Ultrastructural and functional aspects of the spermatheca in the american harlequin bug, Murgantia histrionica (Hemiptera: Pentatomidae)
The spermatheca of Murgantia histrionica (Hahn) was investigated using fluorescence, scanning and transmission electron microscopy. The aim of the study was to elucidate the structure of this organ, pointing out differences between mated and unmated females. Results have shown an elaborated cuticular structure associated with muscular and glandular tissues. The spermatheca is joined with the common oviduct by the spermathecal duct, forming a thin saccular dilation through two consecutive invaginations. The distal part of the organ is formed by a series of two communicating cuticular chambers. The first cylindrical-shaped chamber, corresponding to the coiled region, is wrapped by longitudinal muscular fibers suspended between two cuticular flanges. The contractions of these fibers compress a deformable zone of the cylinder, pumping the sperm toward the spermathecal duct. Without contractions the cylinder results to be isolated from the proximal part of the spermatheca by means of a valve. The second chamber, corresponding to the spermatheca, is made of two parts: a truncated-conical sub chamber, with a constant cuticular thickness, bearing on itself the distal flange, where muscular fibers are attached. The second part is a bulb-like structure wrapped in a glandular epithelium. The secretory units are composed by two cells: a secretory cell and an associated duct cell. Every evacuating duct shows a little reservoir just after the terminal apparatus, and converge inside the distal bulb after a tortuous path. The functional implications of this structure in the reproductive biology of M. histrionica are discussed
Semiochemicals, semiophysicals and their integration for the development of innovative multi-modal systems for agricultural pestsâ monitoring and control
4openInternationalSemiochemicals are informative molecules emitted by living organisms that affect the behavior of receivers. As herbivorous insects are primarily thought to depend on olfaction and taste for their intra- and interspecific communication, semiochemicals have been widely studied for pest management applications. However, given that pest behavior does not rely on just one communication modality, stimuli of physical nature, such as light, sounds and vibrations, can also be used to manipulate insect-insect or insect-plant interactions. Moreover, stimuli of different natures can be combined in a multi-modal pest management program to increase the overall efficacy. Besides the widespread use of both chemical and physical signals in multimodal insect communication, the integration of stimuli has hardly been implemented for hardly any crop. This review introduces the term semiophysicals as opposed to semiochemicals and focuses on pest behavioral manipulation by discussing three main approaches; i) manipulation of pest orientation through attractive/repellent stimuli, ii) inhibition or promotion of specific pest behaviors and iii) interference with intraspecific communication through disruptive stimuli. For each approach, we provide examples of use of both semiochemicals and semiophysicals. Influenza degli stimoli vibrazionali sulla ricerca dell'ospite da parte di G1 Ganaspis cf. brasiliensis
Il parassitoide larvale G1 Ganaspis cf. brasiliensis (Hymenoptera: Figitidae) è stato selezionato come agente di controllo per il programma di controllo biologico classico di Drosophila suzukii (SWD). Il parassitoide mostra alta specificità e attacca 1° e 2° stadio larvale dell'ospite all'interno della frutta infestata. Mentre la ricerca dell'ospite a lungo e medio raggio è fortemente influenzata da stimoli chimici, il riconoscimento dell'ospite all'interno del suo microhabitat è possibilmente mediato da vibrazioni incidentali prodotte dalla larva in fase di sviluppo all'interno del frutto stesso. Abbiamo identificato e descritto tali vibrazioni mettendole in relazione al livello e tempo di infestazione. Abbiamo poi svolto delle prove comportamentali per investigare se G.brasiliensis utilizzi stimoli vibrazionali per riconoscere e discriminare il proprio ospite. Le risposte del parassitoide sono state analizzate considerando il tempo dedicato alla ricerca, ed il numero di inserzioni dell'ovipositore
Aggiornamento sul programma nazionale italiano di lotta biologica classica al moscerino asiatico dei piccoli frutti, Drosophila suzukii
