    Isotopic compositions of lipid biomarker compounds in estuarine plants and surface sediments

    We examined the isotopic compositions of fatty acids, sterols, and hydrocarbons isolated from three coastal macrophytes (Zostera marina, Spartina alterniflora, and Juncus roemerianus) in order to investigate the relative contribution of these vascular plants as sources of organic matter in coastal sediments such as Cape Lookout Eight, North Carolina. On average, lipid biomarker compounds extracted from the plants were depleted in C-13 by 3-5 parts per thousand relative to delta(13)C total organic carbon (TOC). However, individual compounds within each lipid class varied by up to 5.6 parts per thousand. Trends in the isotopic compositions of lipids were consistent with delta(13)C(TOC); compounds obtained from Z. marina were the most enriched in C-13 and those from J. roemerianus were the most depleted. The range in isotopic abundances and molecular compositions of the sediments was greater than that obtained from the plants, indicating that additional, presently unidentified sources of organic matter contribute to the Cape Lookout Eight sediments. Similarity between the signatures for suspended particulate matter and the sediments indicates that much of the sedimentary organic matter is derived from algal and bacterial sources. Bacterial sources of organic matter are Likely greater during summer/early fall, and incorporation of C-13-enriched bacterial biomass may contribute to observed seasonal shifts in delta(13)C(TOC) in the surficial sediments

    Bioethanol Production from Brewers Spent Grains Using a Fungal Consolidated Bioprocessing (CBP) Approach.

    Production of bioethanol from brewers spent grains (BSG) using consolidated bioprocessing (CBP) is reported. Each CBP system consists of a primary filamentous fungal species, which secretes the enzymes required to deconstruct biomass, paired with a secondary yeast species to ferment liberated sugars to ethanol. Interestingly, although several pairings of fungi were investigated, the sake fermentation system (A. oryzae and S. cerevisiae NCYC479) was found to yield the highest concentrations of ethanol (37 g/L of ethanol within 10 days). On this basis, 1 t of BSG (dry weight) would yield 94 kg of ethanol using 36 hL of water in the process. QRT-PCR analysis of selected carbohydrate degrading (CAZy) genes expressed by A. oryzae in the BSG sake system showed that hemicellulose was deconstructed first, followed by cellulose. One drawback of the CBP approach is lower ethanol productivity rates; however, it requires low energy and water inputs, and hence is worthy of further investigation and optimisation

    Testing for HFE-related haemochromatosis

    HFE-haemochromatosis is a genetic disorder resulting from mutations of the HFE gene. It primarily affects people of Northern European descent. Clinical manifestations result from the progressive deposition of iron into various organs including the liver. An elevated serum ferritin concentration greater than 300 microgram/L and a transferrin saturation of greater than 45% will identify almost all patients with HFE-haemochromatosis. HFE genotyping confirms the diagnosis. In some patients, liver biopsy may still be necessary as the degree of hepatic fibrosis has prognostic implications

    SURFACE MEASURED ACCELERATIONS DURING CRICKET FAST-BOWLING

    The aim of this research was to quantify the magnitude and timing of surface measured accelerations during the fast-bowling action. Eleven males performed 6 maximum velocity deliveries with accelerometers positioned over: both ankles; knees; hips; L5; L1; and the C7 vertebrae. Accelerometer signals exhibited decreased peak and increased time to peak acceleration from the ankle to the C7 sensor. Even when distal accelerations were largest at front foot contact, the body was still able to dissipate more than 90% of the acceleration. Active and passive mechanisms such as joint compliance and spinal compression within the body therefore likely contribute to the progressive attenuation of accelerations. The effects of such compliance on investigations of the intersegmental forces and moments during cricket fast-bowling via inverse dynamics warrants further investigation. The data we obtained using APCI-MS correlated significantly with high-performance liquid chromatography analysis whilst offering the analyst minimal sample preparation and faster sample throughput

    Proton Pump Inhibitors Inhibit PHOSPHO1 Activity and Matrix Mineralisation In Vitro

    Proton pump inhibitors (PPIs) have been associated with an increased risk of fragility fractures in pharmaco-epidemiological studies. The mechanism is unclear, but it has been speculated that by neutralising gastric acid, they may reduce intestinal calcium absorption, causing secondary hyperparathyroidism and bone loss. Here we investigated that hypothesis that the skeletal effects of PPI might be mediated by inhibitory effects on the bone-specific phosphatase PHOSPHO1. We found that the all PPIs tested inhibited the activity of PHOSPHO1 with IC50 ranging between 0.73 ”M for esomeprazole to 19.27 ”M for pantoprazole. In contrast, these PPIs did not inhibit TNAP activity. We also found that mineralisation of bone matrix in primary osteoblast cultures was inhibited by several PPIs in a concentration dependent manner. In contrast, the histamine-2 receptor antagonists (H2RA) nizatidine, famotidine, cimetidine and ranitidine had no inhibitory effects on PHOSPHO1 activity. Our experiments show for the first time that PPIs inhibit PHOSPHO1 activity and matrix mineralisation in vitro revealing a potential mechanism by which these widely used drugs are associated with the risk of fractures

    Computer mouse movement patterns: A potential marker of mild cognitive impairment

    AbstractIntroductionSubtle changes in cognitively demanding activities occur in mild cognitive impairment (MCI) but are difficult to assess with conventional methods. In an exploratory study, we examined whether patterns of computer mouse movements obtained from routine home computer use discriminated between older adults with and without MCI.MethodsParticipants were 42 cognitively intact and 20 older adults with MCI enrolled in a longitudinal study of in-home monitoring technologies. Mouse pointer movement variables were computed during one week of routine home computer use using algorithms that identified and characterized mouse movements within each computer use session.ResultsMCI was associated with making significantly fewer total mouse moves (P < .01) and making mouse movements that were more variable, less efficient, and with longer pauses between movements (P < .05). Mouse movement measures were significantly associated with several cognitive domains (P values <.01–.05).DiscussionRemotely monitored computer mouse movement patterns are a potential early marker of real-world cognitive changes in MCI

    Maximising high solid loading enzymatic saccharification yield from acid-catalysed hydrothermally-pretreated brewers spent grain

    Enzyme saccharification of pretreated brewers spent grains (BSG) was investigated, aiming at maximising glucose production. Factors investigated were; variation of the solids loadings at different cellulolytic enzyme doses, reaction time, higher energy mixing methods, supplementation of the cellulolytic enzymes with additional enzymes (and cofactors) and use of fed-batch methods. Improved slurry agitation through aerated high-torque mixing offered small but significant enhancements in glucose yields (to 53 ± 2.9 g/L and 45% of theoretical yield) compared to only 41 ± 4.0 g/L and 39% of theoretical yield for standard shaking methods (at 15% w/v solids loading). Supplementation of the cellulolytic enzymes with additional enzymes (acetyl xylan esterases, ferulic acid esterases and α-L- arabinofuranosidases) also boosted achieved glucose yields to 58 – 69 ± 0.8 - 6.2 g/L which equated to 52 - 58% of theoretical yield. Fed-batch methods also enhanced glucose yields (to 58 ± 2.2 g/L and 35% of theoretical yield at 25% w/v solids loading) compared to non-fed-batch methods. From these investigations a novel enzymatic saccharification method was developed (using enhanced mixing, a fed-batch approach and additional carbohydrate degrading enzymes) which further increased glucose yields to 78 ± 4.1 g/L and 43% of theoretical yield when operating at high solids loading (25% w/v)
