    Qualitative process evaluation of an Australian alcohol media literacy study: recommendations for designing culturally responsive school-based programs

    Background: Alcohol media literacy programs seek to mitigate the potentially harmful effects of alcohol advertising on children\u27s drinking intentions and behaviours through equipping them with skills to challenge media messages. In order for such programs to be effective, the teaching and learning experiences must be tailored to their specific cultural context. Media in the Spotlight is an alcohol media literacy program aimed at 9 to 12 year old Australian children. This study evaluates the process and implementation of the program, outlining the factors that facilitated and inhibited implementation. From this evaluation, a pedagogical framework has been developed for health professionals implementing culturally responsive programs in school settings. Methods: Process measures included: semi-structured interviews with teachers before and after the program was implemented (n = 11 interviews), program evaluation questionnaires completed by children (n = 166), lesson observations completed by teachers (n = 35 observations), and reflective journal entries completed by the researcher (n = 44 entries). A thematic analysis approach was used to analyse all of the data sets using NVivo. Inductive coding was used, whereby the findings were derived from the research objectives and multiple readings and interpretations of the data. Results: Five key pedagogical considerations were identified that facilitated implementation. These were: connecting to the students\u27 life worlds to achieve cultural significance; empowering students with real-world skills to ensure relevance; ensuring programs are well structured with strong connections to the school curriculum; creating developmentally appropriate activities while providing a range of assessment opportunities; and including hands-on and interactive activities to promote student engagement. Three potential inhibitors to implementing the alcohol media literacy program in upper-elementary school classrooms were identified. These included topic sensitivities, classroom management challenges, and fitting new programs into already busy school schedules. Conclusion: Overall, the program content and individual lessons were well received by the teachers and students. The lessons learned from the development, implementation and evaluation of this program can provide health professionals with key pedagogical strategies for designing culturally responsive educational programs.     Ternatin and improved synthetic variants kill cancer cells by targeting the elongation factor-1A ternary complex.

    Cyclic peptide natural products have evolved to exploit diverse protein targets, many of which control essential cellular processes. Inspired by a series of cyclic peptides with partially elucidated structures, we designed synthetic variants of ternatin, a cytotoxic and anti-adipogenic natural product whose molecular mode of action was unknown. The new ternatin variants are cytotoxic toward cancer cells, with up to 500-fold greater potency than ternatin itself. Using a ternatin photo-affinity probe, we identify the translation elongation factor-1A ternary complex (eEF1A·GTP·aminoacyl-tRNA) as a specific target and demonstrate competitive binding by the unrelated natural products, didemnin and cytotrienin. Mutations in domain III of eEF1A prevent ternatin binding and confer resistance to its cytotoxic effects, implicating the adjacent hydrophobic surface as a functional hot spot for eEF1A modulation. We conclude that the eukaryotic elongation factor-1A and its ternary complex with GTP and aminoacyl-tRNA are common targets for the evolution of cytotoxic natural products

    Mapping quantitative trait loci for seizure response to a GABAA receptor inverse agonist in mice

    To define the genetic contributions affecting individual differences in seizure threshold, a beta carboline [methyl-beta-carboline-3-carboxylate (beta-CCM)]-induced model of generalized seizures was genetically dissected in mice. beta-CCM is a GABAA receptor inverse agonist and convulsant. By measuring the latency to generalized seizures after beta-CCM administration to A/J and C57BL6/J mice and their progeny, we estimated a heritability of 0.28 +/- 0.10. A genome wide screen in an F2 population of these parental strains (n = 273) mapped quantitative trait loci (QTLs) on proximal chromosome 7 [logarithm of the likelihood for linkage (LOD) = 3.71] and distal chromosome 10 (LOD = 4.29) for seizure susceptibility, explaining approximately 22 and 25%, respectively, of the genetic variance for this seizure trait. The best fitting logistic regression model suggests that the A/J allele at each locus increases the likelihood of seizures approximately threefold. In a subsequent backcross population (n = 223), we mapped QTLs on distal chromosome 4 (LOD = 2.88) and confirmed the distal chromosome 10 QTLs (LOD = 4.36). In the backcross, the C57BL/6J allele of the chromosome 10 QTL decreases the risk of seizures approximately twofold. These QTLs may ultimately lead to the identification of genes influencing individual differences in seizure threshold in mice and the discovery of novel anticonvulsant agents. The colocalization on distal chromosome 10 of a beta-CCM susceptibility QTL and a QTL for open field ambulation and vertical movement suggests the existence of a single, pleiotropic locus, which we have named Exq1

    Hydrographic Properties and Inferred Circulation Over the Northeastern Shelves of the Gulf of Mexico During Spring to Midsummer of 1998

