    Baited technique improves censuses of cryptic fish in complex habitats

    Many fish species are cryptic by nature or show a negative response to divers. This may make traditional censusing techniques difficult to perform and the results obtained using these methods may be inaccurate. This problem will be exacerbated in complex habitats, such as those found on coral reefs, because the cryptic species present will be even more difficult to locate and identify. Predation studies on coral reefs have been especially hampered by this problem because many predatory species are cryptic and so it has been difficult to obtain reliable abundance estimates. This study tested a visual census technique that used bait to bring cryptic predatory fish into view. Results from this census were then compared to those from a traditional survey using belt transects, and to a patch reef tagging study where all individuals of 3 key species were located. The baited technique produced significantly higher density estimates for 3 of the 4 most abundant cryptic species. The patch reef experiment demonstrated that the baited technique accounted for 85 to 96% of fish present. Censuses without bait observed only 40 to 61% of fish present. For mobile species, on the other hand, the baited census appeared to overestimate abundance, due to movement of fish into the census area. We therefore recommend combining baited censuses of sedentary cryptic species with traditional censuses of mobile species to gain an accurate picture of piscivorous reef fish communities. Using this approach at Lizard Island on the Great Barrier Reef, the proportion of cryptic piscivores in the community was found to be almost double that which would have otherwise been observed. We also found considerable spatial variation in the abundance and distribution of piscivorous fish. These patterns would have been quite different had the survey been based on belt transects alone. Previous studies using belt transects may therefore have underestimated the importance of cryptic piscivorous species in communities of coral reef fish

    Cumulative risk of compromised physical, mental and social health in adulthood due to family conflict and financial strain during childhood: A retrospective analysis based on survey data representative of 19 European countries

    BACKGROUND: Childhood adversity (CA) has previously been linked to various health problems in adulthood. Investigations into the differential impact of distinct types of CA on a wide range of outcomes are scarce. This study aimed to assess the impact of self-reported childhood family conflict and/or financial strain on health and social functioning in adulthood among Europeans, while taking into account the mediating role of adulthood socioeconomic disadvantage (SED) in these associations. METHODS: Using the European Social Survey (ESS) collected in 2014, nationally representative cross-sectional data from 35 475 participants aged 15 years and older in 19 European countries were analysed. Logistic regressions were conducted to assess associations of retrospectively reported family conflict and/or financial strain in childhood with physical and mental health as well as health behaviours and social functioning in adulthood. RESULTS: A quarter of the European population reported having experienced family conflict, financial strain or both in childhood. Financial strain was reported more among older age groups and conflict more among younger age groups. A dose-response pattern with increased risk was demonstrated for almost all physical, behavioral, mental and social outcomes for these aspects of CA compared with no CA, with the highest risk observed in those who experienced both financial strain and family conflict. Adulthood SED mediated a significant proportion of the associations with financial strain (ranging from 5.4% to 72.4%), but did not mediate the associations with conflict. CONCLUSIONS: Individuals reporting family conflict or financial strain during childhood are at increased risk of developing a wide range of health and social problems. Those who report financial strain in childhood are more likely to experience SED in adulthood, which in turn increases their risk of experiencing health and social problems. Reported family conflict during childhood conferred increased risk of health and social problems, but adulthood SED did not appear to operate as an indirect pathway

    Tetracycline-inducible gene regulation in mycobacteria

    A system for the tetracycline-inducible regulation of gene expression in mycobacteria has been developed. We have sub-cloned the tetRO region from the Corynebacterium glutamicum TetZ locus into a mycobacterial shuttle plasmid, making expression of genes cloned downstream of tetRO responsive to tetracycline. Using the luxAB-encoded luciferase from Vibrio harveyi as a reporter (pMind-Lx), we observed a 40-fold increase in light output from Mycobacterium smegmatis cultures 2 h after adding 20 ng ml(−1) of tetracycline. Similarly, exposure to the drug resulted in up to 20-fold increase in relative light units from M.bovis BCG carrying the reporter construct, and a 10-fold increase for M.tuberculosis. Tetracycline induction was demonstrated in log and stationary phase cultures. To evaluate whether this system is amenable to use in vivo, J774 macrophages were infected with M.bovis BCG[pMind-Lx], treated with amikacin to kill extracellular bacteria, and then incubated with tetracycline. A 10-fold increase in light output was measured after 24 h, indicating that intracellular bacteria are accessible and responsive to exogenously added tetracycline. To test the use of the tetracycline-inducible system for conditional gene silencing, mycobacteria were transformed with a pMind construct with tetRO driving expression of antisense RNA for the ftsZ gene. Bacterial cells containing the antisense construct formed filaments after 24 h exposure to tetracycline. These results demonstrate the potential of this tetracycline-regulated system for the manipulation of mycobacterial gene expression inside and outside cells

