    Ice Crystal Classification Using Two Dimensional Light Scattering Patterns

    An investigation is presented into methods of characterising cirrus ice crystals from in-situ light scattering data. A database of scattering patterns from modelled crystals was created using the Ray Tracing with Diffraction on Facets (RTDF) model from the University of Hertfordshire, to which experimental and modelled data was fitted. Experimental data was gathered in the form of scattering patterns from ice analogue crystals with similar optical properties and hexagonal symmetry to ice, yet stable at room temperature. A laboratory rig is described which images scattering patterns from single particles while allowing precise control over the orientation of the particle with respect to the incident beam. Images of scattering patterns were captured and compared to patterns from modelled crystals with similar geometry. Methods for introducing particles en-masse and individually to the Small Ice Detector (SID) instruments are discussed, with particular emphasis on the calibration of the gain of the SID-2 instrument. The variation in gain between detector elements is found to be significant, variable over the life of the detector, and different for different detectors. Fitting was performed by comparison of test scattering patterns (either modelled or experimental) to the reference database. Representation of the two dimensional scattering patterns by asymmetry factor, moment invariants, azimuthal intensity patterns (AIP) and the Fourier transform of the AIP are compared for fitting accuracy. Direct comparison of the AIP is found to be the most accurate method. Increased resolution of the AIP is shown to improve the fitting substantially. Case studies are presented for the fitting of two ice analogue crystals to the modelled database. Fitting accuracy is found to be negatively influenced by small amounts of surface roughness and detail not currently considered by the RTDF model. Fitting of in-situ data gathered by the SID-3 instrument during the HALO 02 campaign at the AIDA cloud chamber in Germany is presented and discussed. Saturation of detector pixels is shown to affect pattern fitting. In-flight operation of the instrument involves the variation of gain of the whole detector (as opposed to individual elements) in order to obtain unsaturated images of both large and small particles

    A framework for the simulation of structural software evolution

Copyright @ 2008 ACM.As functionality is added to an aging piece of software, its original design and structure will tend to erode. This can lead to high coupling, low cohesion and other undesirable effects associated with spaghetti architectures. The underlying forces that cause such degradation have been the subject of much research. However, progress in this field is slow, as its complexity makes it difficult to isolate the causal flows leading to these effects. This is further complicated by the difficulty of generating enough empirical data, in sufficient quantity, and attributing such data to specific points in the causal chain. This article describes a framework for simulating the structural evolution of software. A complete simulation model is built by incrementally adding modules to the framework, each of which contributes an individual evolutionary effect. These effects are then combined to form a multifaceted simulation that evolves a fictitious code base in a manner approximating real-world behavior. We describe the underlying principles and structures of our framework from a theoretical and user perspective; a validation of a simple set of evolutionary parameters is then provided and three empirical software studies generated from open-source software (OSS) are used to support claims and generated results. The research illustrates how simulation can be used to investigate a complex and under-researched area of the development cycle. It also shows the value of incorporating certain human traits into a simulation—factors that, in real-world system development, can significantly influence evolutionary structures

    Estuary-associated syndrome in North Carolina: an occupational prevalence study.

    Atlantic coast estuaries recently have experienced fish kills and fish with lesions attributed to Pfiesteria piscicida and related dinoflagellates. Human health effects have been reported from laboratory exposure and from a 1997 Maryland fish kill. North Carolina has recorded Pfiesteria-related fish kill events over the past decade, but human health effects from environmental exposure have not been systematically investigated or documented here. At the request of the state health agency, comprehensive examinations were conducted in a cross-sectional prevalence study of watermen working where Pfiesteria exposure may occur: waters where diseased or stressed fish were reported from June to September 1997, and where Pfiesteria had been identified in the past. Controls worked on unaffected waterways. The study was conducted 3 months after the last documented Pfiesteria-related fish kill. The goal was to document any persistent health effects from recent or remote contact with fish kills, fish with lesions, or affected waterways, using the 1997 U.S. Centers for Disease Control and Prevention case description for estuary-associated syndrome (EAS). Examinations included comprehensive medical, occupational, and environmental history, general medical, dermatologic, and neurologic examinations, vision testing, and neuropsychologic evaluations. Seventeen of 22 watermen working in affected waters and 11 of 21 in unaffected waters reported exposure to a fish kill or to fish with lesions. We found no pattern of abnormalities on medical, neurologic, neuropsychologic, or NES-2 evaluation. By history, one subject in each group met the EAS criteria, neither of whom had significant neuropsychological impairment when examined. Watermen from affected waterways had a significant reduction in visual contrast sensitivity (VCS) at the midspatial frequencies, but we did not identify a specific factor or exposure associated with this reduction. The cohorts did not differ in reported occupational exposure to solvents (qualitative) or to other neurotoxicants; however, exposure history was not sufficiently detailed to measure or control for solvent exposure. This small prevalence study in watermen, conducted 3 months after the last documented fish kill related to Pfiesteria, did not identify an increased risk of estuary-associated syndrome in those working on affected waterways. A significant difference between the estuary and ocean watermen was found on VCS, which could not be attributed to any specific factor or exposure. VCS may be affected by chemicals, drugs, alcohol, and several developmental and degenerative conditions; it has not been validated as being affected by known exposure to dinoflagellate secretions. VCS should be considered for inclusion in further studies, together with documentation or quantification of its potential confounders, to assess whether it has utility in relationship to dinoflagellate exposure

