    Psychoeducational module to promote knowledge and reduce stigma towards recreational marijuana users

    Stigma can affect many aspects of life for those who are subject to stigmatization. As contact with stigmatized groups increases, less stigmatization occurs. Marijuana is one of the most commonly used illegal substances and is growing in popularity. Relatively few studies have examined the relations between stigma and specific substance use or particular characteristics of non-users that may influence stigma. Additionally, current anti-stigma interventions targeted at the general population have been largely unsuccessful in the reduction of stigma. The current study assesses a 25- minute online psychoeducational module designed to increase knowledge and reduce the stigma of recreational marijuana users in a more efficient, practical, engaging, and costeffective way which can be easily applied to any anti-stigma intervention program. The module surveyed 201 college students from a public Midwestern university where participants were randomly assigned to one of two groups; a module group (N = 104) and a control group (N = 97). Specifically, the module was shown to increase knowledge regarding marijuana, however, there was little to no impact on stigma (i.e., preferred social distance, perceived dangerousness, negative emotions regarding marijuana use). Additionally, findings indicate strong associations between the level of familiarity, the level of contact, and stigma. Specifically, as the level of familiarity or level of contact increases, less stigmatization occurs. In addition, limited support was found for the moderating role of gender, past marijuana use, and age on the established association between the level of familiarity and level of contact on stigma levels. Furthermore, we explored the relationship between the level of familiarity, the level of contact, and the likelihood of future use. After establishing this strong relationship, we examined whether sex, any past marijuana use, and age would moderate this potential relationship. Results indicate that when familiarity is high, individuals will have a higher likelihood of future use if they already have a prior history of marijuana use. However, there were little to no differences in the likelihood of future use when familiarity was low, even with a prior history of marijuana use. No other characteristics seemed to impact this relationship. Findings have implications for anti-stigma interventions

    Increased Mood Disorder Symptoms, Perceived Stress, and Alcohol Use Among College Students During the COVID-19 Pandemic

    The COVID-19 pandemic caused significant disruption during the spring of 2020. Many college students were told to leave campus at spring break and to complete the semester remotely. This study evaluates effects of this disruption on student well-being. Measures of psychological symptoms, perceived stress, and alcohol use during the pandemic were completed by 148 students in spring 2020 and 352 students in fall 2020 at a university in the southeastern U.S. Results from both cohorts were compared to 240 students who completed the same measures in the fall 2019 semester. Participants in spring 2020 reported more mood disorder symptoms, perceived stress, and alcohol use than did pre-pandemic participants and worry about COVID-19 was negatively associated with well-being. By fall 2020 symptoms had largely returned to pre-pandemic levels. In general, White students reported a greater effect of the pandemic on well-being than did African American students. Young adults appear to be less vulnerable to the most serious medical complications associated with COVID-19, but nonetheless experience psychological effects from the pandemic. Universities and practitioners who work with college students can help young adults manage their symptoms and avoid behaviors like risky alcohol use when confronted with stressors such as the COVID-19 pandemic

    Economic Evaluation of Coastal Land Loss in Louisiana

    Louisiana has lost approximately 1,880 square miles of land over the past eighty years. Projections suggest that in a future without action, the next fifty years could result in the loss of 1,750 additional square miles of land area. As land loss continues, a large portion of the natural and man-made capital stocks of coastal Louisiana will be at greater risk of damage, either from land loss or from the associated increase in storm damage. We estimate the replacement cost of capital stock directly at risk from land loss ranges from approximately 2.1billionto2.1 billion to 3.5 billion with economic activity at risk ranging from 2.4billionto2.4 billion to 3.1 billion in output. Increases in storm damage to capital stock range from 8.7billiontoasmuchas8.7 billion to as much as 133 billion with associated disruptions to economic activity ranging from an additional 1.9billionto1.9 billion to 23 billion in total lost output

    The Future of Data Management Planning: Tools, Policies, and Players

    DMPonline and the DMPTool are well-established tools for data management planning. As the software of each matures and the user communities grow, we turn our attention to issues of sustainability, culture change, and international collaboration. Here we outline strategies for addressing these issues. We propose to build a new, global framework for data management planning that links plans to researchers, funders, publications, data, and other components of the research lifecycle. By refocusing our efforts from promoting the creation of data management plans (DMPs) to comply with funder requirements to supporting the creation of good DMPs that can be implemented, we seek to further enable the open scholarship revolution, advancing science and society

    The role of taxonomic expertise in interpretation of metabarcoding studies

    Abstract The performance of DNA metabarcoding approaches for characterizing biodiversity can be influenced by multiple factors. Here, we used morphological assessment of taxa in zooplankton samples to develop a large barcode database and to assess the congruence of taxonomic identification with metabarcoding under different conditions. We analysed taxonomic assignment of metabarcoded samples using two genetic markers (COI, 18S V1–2), two types of clustering into molecular operational taxonomic units (OTUs, ZOTUs), and three methods for taxonomic assignment (RDP Classifier, BLASTn to GenBank, BLASTn to a local barcode database). The local database includes 1042 COI and 1108 18S (SSU) barcode sequences, and we added new high-quality sequences to GenBank for both markers, including 109 contributions at the species level. The number of phyla detected and the number of taxa identified to phylum varied between a genetic marker and among the three methods used for taxonomic assignments. Blasting the metabarcodes to the local database generated multiple unique contributions to identify OTUs and ZOTUs. We argue that a multi-marker approach combined with taxonomic expertise to develop a curated, vouchered, local barcode database increases taxon detection with metabarcoding, and its potential as a tool for zooplankton biodiversity surveys