    Interstitial lung fluid balance in healthy lowlanders exposed to high-altitude

    We aimed to assess lung fluid balance before and after gradual ascent to 5,150 m. Lung diffusion capacity for carbon monoxide (DLCO), alveolar-capillary membrane conductance (DmCO) and ultrasound lung comets (ULCs) were assessed in 12 healthy lowlanders at sea-level, and on Day 1, Day 5 and Day 9 after arrival at Mount Everest Base Camp (EBC). EBC was reached following an 8-day hike at progressively increasing altitudes starting at 2,860 m. DLCO was unchanged from sea-level to Day 1 at EBC, but increased on Day 5 (11 ± 10%) and Day 9 (10 ± 9%) vs. sea-level (P ≤ 0.047). DmCO increased from sea-level to Day 1 (9 ± 6%), Day 5 (12 ± 8%), and Day 9 (17 ± 11%) (all P ≤ 0.001) at EBC. There was no change in ULCs from sea-level to Day 1, Day 5 and Day 9 at EBC. These data provide evidence that interstitial lung fluid remains stable or may even decrease relative to at sea-level following 8 days of gradual exposure to high-altitude in healthy humans

    An effective multiscale methodology for the analysis of marine flexible risers

    Life extension is an attractive option for subsea flexible risers nearing the end of their design lives. However, techniques for assessing accumulated fatigue damage in flexible risers are often associated with large uncertainties due to the simplified calculation approaches typically used. One approach to reducing uncertainties is the inclusion of nonlinearities in riser structural response and consistent linking between global and local models. In this article, we present the elements of a numerical multiscale procedure capable of predicting the stresses that lead to fatigue damage in flexible pipes, namely: a nonlinear beam element, a nonlinear section response model and a detailed finite element model; the consistent integration of models developed for different length scales; and finally a validation of the flexible riser large-scale model

    Whole-genome association analysis of treatment response in obsessive-compulsive disorder.

    Up to 30% of patients with obsessive-compulsive disorder (OCD) exhibit an inadequate response to serotonin reuptake inhibitors (SRIs). To date, genetic predictors of OCD treatment response have not been systematically investigated using genome-wide association study (GWAS). To identify specific genetic variations potentially influencing SRI response, we conducted a GWAS study in 804 OCD patients with information on SRI response. SRI response was classified as 'response' (n=514) or 'non-response' (n=290), based on self-report. We used the more powerful Quasi-Likelihood Score Test (the MQLS test) to conduct a genome-wide association test correcting for relatedness, and then used an adjusted logistic model to evaluate the effect size of the variants in probands. The top single-nucleotide polymorphism (SNP) was rs17162912 (P=1.76 × 10(-8)), which is near the DISP1 gene on 1q41-q42, a microdeletion region implicated in neurological development. The other six SNPs showing suggestive evidence of association (P<10(-5)) were rs9303380, rs12437601, rs16988159, rs7676822, rs1911877 and rs723815. Among them, two SNPs in strong linkage disequilibrium, rs7676822 and rs1911877, located near the PCDH10 gene, gave P-values of 2.86 × 10(-6) and 8.41 × 10(-6), respectively. The other 35 variations with signals of potential significance (P<10(-4)) involve multiple genes expressed in the brain, including GRIN2B, PCDH10 and GPC6. Our enrichment analysis indicated suggestive roles of genes in the glutamatergic neurotransmission system (false discovery rate (FDR)=0.0097) and the serotonergic system (FDR=0.0213). Although the results presented may provide new insights into genetic mechanisms underlying treatment response in OCD, studies with larger sample sizes and detailed information on drug dosage and treatment duration are needed

    Internal construct validity of the Warwick-Edinburgh Mental Well-being Scale (WEMWBS): a Rasch analysis using data from the Scottish Health Education Population Survey

    Background: The Warwick-Edinburgh Mental Well-Being Scale (WEMWBS) was developed to meet demand for instruments to measure mental well-being. It comprises 14 positively phrased Likert-style items and fulfils classic criteria for scale development. We report here the internal construct validity of WEMWBS from the perspective of the Rasch measurement model. Methods: The model was applied to data collected from 779 respondents in Wave 12 (Autumn 2006) of the Scottish Health Education Population Survey. Respondents were aged 16–74 (average 41.9) yrs. Results: Initial fit to model expectations was poor. The items 'I've been feeling good about myself', 'I've been interested in new things' and 'I've been feeling cheerful' all showed significant misfit to model expectations, and were deleted. This led to a marginal improvement in fit to the model. After further analysis, more items were deleted and a strict unidimensional seven item scale (the Short Warwick Edinburgh Mental Well-Being Scale (SWEMWBS)) was resolved. Many items deleted because of misfit with model expectations showed considerable bias for gender. Two retained items also demonstrated bias for gender but, at the scale level, cancelled out. One further retained item 'I've been feeling optimistic about the future' showed bias for age. The correlation between the 14 item and 7 item versions was 0.954. Given fit to the Rasch model, and strict unidimensionality, SWEMWBS provides an interval scale estimate of mental well-being. Conclusion: A short 7 item version of WEMWBS was found to satisfy the strict unidimensionality expectations of the Rasch model, and be largely free of bias. This scale, SWEMWBS, provides a raw score-interval scale transformation for use in parametric procedures. In terms of face validity, SWEMWBS presents a more restricted view of mental well-being than the 14 item WEMWBS, with most items representing aspects of psychological and eudemonic well-being, and few covering hedonic well-being or affect. However, robust measurement properties combined with brevity make SWEMWBS preferable to WEMWBS at present for monitoring mental well-being in populations. Where face validity is an issue there remain arguments for continuing to collect data on the full 14 item WEMWBS
