    Ecological communities are increasingly being recognized as the products of contemporary drivers and historical legacies that are both biotic and abiotic. In an attempt to unravel multiple layers of ecological contingency, we manipulated (i) competition with exotic annual grasses, (ii) the timing of this competition (temporal priority in arrival/seeding times) and (iii) watering (simulated rainfall) in a restoration-style planting of native perennial grasses. In addition, we replicated this experiment simultaneously at three sites in north-central California. Native perennial grasses had 73-99 % less cover when planted with exotic annuals than when planted alone, but this reduction was greatly ameliorated by planting the natives 2 weeks prior to the exotics. In a drought year, irrigation significantly reduced benefits of early planting so that these benefits resembled those observed in a non-drought year. There were significant differences across the three sites (site effects and interactions) in (i) overall native cover, (ii) the response of natives to competition, (iii) the strength of the temporal priority effect and (iv) the degree to which supplemental watering reduced priority effects. These results reveal the strong multi-layered contingency that underlies even relatively simple communities

    Investigating the role of Hedgehog/GLI1 signaling in glioblastoma cell response to temozolomide.

    Resistance to chemotherapy substantially hinders successful glioblastoma (GBM) treatment, contributing to an almost 100% mortality rate. Resistance to the frontline chemotherapy, temozolomide (TMZ), arises from numerous signaling pathways that are deregulated in GBM, including Hedgehog (Hh) signaling. Here, we investigate suppression of Hh signaling as an adjuvant to TMZ using U87-MG and T98G cell lines as in vitro models of GBM. We found that silencing GLI1 with siRNA reduces cell metabolic activity by up to 30% in combination with TMZ and reduces multidrug efflux activity by 2.5-fold. Additionally, pharmacological GLI inhibition modulates nuclear p53 levels and decreases MGMT expression in combination with TMZ. While we surprisingly found that silencing GLI1 does not induce apoptosis in the absence of TMZ co-treatment, we discovered silencing GLI1 without TMZ co-treatment induces senescence as evidenced by a significant 2.3-fold increase in senescence associated β-galactosidase staining, and this occurs in a loss of PTEN-dependent manner. Finally, we show that GLI inhibition increases apoptosis in glioma stem-like cells by up to 6.8-fold in combination with TMZ, and this reduces the size and number of neurospheres grown from glioma stem-like cells. In aggregate, our data warrant the continued investigation of Hh pathway inhibitors as adjuvants to TMZ chemotherapy and highlight the importance of identifying signaling pathways that determine whether co-treatment will be successful

    Home Delinquency Rates Are Lower Among ACA Marketplace Households: Evidence From a Natural Experiment

    This brief uses administrative income tax data coupled with survey responses from roughly 5,000 households living near the poverty line to estimate how access to the Affordable Care Act’s health insurance Marketplaces have affected households’ experiences of extreme illiquidity, which is measured by delinquencies on home payments. To estimate this relationship, we exploit a natural experiment underway in states that did not expand Medicaid and created by the eligibility rules for Marketplace subsidies. Results suggest that insured households living near the poverty line are better able to make timely rent and mortgage payments compared with similar, uninsured households. Given housing instability’s well-documented links with downstream financial and developmental outcomes, findings presented in this brief suggest that the Affordable Care Act’s health insurance programs have improved the short-term financial well-being of low- and moderate-income households in a way that may have longer term implications

    Light availability controls in the benthic nearshore ecosystem of the Elwha River

    The Elwha River Restoration Project was the largest US dam removal project to date, both in dam height and sediment released. During dam removal in 2011–2014, ~18 Mt of sediment washed downriver, and macroalgae virtually disappeared from the adjacent nearshore ecosystem. The link between current benthic light availability and sediment delivery and transport has been investigated in order to understand conditions during dam removal. Seven instrument platforms were deployed on the 10-m isobath along a 16 km transect centered on the river mouth for seven fortnightly periods in 2016 and 2017 to monitor near-bed photosynthetically available radiation (PAR), suspended sediment, wave climate, current velocity, temperature, and salinity. Water-column profiles, bed sediment, and water samples were collected during deployments. Seasonally variable chlorophyll-a and colored dissolved organic matter did not contribute substantially to light attenuation compared to suspended sediment. Along the 10-m isobath within 1.5 km of the river mouth, the greatest light attenuation occurred when wave events coincided with or followed periods of high river discharge. However, discharge events lasting attenuation; energetic tidal currents promote rapid sediment export out of the nearshore environment. In the buoyant plume, maximum light attenuation occurred within 1 m of the surface, reducing light through the rest of the water column. Benthic PAR varied more during spring tides when plume location was more variable. Alongshore 1.5 to 8 km from the river mouth, light availability was not directly coupled to river discharge. Light attenuation occurred throughout the water column, influenced by resuspension due to strong currents and wave events. This subsurface attenuation would not be captured by remote sensing. Predicting benthic light availability over event, tidal, and seasonal timescales will improve management strategies designed to limit ecosystem damage during other dam removals or sediment delivery events