    The role of the melanoma gene MC1R in Parkinson disease and REM sleep behavior disorder

    The MC1R gene, suggested to be involved in Parkinson disease (PD) and melanoma, was sequenced in PD patients (n=539) and controls (n=265) from New-York, and PD patients (n=551), rapid eye movement sleep behavior disorder (RBD) patients (n=351) and controls (n=956) of European ancestry. Sixty-eight MC1R variants were identified, including 7 common variants with frequency>0.01. None of the common variants was associated with PD or RBD in the different regression models. In a meta-analysis with fixed-effect model, the p.R160W variant was associated with an increased risk for PD (OR=1.22, 95%CI 1.02-1.47, p=0.03) but with significant heterogeneity (p=0.048). Removing one study that introduced the heterogeneity resulted in nonsignificant association (OR=1.11, 95%CI 0.92-1.35, p=0.27, heterogeneity p=0.57). Rare variants had similar frequencies in patients and controls (10.54% and 10.15%, respectively, p=0.75), and no cumulative effect of carrying more than one MC1R variant was found. The current study does not support a role for the MC1R p.R160W and other variants in susceptibility for PD or RBD

    Genomic and microbiological analyses of iron acquisition pathways among respiratory and environmental nontuberculous mycobacteria from Hawai’i

    As environmental opportunistic pathogens, nontuberculous mycobacteria (NTM) can cause severe and difficult to treat pulmonary disease. In the United States, Hawai’i has the highest prevalence of infection. Rapid growing mycobacteria (RGM) such as Mycobacterium abscessus and M. porcinum and the slow growing mycobacteria (SGM) including M. intracellulare subspecies chimaera are common environmental NTM species and subspecies in Hawai’i. Although iron acquisition is an essential process of many microorganisms, iron acquisition via siderophores among the NTM is not well-characterized. In this study, we apply genomic and microbiological methodologies to better understand iron acquisition via siderophores for environmental and respiratory isolates of M. abscessus, M. porcinum, and M. intracellulare subspecies chimaera from Hawai’i. Siderophore synthesis and transport genes, including mycobactin (mbt), mmpL/S, and esx-3 were compared among 47 reference isolates, 29 respiratory isolates, and 23 environmental Hawai’i isolates. Among all reference isolates examined, respiratory isolates showed significantly more siderophore pertinent genes compared to environmental isolates. Among the Hawai’i isolates, RGM M. abscessus and M. porcinum had significantly less esx-3 and mbt genes compared to SGM M. chimaera when stratified by growth classification. However, no significant differences were observed between the species when grown on low iron culture agar or siderophore production by the chrome azurol S (CAS) assay in vitro. These results indicate the complex mechanisms involved in iron sequestration and siderophore activity among diverse NTM species

    An evaluation of IASI-NH\u3csub\u3e3\u3c/sub\u3e with ground-based Fourier transform infrared spectroscopy measurements

    Global distributions of atmospheric ammonia (NH3) measured with satellite instruments such as the Infrared Atmospheric Sounding Interferometer (IASI) contain valuable information on NH3 concentrations and variability in regions not yet covered by ground-based instruments. Due to their large spatial coverage and (bi-)daily overpasses, the satellite observations have the potential to increase our knowledge of the distribution of NH3 emissions and associated seasonal cycles. However the observations remain poorly validated, with only a handful of available studies often using only surface measurements without any vertical information. In this study, we present the first validation of the IASI-NH3 product using ground-based Fourier transform infrared spectroscopy (FTIR) observations. Using a recently developed consistent retrieval strategy, NH3 concentration profiles have been retrieved using observations from nine Network for the Detection of Atmospheric Composition Change (NDACC) stations around the world between 2008 and 2015. We demonstrate the importance of strict spatio-temporal collocation criteria for the comparison. Large differences in the regression results are observed for changing intervals of spatial criteria, mostly due to terrain characteristics and the short lifetime of NH3 in the atmosphere. The seasonal variations of both datasets are consistent for most sites. Correlations are found to be high at sites in areas with considerable NH3 levels, whereas correlations are lower at sites with low atmospheric NH3 levels close to the detection limit of the IASI instrument. A combination of the observations from all sites (Nobs = 547) give a mean relative difference of −32.4 ± (56.3) %, a correlation r of 0.8 with a slope of 0.73. These results give an improved estimate of the IASI-NH3 product performance compared to the previous upper-bound estimates (−50 to +100 %)
