Single鈥恈ell profiling and zebrafish avatars reveal LGALS1 as immunomodulating target in glioblastoma
Glioblastoma (GBM) remains the most malignant primary brain tumor, with a median survival rarely exceeding 2鈥墆ears. Tumor heterogeneity and an immunosuppressive microenvironment are key factors contributing to the poor response rates of current therapeutic approaches. GBM鈥恆ssociated macrophages (GAMs) often exhibit immunosuppressive features that promote tumor progression. However, their dynamic interactions with GBM tumor cells remain poorly understood. Here, we used patient鈥恉erived GBM stem cell cultures and combined single鈥恈ell RNA sequencing of GAM鈥怗BM co鈥恈ultures and real鈥恡ime in vivo monitoring of GAM鈥怗BM interactions in orthotopic zebrafish xenograft models to provide insight into the cellular, molecular, and spatial heterogeneity. Our analyses revealed substantial heterogeneity across GBM patients in GBM鈥恑nduced GAM polarization and the ability to attract and activate GAMs鈥攆eatures that correlated with patient survival. Differential gene expression analysis, immunohistochemistry on original tumor samples, and knock鈥恛ut experiments in zebrafish subsequently identified LGALS1 as a primary regulator of immunosuppression. Overall, our work highlights that GAM鈥怗BM interactions can be studied in a clinically relevant way using co鈥恈ultures and avatar models, while offering new opportunities to identify promising immune鈥恗odulating targets.Deutsche Forschungsgemeinschaft (DFG)Fondation Leducq (Leducq Foundation)

Imaging Glioma Progression by Intravital Microscopy.
We describe here a method for generating mouse orthotopic gliomas in order to follow their progression over time by multi-photon laser scanning microscopy. After craniotomy of the parietal bone, glioma cells are implanted in the brain cortex and a glass window is cemented atop, allowing chronical imaging of the tumor. The expression of different fluorescent proteins in tumor cells and in specific cell types of a number of currently available transgenic mouse strains allows obtaining multicolor 3D images of the tumor over time. This technique is suitable both to evaluate the effect of pharmacological treatments and to unravel basic mechanisms of tumor-host interactions.status: publishe
Epidemiology of latrodectism in the province of Buenos Aires, Argentina
Epidemiological data related to Latrodectus bites over a 10-year period (1979-1988) in Buenos Aires Province, Argentina, are presented. Data on distribution of accidents by year, month, sex, residence of person bitten and general symptomatology are given. Statistical analysis showed a mean of 28.1 bites cases per year, 80% in males, most of whom were farm workers. The Latrodectus bites were localized in arms, pelvic waist and legs. Forty-six per cent of bitten persons visit the clinic within 1 to 3 hours after the incident, and only 15% capture the animal responsible for the accident.El presente trabajo tiene por objeto dar a conecer los resultados del Latrodectismo y su tratamiento especifico, durante un periodo de 10 a帽os (1979-1988) en la Provincia de Buenos Aires. Se suministran datos de la distribuci贸n de casos por a帽o, por meses, incidencia seg煤n sexo, residencia del accidentado, regi贸n corporal de la picadura, sintomatologia presentada, tiempo transcurrido entre el accidente y la aplicaci贸n del suero antilatrodectus y captura del animal agresor. Con respecto al a帽o se produjo um pico entre 1982 y 1983 con 38 accidentes, correspondiendo al periodo de 10 a帽os una media de 28,1 accidentes anuales los que presentan mayor incidencia de diciembre a marzo. Con respecto al sexo y residencia del accidentado un 80% corresponde a hombres, siendo principalmente trabajadores rurales los afectados. El mayor porcentaje de accidentados se produjo en antebrazos, cintura p茅lvica y muslos. Con respecto al tiempo transcorrido desde el accidente hasta la aplicaci贸n del suero, el 46% lo realiza entre las tres primeras horas. Endothelial PKA activity regulates angiogenesis by limiting autophagy through phosphorylation of ATG16L1
The cAMP-dependent protein kinase A (PKA) regulates various cellular functions in health and disease. In endothelial cells PKA activity promotes vessel maturation and limits tip cell formation. Here, we used a chemical genetic screen to identify endothelial-specific direct substrates of PKA in human umbilical vein endothelial cells (HUVEC) that may mediate these effects. Amongst several candidates, we identified ATG16L1, a regulator of autophagy, as novel target of PKA. Biochemical validation, mass spectrometry and peptide spot arrays revealed that PKA phosphorylates ATG16L1伪 at Ser268 and ATG16L1尾 at Ser269, driving phosphorylation-dependent degradation of ATG16L1 protein. Reducing PKA activity increased ATG16L1 protein levels and endothelial autophagy. Mouse in vivo genetics and pharmacological experiments demonstrated that autophagy inhibition partially rescues vascular hypersprouting caused by PKA deficiency. Together these results indicate that endothelial PKA activity mediates a critical switch from active sprouting to quiescence in part through phosphorylation of ATG16L1, which in turn reduces endothelial autophagy.status: publishe