    Household-level risk factors for secondary influenza-like illness in a rural area of Bangladesh

These permissions are granted for the duration of the World Health Organization (WHO) declaration of COVID-19 as a global pandemic.Objective To describe household‐level risk factors for secondary influenza‐like illness (ILI), an important public health concern in the low‐income population of Bangladesh. Methods Secondary analysis of control participants in a randomised controlled trial evaluating the effect of handwashing to prevent household ILI transmission. We recruited index‐case patients with ILI – fever (<5 years); fever, cough or sore throat (≥5 years) – from health facilities, collected information on household factors and conducted syndromic surveillance among household contacts for 10 days after resolution of index‐case patients’ symptoms. We evaluated the associations between household factors at baseline and secondary ILI among household contacts using negative binomial regression, accounting for clustering by household. Results Our sample was 1491 household contacts of 184 index‐case patients. Seventy‐one percentage reported that smoking occurred in their home, 27% shared a latrine with one other household and 36% shared a latrine with >1 other household. A total of 114 household contacts (7.6%) had symptoms of ILI during follow‐up. Smoking in the home (RRadj 1.9, 95% CI: 1.2, 3.0) and sharing a latrine with one household (RRadj 2.1, 95% CI: 1.2, 3.6) or >1 household (RRadj 3.1, 95% CI: 1.8–5.2) were independently associated with increased risk of secondary ILI. Conclusion Tobacco use in homes could increase respiratory illness in Bangladesh. The mechanism between use of shared latrines and household ILI transmission is not clear. It is possible that respiratory pathogens could be transmitted through faecal contact or contaminated fomites in shared latrines

    Case-fatality ratio of blood culture–confirmed typhoid fever in Dhaka, Bangladesh

    With impending rollout of new conjugate typhoid vaccines, better estimates of typhoid case-fatality ratio are needed for countries to set priorities for public health programs. We enrolled 1425 patients of all ages with blood culture–confirmed Salmonella Typhi from laboratory networks serving inpatients and outpatients in Dhaka, Bangladesh. Participants were asked about symptoms and complications including death experienced over a median 3-month period following blood culture diagnosis. Four fatal cases were identified (case-fatality ratio of 0.3% [95% confidence interval, .05%–.55%]). Applying this case-fatality ratio to global typhoid burden estimates would reduce deaths by 70%

    Axin2 as regulatory and therapeutic target in newborn brain injury and remyelination.

    Permanent damage to white matter tracts, comprising axons and myelinating oligodendrocytes, is an important component of brain injuries of the newborn that cause cerebral palsy and cognitive disabilities, as well as multiple sclerosis in adults. However, regulatory factors relevant in human developmental myelin disorders and in myelin regeneration are unclear. We found that AXIN2 was expressed in immature oligodendrocyte progenitor cells (OLPs) in white matter lesions of human newborns with neonatal hypoxic-ischemic and gliotic brain damage, as well as in active multiple sclerosis lesions in adults. Axin2 is a target of Wnt transcriptional activation that negatively feeds back on the pathway, promoting β-catenin degradation. We found that Axin2 function was essential for normal kinetics of remyelination. The small molecule inhibitor XAV939, which targets the enzymatic activity of tankyrase, acted to stabilize Axin2 levels in OLPs from brain and spinal cord and accelerated their differentiation and myelination after hypoxic and demyelinating injury. Together, these findings indicate that Axin2 is an essential regulator of remyelination and that it might serve as a pharmacological checkpoint in this process
