    Observation of sub-Poisson photon statistics in the cavity-QED microlaser

    We have measured the second-order correlation function of the cavity-QED microlaser output and observed a transition from photon bunching to antibunching with increasing average number of intracavity atoms. The observed correlation times and the transition from super- to sub-Poisson photon statistics can be well described by gain-loss feedback or enhanced/reduced restoring action against fluctuations in photon number in the context of a quantum microlaser theory and a photon rate equation picture. However, the theory predicts a degree of antibunching several times larger than that observed, which may indicate the inadequacy of its treatment of atomic velocity distributions.Comment: 4 pages, 4 figure

    Atomic position localization via dual measurement

    We study localization of atomic position when a three-level atom interacts with a quantized standing-wave field in the Ramsey interferometer setup. Both the field quadrature amplitude and the atomic internal state are measured to obtain the atomic position information. It is found that this dual measurement scheme produces an interference pattern superimposed on a diffraction-like pattern in the atomic position distribution, where the former pattern originates from the state-selective measurement and the latter from the field measurement. The present scheme results in a better resolution in the position localization than the field-alone measurement schemes. We also discuss the measurement-correlated mechanical action of the standing-wave field on the atom in the light of Popper's test.Comment: 6.5 pages and 5 figure

    A scoping review of ontologies related to human behaviour change.

    Ontologies are classification systems specifying entities, definitions and inter-relationships for a given domain, with the potential to advance knowledge about human behaviour change. A scoping review was conducted to: (1) identify what ontologies exist related to human behaviour change, (2) describe the methods used to develop these ontologies and (3) assess the quality of identified ontologies. Using a systematic search, 2,303 papers were identified. Fifteen ontologies met the eligibility criteria for inclusion, developed in areas such as cognition, mental disease and emotions. Methods used for developing the ontologies were expert consultation, data-driven techniques and reuse of terms from existing taxonomies, terminologies and ontologies. Best practices used in ontology development and maintenance were documented. The review did not identify any ontologies representing the breadth and detail of human behaviour change. This suggests that advancing behavioural science would benefit from the development of a behaviour change intervention ontology.Wellcome Trus

    Exploring Profitable, Sustainable Livestock Businesses in an Increasingly Variable Climate

    Livestock production systems will need to change in many regions of the world to meet the dual challenges of adaptation to a changing climate and reducing net greenhouse gas (GHG) emissions. The ‘Nexus project’ is exploring these options using seven case studies in eastern Australia spanning a range of production systems from semi-arid beef production in northern Australia to mixed beef and sheep production in cool temperate climates of southern Australia. Regional reference groups, made up of 4-6 local farmers and advisers, identified a series of farm systems changes that could be made to meet these challenges. Options were categorised under four themes: feedbase, animal genetics and management, management and technology, and transformational options. More options for adaptation were identified compared to options to mitigate greenhouse gas emission of sequestered carbon. Adaptation options focussed on maximising pasture production in variable climates, and having efficient and flexible animal production systems. A focus on infrastructure for water and feed storage and use was also observed. Options to reduce GHG emissions focussed on reducing feed demand and sequestering carbon in soils and trees, and limited possibilities for feed additives were noted in these pasture-based systems. Further work will explore how combinations of these options will impact production, profit and greenhouse gas emissions from these production systems in current and future climate scenarios

    Paroxetine suppresses recombinant human P2X7 responses

    P2X7 receptor (P2X7) activity may link inflammation to depressive disorders. Genetic variants of human P2X7 have been linked with major depression and bipolar disorders, and the P2X7 knockout mouse has been shown to exhibit anti-depressive-like behaviour. P2X7 is an ATP-gated ion channel and is a major regulator of the pro-inflammatory cytokine interleukin 1ÎČ (IL-1ÎČ) secretion from monocytes and microglia. We hypothesised that antidepressants may elicit their mood enhancing effects in part via modulating P2X7 activity and reducing inflammatory responses. In this study, we determined whether common psychoactive drugs could affect recombinant and native human P2X7 responses in vitro. Common antidepressants demonstrated opposing effects on human P2X7-mediated responses; paroxetine inhibited while fluoxetine and clomipramine mildly potentiated ATP-induced dye uptake in HEK-293 cells stably expressing recombinant human P2X7. Paroxetine inhibited dye uptake mediated by human P2X7 in a concentration-dependent manner with an IC50 of 24 ÎŒM and significantly reduces ATP-induced inward currents. We confirmed that trifluoperazine hydrochloride suppressed human P2X7 responses (IC50 of 6.4 ÎŒM). Both paroxetine and trifluoperazine did not inhibit rodent P2X7 responses, and mutation of a known residue (F 95L) did not alter the effect of either drug, suggesting neither drug binds at this site. Finally, we demonstrate that P2X7-induced IL-1ÎČ secretion from lipopolysaccharide (LPS)-primed human CD14+ monocytes was suppressed with trifluoperazine and paroxetine

