    Brief communication: The effects of disuse on the mechanical properties of bone: What unloading tells us about the adaptive nature of skeletal tissue

    The intricate link between load environment and skeletal health is exemplified by the severe osteopenia that accompanies prolonged periods of immobilization, frequently referred to as disuse osteoporosis. Investigating the effects disuse has on the structural properties of bone provides a unique opportunity to better understand how mechanical loads influence the adaptation and maintenance of skeletal tissue. Here, we report results from an examination of multiple indicators of bone metabolism (e.g., mean osteon density, mean osteon size, bone mass, and bone area distribution) within the major long bones of individuals with distinct activity level differences. Results are based on a sample comprising two subjects that suffered from long‐term quadriplegia and 28 individuals of comparable age that had full limb mobility. Although limited in sample size, our findings suggest bones associated with long‐term disuse have lower osteon densities and larger osteon areas compared to individuals of normal mobility, reflecting dramatically lower remodeling rates potentially related to reduced strain levels. Moreover, immobilized skeletal elements demonstrate a reduced percentage of cortical area present resulting from endosteal resorption. Differences between mobility groups in the percentage of cortical area present and bone distribution of all skeletal elements, suggests bone modeling activity is negligible in the unloaded adult skeleton. Additional histomorphometric comparisons reveal potential intraskeletal differences in bone turnover rates suggesting remodeling rates are highest within the humeri and femora. Addition of more immobilized individuals in the future will allow for quantitative statistical analyses and greater consideration of human variation within and between individuals.     On the psychological origins of tool use

    The ubiquity of tool use in human life has generated multiple lines of scientific and philosophical investigation to understand the development and expression of humans' engagement with tools and its relation to other dimensions of human experience. However, existing literature on tool use faces several epistemological challenges in which the same set of questions generate many different answers. At least four critical questions can be identified, which are intimately intertwined-(1) What constitutes tool use? (2) What psychological processes underlie tool use in humans and nonhuman animals? (3) Which of these psychological processes are exclusive to tool use? (4) Which psychological processes involved in tool use are exclusive to Homo sapiens? To help advance a multidisciplinary scientific understanding of tool use, six author groups representing different academic disciplines (e.g., anthropology, psychology, neuroscience) and different theoretical perspectives respond to each of these questions, and then point to the direction of future work on tool use. We find that while there are marked differences among the responses of the respective author groups to each question, there is a surprising degree of agreement about many essential concepts and questions. We believe that this interdisciplinary and intertheoretical discussion will foster a more comprehensive understanding of tool use than any one of these perspectives (or any one of these author groups) would (or could) on their own

    Geographic variation and new taxa of western North American butterflies, especially from Colorado

    Dec. 3, 2008.Includes bibliographical references (pages 67-68).Michael Fisher is currently updating the 1957 book Colorado Butterflies, by F. Martin Brown, J. Donald Eff, and Bernard Rotger (Fisher 2005a, 2005b, 2006). This project has emphasized the necessity of naming certain butterflies in Colorado and vicinity that are distinctive, but currently have no name, as part of our goal of applying correct species/subspecies names to all Colorado butterflies. Eleven of those distinctive butterflies are named here, in the genera Anthocharis, Neominois, Asterocampa, Argynnis (Speyeria), Euphydryas, Lycaena, and Hesperia. New life histories are reported for species or subspecies of Neominois & Oeneis & Euphydryas & Lycaena that were recently described or recently elevated in status. Lycaena florus differs in hostplant, egg morphology, and somewhat in a seta on 1st-stage larvae. We also report the results of research elsewhere in North America that was needed to determine which of the current subspecies names should be applied to other butterflies in Colorado, in the genera Anthocharis, Neominois, Apodemia, Callophrys, Atlides, Euphilotes, PlebeJus, Polites, & Hylephila. This research has added additional species to the total of Colorado butterflies. Nomenclatural problems in Colorado Lycaena & Callophrys are settled with lectotypes and designations of type localities and two pending petitions to suppress toxotaxa. Difficulties with the ICZN Code in properly applying names to clines are explored, and new terminology is given to some necessary biological solutions

    Gas emissions, minerals, and tars associated with three coal fires, Powder River Basin, USA.

    Ground-based surveys of three coal fires and airborne surveys of two of the fires were conducted near Sheridan, Wyoming. The fires occur in natural outcrops and in abandoned mines, all containing Paleocene-age subbituminous coals. Diffuse (carbon dioxide (CO(2)) only) and vent (CO(2), carbon monoxide (CO), methane, hydrogen sulfide (H(2)S), and elemental mercury) emission estimates were made for each of the fires. Additionally, gas samples were collected for volatile organic compound (VOC) analysis and showed a large range in variation between vents. The fires produce locally dangerous levels of CO, CO(2), H(2)S, and benzene, among other gases. At one fire in an abandoned coal mine, trends in gas and tar composition followed a change in topography. Total CO(2) fluxes for the fires from airborne, ground-based, and rate of fire advancement estimates ranged from 0.9 to 780mg/s/m(2) and are comparable to other coal fires worldwide. Samples of tar and coal-fire minerals collected from the mouth of vents provided insight into the behavior and formation of the coal fires

    Macrophage origin limits functional plasticity in helminth-bacterial co-infection

    Rapid reprogramming of the macrophage activation phenotype is considered important in the defense against consecutive infection with diverse infectious agents. However, in the setting of persistent, chronic infection the functional importance of macrophage-intrinsic adaptation to changing environments vs. recruitment of new macrophages remains unclear. Here we show that resident peritoneal macrophages expanded by infection with the nematode Heligmosomoides polygyrus bakeri altered their activation phenotype in response to infection with Salmonella enterica ser. Typhimurium in vitro and in vivo. The nematode-expanded resident F4/80high macrophages efficiently upregulated bacterial induced effector molecules (e.g. MHC-II, NOS2) similarly to newly recruited monocyte-derived macrophages. Nonetheless, recruitment of blood monocyte-derived macrophages to Salmonella infection occurred with equal magnitude in co-infected animals and caused displacement of the nematode-expanded, tissue resident-derived macrophages from the peritoneal cavity. Global gene expression analysis revealed that although nematode-expanded resident F4/80high macrophages made an anti-bacterial response, this was muted as compared to newly recruited F4/80low macrophages. However, the F4/80high macrophages adopted unique functional characteristics that included enhanced neutrophil-stimulating chemokine production. Thus, our data provide important evidence that plastic adaptation of MΊ activation does occur in vivo, but that cellular plasticity is outweighed by functional capabilities specific to the tissue origin of the cell