Vengono presentate le attivitaĚ del Tavolo tecnico-scientifico nazionale di coordinamento per la lotta al moscerino asiatico dei piccoli frutti, Drosophila suzukii Matsumura, istituito formalmente su indicazione del Comitato Fitosanitario Nazionale nel gennaio 2021. La principale finalitaĚ del Tavolo eĚ quella di definire e condividere le attivitaĚ di contrasto al fitofago invasivo attraverso la programmazione di un piano triennale di controllo biologico classico che prevede lâimpiego del parassitoide esotico Ganaspis brasiliensis Ihering. La prima campagna di lanci del parassitoide, partita nellâagosto del 2021 a seguito dellâautorizzazione da parte dellâex Ministero della Transizione Ecologica (ora Ministero dellâAmbiente e della Sicurezza Energetica), ha visto la partecipazione delle regioni Campania, Emilia-Romagna, Piemonte, Puglia, Sicilia, Valle dâAosta e Veneto e delle Province autonome di Trento e Bolzano. Nel 2023 si aggiungeranno le regioni Lombardia e Toscana. I risultati dei monitoraggi delle prime due stagioni, eseguiti sul 100% dei siti di lancio tra maggio e novembre di ogni anno, suggeriscono che G. brasiliensissi stia insediando in varie aree del territorio nazionale e ne confermano la specificitaĚ nei confronti di D. suzukii, giaĚ osservata negli studi di laboratorio
Comparative neuroanatomy of the antennal lobes of 2 homopteran species
We compared the morphology of the primary olfactory center, the antennal lobe (AL), in 2 homopteran insects, Hyalesthes obsoletus Signoret (Homoptera: Cixiidae) and Scaphoideus titanus Ball (Homoptera: Cicadomorpha). The comparison between the ALs of the 2 species is particularly interesting considering that, although both use volatile cues to locate their host plants, their feeding behavior differs considerably: specifically, H. obsoletus is a highly polyphagous species, whereas S. titanus is strictly monophagous (on grapevine). Our investigation of the AL structure using immunocytochemical staining and antennal backfills did not reveal any sexual dimorphism in either the size of the ALs or in the size of individual glomeruli for either species. Instead, the AL of H. obsoletus displayed numerous and well-delineated glomeruli (about 130 in both sexes) arranged in a multilayered structure, whereas the smaller AL of S. titanus contained fewer than 15 glomerular-like structures. This difference is likely to reflect the comparatively reduced olfactory abilities in S. titanus, probably as a consequence of the reduced number of volatiles coming from the single host plant. Instead, in H. obsoletus, the ability to distinguish among several host plants may require a more complex olfactory neuronal networ
Fine Structure of the Antennal Glands of the Ant Nest Beetle Paussus favieri (Coleoptera, Carabidae, Paussini).Corresponding author
The antennae of the ant nest beetle Paussus favieri are studied by using both SEM and TEM. In the myrmecophilous genus Paussus, these structures are composed of three joints: scape, pedicel and a wide third joint, the ââantennal clubââ, resulting from the fusion of antennomeres A3âA11 (flagellum). The antennal club shows an exceptional glandular activity, with the presence of pores mostly crowded in special hairless cuticular areas, surrounding the base of single setae, grouped at the base of tufts of setae, or positioned inside deep pockets that store the secretions, with filiform material arising from them. The surface of A1 and A3 are covered by mechanoreceptors, modified to spread the glandular exudates, while the chemoreceptors are restricted to the apex of the club. The fine structural analysis shows a great number of antennal glands, that can be referred to three main typologies: type A (GhA) bi-cellular, composed of a large secretory cell and a small duct cell, positioned close to the antennal surface; type B (GhB), tri-cellular, composed of two secretory cells and one duct cell, less frequent and positioned deep inside the antennal club; type C (GhC), rare, located deeply within the antennal lumen, in the vicinity of the trophocytes. This complexity indicates that more than one substance could be released from the antennae. Possible functional aspects of the secretions dealing with symbiotic interaction with the host ants are discussed
Verso una semplificazione strutturale: il caso delle antenne degli Auchennorrhyncha
Gli Auchennorrhyncha sono un gruppo di Emitteri caratterizzati da dimensioni
generalmente ridotte e corpo allungato. Numerose specie, appartenenti ai due diversi
infraordini dei Cicadomorpha e Fulgoromorpha, rivestono un importante ruolo allâinterno
di vari ecosistemi (sia naturali che coltivati) in quanto si comportano da vettori di
pericolose fitopatie. Esempi a tale riguardo sono Scaphoideus titanus Ball (vettore del
