    Impact of Waddlia chondrophila infection on pregnancy in the mouse.

    The intracellular bacterium Waddlia chondrophila, which belongs to the Chlamydiales order, was found to be associated with miscarriage in humans. There is little to no knowledge regarding the mode of infection, impact on the neonate and pathophysiology of this emerging bacterium. We have previously shown that W. chondrophila induces a systemic infection, organ pathology and elicits T helper type 1-associated humoral immunity in a murine model of genital infection. In the present study, we took advantage of this model of infection to evaluate the impact of this bacterium on the mouse pregnancy. We used two routes of inoculation, vaginal and intrauterine, to introduce infection before and after mating. Our results show that genital infection by W. chondrophila did not have any significant impact on gestation length and maternal weight gain, nor on the number of offspring and their weight. This observation indicates that the mouse model of infection is not suitable to study the effect of W. chondrophila on pregnancy and alternative models of infection, including in vitro ones, should be used. Moreover, an indirect immunopathological mechanism activated by this bacterium should be further explored

    Waddlia chondrophila is a known bovine abortigenic Chlamydia-related bacterium that has been associated with adverse pregnancy outcomes in human. However, there is a lack of knowledge regarding how W. chondrophila infection spreads, its ability to elicit an immune response and induce pathology. A murine model of genital infection was developed to investigate the pathogenicity and immune response associated with a W. chondrophila infection. Genital inoculation of the bacterial agent resulted in a dose-dependent infection that spread to lumbar lymph nodes and successively to spleen and liver. Bacterial-induced pathology peaked on day 14, characterized by leukocyte infiltration (uterine horn, liver, and spleen), necrosis (liver) and extramedullary hematopoiesis (spleen). Immunohistochemistry demonstrated the presence of a large number of W. chondrophila in the spleen on day 14. Robust IgG titers were detected by day 14 and remained high until day 52. IgG isotypes consisted of high IgG2a, moderate IgG3 and no detectable IgG1, indicating a Th1-associated immune response. This study provides the first evidence that W. chondrophila genital infection is capable of inducing a systemic infection that spreads to major organs, induces uterus, spleen, and liver pathology and elicits a Th1-skewed humoral response. This new animal model will help our understanding of the mechanisms related to intracellular bacteria-induced miscarriages, the most frequent complication of pregnancy that affects one in four women

    Short-term liquid storage of ram semen in various extenders

    The aim of this study was to investigate the effects of three extenders on ram sperm quality after short-term liquid storage (24 hours’ holding time). The study included 20 crossbred rams (Pirot Pramenka x Wurttemberg x Ile de France), 12 months old. Animals were housed at the experimental sheep farm of the Institute for Animal Husbandry in Belgrade, Serbia. Semen was collected through electro ejaculation. The ejaculates were obtained from single services and routine field analysis of the semen was performed immediately after the collection. The semen was split and diluted with three extenders, namely Optidyl®, Andromed® and ultrahigh temperature processed (UHT) milk, in ratios of 1 : 50 or 1 : 100. The ejaculates were examined for sperm motility variables (sperm cell motility percentage, the progressive motility percentage, curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP), sperm linearity (LIN), straightness (STR), amplitude of lateral sperm head displacement (ALH), beat cross frequency (BCF) and circular tracks), and sperm morphology (live sperm percentage, percentage of normal sperm forms with intact acrosome, percentage of abnormal sperm forms and total damaged acrosome) by computer-assisted sperm analysis (CASA) and classic sperm cytology after supravital eosin/nigrosine/trypan blue staining, respectively. It was observed that the type of extender used in diluting ram semen is an important factor in the successful short-term liquid preservation (at 4 °C) of ram spermatozoa. In conclusion, this study showed that egg yolk (Optidyl) and soybean (Andromed)-based extenders gave better results of both sperm morphology and sperm motility parameters compared with UHT milk.Keywords: Diluents, morphology, motility, sper

We herein report a case of a ruptured aneurysm at a basilar artery fenestration. The diagnosis was assessed with 16-row multisection CT angiography and was confirmed by using digital subtraction angiography. The patient was successfully treated with coil placement

    Nano-Motion Analysis for Rapid and Label Free Assessing of Cancer Cell Sensitivity to Chemotherapeutics.