    A hydrographic cruise was conducted 5-16 May 1998 over the northeastern shelves of the Gulf of Mexico, Observed distributions of temperature, salinity, oxygen, and nutrients were consonant with prior occurrences of upwelling, particularly near the head of DeSoto Canyon. Shipboard, moored, and satellite observations indicated these upwelling events were related to the presence of an anticyclonic circulation feature over the canyon. In addition, several cool water events occurred during spring in the nearshore region west of Pensacola; these may be attributed to atmospheric effects. High river discharges from rivers west of the Apalachicola during winter and spring likely resulted in the extensive surface distributions of low-salinity water observed from Mississippi Sound to Cape San Bias during the cruise. The combination of cool bottom temperatures and relatively low surface salinities over the inshore shelf west of Cape San Bias, with the usual seasonal warming, resulted in enhanced vertical stability. This stability likely inhibited vertical mixing and contributed to the development of the relatively low concentrations of dissolved oxygen observed in the bottom waters

This article may be used for non-commercial purposes in accordance with Wiley Terms and Conditions for Self-Archiving."We tested how the addition of oxaloacetate (OAA) to SH-SY5Y cells affected bioenergetic fluxes and infrastructure, and compared the effects of OAA to malate, pyruvate, and glucose deprivation. OAA displayed pro-glycolysis and pro-respiration effects. OAA pro-glycolysis effects were not a consequence of decarboxylation to pyruvate because unlike OAA, pyruvate lowered the glycolysis flux. Malate did not alter glycolysis flux and reduced mitochondrial respiration. Glucose deprivation essentially eliminated glycolysis and increased mitochondrial respiration. OAA increased, while malate decreased, the cell NAD+/NADH ratio. Cytosolic malate dehydrogenase 1 (MDH1) protein increased with OAA treatment, but not with malate or glucose deprivation. Glucose deprivation increased protein levels of ATP citrate lyase, an enzyme which produces cytosolic OAA, while OAA altered neither ATP citrate lyase mRNA nor protein levels. OAA, but not glucose deprivation, increased COX2, PGC1α, PGC1β, and PRC protein levels. OAA increased total and phosphorylated SIRT1 protein. We conclude that adding OAA to SH-SY5Y cells can support or enhance both glycolysis and respiration fluxes. These effects appear to depend, at least partly, on OAA causing a shift in the cell redox balance to a more oxidized state, that it is not a glycolysis pathway intermediate, and possibly its ability to act in an anaplerotic fashion

    Development, Operation, and Results From the Texas Automated Buoy System

    The Texas Automated Buoy System (TABS) is a coastal network of moored buoys that report near-real-time observations about currents and winds along the Texas coast. Established in 1995, the primary mission of TABS is ocean observations in the service of oil spill preparedness and response. The state of Texas funded the system with the intent of improving the data available to oil spill trajectory modelers. In its 12 years of operation, TABS has proven its usefulness during realistic oil spill drills and actual spills. The original capabilities of TABS, i.e., measurement of surface currents and temperatures, have been extended to the marine surface layer, the entire water column, and the sea floor. In addition to observations, a modeling component has been integrated into the TABS program. The goal is to form the core of a complete ocean observing system for Texas waters. As the nation embarks on the development of an integrated ocean observing system, TABS will continue to be an active participant of the Gulf of Mexico Coastal Ocean Observing System (GCOOS) regional association and the primary source of near-surface current measurements in the northwestern Gulf of Mexico. This article describes the origin of TABS, the philosophy behind the operation and development of the system, the resulting modifications to improve the system, the expansion of the system to include new sensors, the development of TABS forecasting models and real-time analysis tools, and how TABS has met many of the societal goals envisioned for GCOOS

    The Availability of Advanced Airway Equipment and Experience with Videolaryngoscopy in the UK: Two UK Surveys

    Fibreoptic intubation, high frequency jet ventilation, and videolaryngoscopy form part of the Royal College of Anaesthetists compulsory higher airway training module. Curriculum delivery requires equipment availability and competent trainers. We sought to establish (1) availability of advanced airway equipment in UK hospitals (Survey I) and (2) if those interested in airway management (Difficult Airway Society (DAS) members) had access to videolaryngoscopes, their basic skill levels and teaching competence with these devices and if they believed that videolaryngoscopy was replacing conventional or fibreoptic laryngoscopy (Survey II). Data was obtained from 212 hospitals (73.1%) and 554 DAS members (27.6%). Most hospitals (202, 99%) owned a fiberscope, 119 (57.5%) had a videolaryngoscope, yet only 62 (29.5%) had high frequency jet ventilators. DAS members had variable access to videolaryngoscopes with Airtraq 319 (59.6%) and Glidescope 176 (32.9%) being the most common. More DAS members were happy to teach or use videolaryngoscopes in a difficult airway than those who had used them more than ten times. The majority rated Macintosh laryngoscopy as the most important airway skill. Members rated fibreoptic intubation and videolaryngoscopy skills equally. Our surveys demonstrate widespread availability of fibreoptic scopes, limited availability of videolaryngoscopes, and limited numbers of experienced videolaryngoscope tutors
