of dexamethasone on the histomorphometric characteristics of perirenal adipocytes of adrenalectomized, dexamethasone-treated rats (ADR+Dexa) and the association of dexamethasone treatment with the expression and activity of 11 &#946;-hydroxysteroid dehydrogenase type 1 (11 &#946;-hydroxysteroid dehydrogenase type 1). METHODS: A total of 20 male Sprague Dawley rats were divided into 3 groups: a baseline control group (n = 6), a sham-operated group (n = 7) and an adrenalectomized group (n=7). The adrenalectomized group was given intramuscular dexamethasone (ADR+Dexa) 2 weeks post adrenalectomy, and the rats from the sham-operated group were administered intramuscular vehicle (olive oil). RESULTS: Treatment with 120 &#956;g/kg intramuscular dexamethasone for 8 weeks resulted in a significant decrease in the diameter of the perirenal adipocytes (p<0.05) and a significant increase in the number of perirenal adipocytes (p<0.05). There was minimal weight gain but pronounced fat deposition in the dexamethasone-treated rats. These changes in the perirenal adipocytes were associated with high expression and dehydrogenase activity of 11&#946;-hydroxysteroid dehydrogenase type 1. CONCLUSIONS: In conclusion, dexamethasone increased the deposition of perirenal fat by hyperplasia, which causes increases in the expression and dehydrogenase activity of 11 &#946;-hydroxysteroid dehydrogenase type 1 in adrenalectomized rats

    The effect of holmium on microstructure and mechanical properties of light metal alloy

    The thesis project investigates the effect of Holmium addition on the microstructure and mechanical properties of cast ZRE1 magnesium alloy. In the advancement of automotive and aerospace industries, the next generation of light metal alloy is magnesium due to its manufacturing abilities and lightweight material. The main limitation of Mg alloys is the ability to improve its mechanical properties. Rare earth elements were used as alloying elements for improving the mechanical properties and producing a new Mg-RE alloy with modified structure and strength. In this studies, it demonstrates the addition of RE at specific amount, which could be considered as main alloy element that may lead to extend limitation of RE application. The amount of Holmium (Ho) at 0.2, 0.4 and 0.6 wt. % were added separately to ZRE1 magnesium alloy. The Optical Microsope, FESEM coupled with EDX, and XRD were used to investigate the microstructure and mechanical properties, including tensile and hardness tests. The results showed that the Ho content at 0.2wt. % increased the ultimate tensile strength and yield strength compared to others Ho composition. Finally, the hardness value was increased and the additive had a significant improvement of mechanical properties, thus the ZRE1 magnesium alloy has been improved with Holmium addition

    The Usability of C++ Interactive Self-Assessment Quiz (i-SAQ)

    The rising of technologies these days affected most of the sectors in the country including education.  Traditional assessment method gives difficulty to both students and lecturers in term of learning and teaching process. Therefore, this study proposed an online quizzes application called i-SAQ (C++ Interactive Self-Assessment Quiz) which adopt formative assessment method to help students and lecturers in their learning and teaching process. The feature of i-SAQ which offers immediate feedback while answering the quiz attracts students in having a hassle-free way of discovering knowledge. This feature also helps lecturers in handling large number of students in term of giving the feedback from formative assessment. The usability test was conducted to a group of 64 students to test the functionality of i-SAQ by using questionnaire approach. From the analysis, most of the students gave highest points for every questionnaire usability factors which shows that i-SAQ is one useful and interesting learning support tool