    Background and Objectives: Optimization of chemotherapy is crucial for cancer patients. Timely and costly efficient treatments are emerging due to the increasing incidence of cancer worldwide. Here, we present a methodology of nano-motion analysis that could be developed to serve as a screening tool able to determine the best chemotherapy option for a particular patient within hours. Materials and Methods: Three different human cancer cell lines and their multidrug resistant (MDR) counterparts were analyzed with an atomic force microscope (AFM) using tipless cantilevers to adhere the cells and monitor their nano-motions. Results: The cells exposed to doxorubicin (DOX) differentially responded due to their sensitivity to this chemotherapeutic. The death of sensitive cells corresponding to the drop in signal variance occurred in less than 2 h after DOX application, while MDR cells continued to move, even showing an increase in signal variance. Conclusions: Nano-motion sensing can be developed as a screening tool that will allow simple, inexpensive and quick testing of different chemotherapeutics for each cancer patient. Further investigations on patient-derived tumor cells should confirm the method's applicability

    Impact of the COVID-19 pandemic on Swiss pregnant and breastfeeding women - a cross-sectional study covering the first pandemic wave.

    Information on the impact of the COVID-19 pandemic on pregnancy and breastfeeding experiences, as well as on perinatal mental health in Switzerland is limited. In Switzerland, there are few national studies and little information. Using an anonymous online survey accessible after the first wave of the outbreak in Switzerland, we have investigated how this pandemic affected pregnant and breastfeeding women. Among women who completed the survey, 69.0% (1050/1518) indicated the first wave of the pandemic affected their personal habits, 61.0% (689/1131) were affected in their work and 40.0% (632/1573) reported impaired relations with healthcare services (different denominators correspond to the number of participants who answered the question). 36.8% (110/299) of women reported an impact of the pandemic on their current pregnancy experience or breastfeeding experience (8.2%, 46/555). Overall, 11.6% (170/1467) of participants who completed the validated screening tests for mental health symptoms (Edinburgh Postnatal Depression Scale, Generalized Anxiety Disorder 7, Perceived Stress Scale) presented a score compatible with symptoms of major depression, severe anxiety or high perceived stress, which is higher than in the pre-pandemic period according to literature. Risk factors independently associated with impaired mental health were being hospitalized, having symptoms of COVID-19, living with a person with COVID-19 symptoms, having comorbidities, having experienced reduced healthcare services, having restricted usual activities and being a housewife. Protective factors independently associated were a high level of education and living with a partner. Our findings suggest that the COVID-19 pandemic might have significantly affected the well-being and mental health of pregnant and breastfeeding women, directly in the case of exposure, and indirectly as a result of the potential modifications in their life habits and in healthcare facilities

    Relativistic three-body bound states and the reduction from four to three dimensions

    Beginning with an effective field theory based upon meson exchange, the Bethe-Salpeter equation for the three-particle propagator (six-point function) is obtained. Using the one-boson-exchange form of the kernel, this equation is then analyzed using time-ordered perturbation theory, and a three-dimensional equation for the propagator is developed. The propagator consists of a pre-factor in which the relative energies are fixed by the initial state of the particles, an intermediate part in which only global propagation of the particles occurs, and a post-factor in which relative energies are fixed by the final state of the particles. The pre- and post-factors are necessary in order to account for the transition from states where particles are off their mass shell to states described by the global propagator with all of the particle energies on shell. The pole structure of the intermediate part of the propagator is used to determine the equation for the three-body bound state: a Schr{\"o}dinger-like relativistic equation with a single, global Green's function. The role of the pre- and post-factors in the relativistic dynamics is to incorporate the poles of the breakup channels in the initial and final states. The derivation of this equation by integrating over the relative times rather than via a constraint on relative momenta allows the inclusion of retardation and dynamical boost corrections without introducing unphysical singularities.Comment: REVTeX, 21 pages, 4 figures, epsf.st