We illustrate our discussion with descriptions of experimental demonstrations of key communication and processing tasks from the last decade and also look forward to the key results likely in the next decade. We examine both discrete (single photon) type processing as well as those which employ continuous variable manipulations. The mathematical formalism is kept to the minimum needed to understand the key theoretical and experimental results

    Pushed and pulled fronts in a discrete reaction-diffusion equation

    We consider the propagation of wave fronts connecting unstable and stable uniform solutions to a discrete reaction-diffusion equation on a one-dimensional integer lattice. The dependence of the wavespeed on the coupling strength ” between lattice points and on a detuning parameter (α) appearing in a nonlinear forcing is investigated thoroughly. Via asymptotic and numerical studies, the speed both of 'pulled' fronts (whereby the wavespeed can be characterised by the linear behaviour at the leading edge of the wave) and of 'pushed' fronts (for which the nonlinear dynamics of the entire front determine the wavespeed) is investigated in detail.     Probenecid Blocks Human P2X7 Receptor-Induced Dye Uptake via a Pannexin-1 Independent Mechanism

    P2X7 is a ligand-gated ion channel which is activated by ATP and displays secondary permeability characteristics. The mechanism of development of the secondary permeability pathway is currently unclear, although a role for the hemichannel protein pannexin-1 has been suggested. In this study we investigated the role of pannexin-1 in P2X7-induced dye uptake and ATP-induced IL-1ÎČ secretion from human monocytes. We found no pharmacological evidence for involvement of pannexin-1 in P2X7-mediated dye uptake in transfected HEK-293 cells with no inhibition seen for carbenoxolone and the pannexin-1 mimetic inhibitory peptide, 10Panx1. However, we found that probenecid inhibited P2X7-induced cationic and anionic dye uptake in stably transfected human P2X7 HEK-293 cells. An IC50 value of 203 ÎŒM was calculated for blockade of ATP-induced responses at human P2X7. Probenecid also reduced dye uptake and IL-1ÎČ secretion from human CD14+ monocytes whereas carbenoxolone and 10Panx1 showed no inhibitory effect. Patch clamp and calcium indicator experiments revealed that probenecid directly blocks the human P2X7 receptor

    Identification and correction of previously unreported spatial phenomena using raw Illumina BeadArray data

    <p>Abstract</p> <p>Background</p> <p>A key stage for all microarray analyses is the extraction of feature-intensities from an image. If this step goes wrong, then subsequent preprocessing and processing stages will stand little chance of rectifying the matter. Illumina employ random construction of their BeadArrays, making feature-intensity extraction even more important for the Illumina platform than for other technologies. In this paper we show that using raw Illumina data it is possible to identify, control, and perhaps correct for a range of spatial-related phenomena that affect feature-intensity extraction.</p> <p>Results</p> <p>We note that feature intensities can be unnaturally high when in the proximity of a number of phenomena relating either to the images themselves or to the layout of the beads on an array. Additionally we note that beads neighbour beads of the same type more often than one might expect, which may cause concern in some models of hybridization. We highlight issues in the identification of a bead's location, and in particular how this both affects and is affected by its intensity. Finally we show that beads can be wrongly identified in the image on either a local or array-wide scale, with obvious implications for data quality.</p> <p>Conclusions</p> <p>The image processing issues identified will often pass unnoticed by an analysis of the standard data returned from an experiment. We detail some simple diagnostics that can be implemented to identify problems of this nature, and outline approaches to correcting for such problems. These approaches require access to the raw data from the arrays, not just the summarized data usually returned, making the acquisition of such raw data highly desirable.</p