    Signal transducer and activator of transcription 3 (Stat3) regulates host defense and protects mice against herpes simplex virus-1 (HSV-1) infection

    Signal transducer and activator of transcription 3 (STAT3) mediates cellular responses to multiple cytokines, governs gene expression, and regulates the development and activation of immune cells. STAT3 also modulates reactivation of latent herpes simplex virus-1 (HSV-1) in ganglia. However, it is unclear how STAT3 regulates the innate immune response during the early phase of HSV-1 lytic infection. Many cell types critical for the innate immunity are derived from the myeloid lineage. Therefore, in this study, we used myeloid-specific Stat3 knockout mice to investigate the role of STAT3 in the innate immune response against HSV-1. Our results demonstrate that Stat3 knockout bone marrow-derived macrophages (BMMs) expressed decreased levels of interferon-α (IFN-α) and interferon-stimulated genes (ISGs) upon HSV-1 infection. In vivo, knockout mice were more susceptible to HSV-1, as marked by higher viral loads and more significant weight loss. Splenic expression of IFN-α and ISGs was reduced in the absence of STAT3, indicating that STAT3 is required for optimal type I interferon response to HSV-1. Expression of TNF-α and IL-12, cytokines that have been shown to limit HSV-1 replication and pathogenesis, was also significantly lower in knockout mice. Interestingly, Stat3 knockout mice failed to expand the CD8+ conventional DC (cDC) population upon HSV-1 infection, and this was accompanied by impaired NK and CD8 T cell activation. Collectively, our data demonstrate that myeloid-specific Stat3 deletion causes defects in multiple aspects of the immune system and that STAT3 has a protective role at the early stage of systemic HSV-1 infection

    BIANKA : A Balloon Field Campaign to Study Climate Change Uncertainties

    Climate change affects our everyday lives and despite a lot of research, many climate uncertainties exist. This work aims to help address some of these uncertainties via the BIANKA (BImodal ANdes KAroo) campaign; a first of its kind for South Africa. The campaign collected observations in the data sparse southern hemisphere and was a tri-lateral campaign between Chile, Germany and South Africa. The BIANKA campaign aimed at observing atmospheric composition, aerosols and cirrus clouds over South Africa as well as South America, with a special focus on the upper troposphere and stratosphere. This work summarises the aims and outlines the observations of the campaign. Moreover, initial results are showcased alongside plans for future work.Peer reviewe

    Cluster Analysis of Symptoms Among Patients with Upper Extremity Musculoskeletal Disorders

    Introduction Some musculoskeletal disorders of the upper extremity are not readily classified. The study objective was to determine if there were symptom patterns in self-identified repetitive strain injury (RSI) patients. Methods Members (n = 700) of the Dutch RSI Patients Association filled out a detailed symptom questionnaire. Factor analysis followed by cluster analysis grouped correlated symptoms. Results Eight clusters, based largely on symptom severity and quality were formulated. All but one cluster showed diffuse symptoms; the exception was characterized by bilateral symptoms of stiffness and aching pain in the shoulder/neck. Conclusions Case definitions which localize upper extremity musculoskeletal disorders to a specific anatomical area may be incomplete. Future clustering studies should rely on both signs and symptoms. Data could be collected from health care providers prospectively to determine the possible prognostic value of the identified clusters with respect to natural history, chronicity, and return to work