fitoplasma agente della flavescenza dorata), Hyalesthes obsoletus Signoret (vettore
naturale del fitoplasma agente causale del Legno nero) e Philaenus spumarius L. (vettore
del batterio Xylella fastidiosa, associato al Complesso del Disseccamento Rapido
dellâOlivo, CoDiRO). Le antenne di diverse specie di Auchenorrhyncha presentano una
struttura generale piuttosto semplificata, caratterizzata generalmente dalla presenza di uno
scapo e un pedicello subcilindrici, ed un flagello molto allungato e sottile. Il flagello
mostra una forte riduzione nel comparto sensoriale, in alcuni casi con una forte riduzione
dei sensilli olfattivi. Il flagello è inoltre articolato sul pedicello attraverso una membrana
articolare associata ad un voluminoso organo di Johnston ospitato nel pedicello. In alcuni
casi i sensilli olfattivi sono presenti in gran numero sulla superficie del pedicello. Diverse
tipologie sensoriali (sensilli basiconici, celoconici e ampullacei) sono presenti nella parte
basale ingrossata del flagello. Il ruolo che queste strutture sensoriali potrebbero avere nella
biologia di alcune specie viene discusso, tracciando ipotesi relative alla riduzione
funzionale dei sensilli a livello del flagello e la riduzione del comparto sensoriale olfattivo
Functional structure of antennal sensilla in the myrmecophilous beetle Paussus favieri (Coleoptera, Carabidae, Paussini). Corresponding author
The evolution of a myrmecophilous lifestyle in beetles is often associated with morphological alterations. In particular, the antennae of all members of the myrmecophilous ground beetle tribe Paussini are greatly modified, with flagellomeres flattened or crassate, frequently reduced in number from 9 to 5 or even 1 single âantennal clubâ. The enhanced glandular function of the an-tennal club has been recently described by scanning (SEM) and transmission (TEM) electron mi-croscopy in Paussus favieri Fairmaire, 1851, where the antenna has become a complex glandular organ, supplying rewarding substances to the ants. In the present work, the antennal sensilla of P. favieri are investigated by SEM, TEM and Focused Ion Beam (FIB/SEM) technology. Most sen-silla of scape and antennal club are highly modified mechanoreceptors (i.e. multipointed, fringed, branched, brush-like, sickle-shaped), singly or grouped in tufts (âantennal symphilous organsâ). These âtrichomesâ, here assigned to 8 different morphotypes of sensilla chaetica (Ch.1â Ch.8), show a variable number of basal pores (present also at the base of the taste sensilla Ch.9), which spread dense substances of unknown chemical composition on the seta. Although hygro-, ther-mo- and chemoreceptors are reduced in number as compared with non-myrmecophilous rela-tives, and mainly relegated to the apex of the antennal club, their diversity is comparable to that of other carabid beetles: two types of sensilla trichodea (Tr.1â Tr.2); three types of basiconica (Ba.1â Ba.3); one type of campaniformia (Ca); one type of coeloconica (Co) and one type of BĂśhm sensilla (Bo). Contrary to the hypothesis that Paussus species lack a Johstonâs organ, a non-connective chordotonal organ composed of 9 groups of scolopidia has been found inside the pedicel. A comparison between sensilla of P. favieri and those of other non-myrmecophilous and myrmecophilous ground beetle species is provided
The sensory structures of the antennal flagellum in Hyalesthes obsoletus (Hemiptera: Fulgoromorpha: Cixiidae): A functional reduction?
Despite their relevance as harmful pests on plants of economic importance, Hemiptera Fulgoromorpha have been poorly studied as regards their antennal sensory structures. In particular, the flagellum has been neglected and, therefore, to date there are no data on its structural organization and sensory equipment. In order to fill this gap, we carried out a study on the sensillum types and distribution on the flagellum of the planthopper Hyalesthes obsoletus Signoret, an efficient vector of the stolbur phytoplasma, the cause of various crop diseases. In this cixiid species the antenna is composed of three segments, the scape, an enlarged pedicel and a long flagellum. This latter is made of a single segment and presents a basal, bulb-like enlargement from which two processes arise, a short spur and a long arista. Combining scanning electron microscopy, transmission electron microscopy and focused ion beam investigations, we discovered the presence of a total number of 6 sensilla, belonging to 4 different types: a single scolopidium extending from the bulb to the arista, three sensilla styloconica within the cuticular spur and two different sensilla coeloconica inside the bulb. As far as structural data can suggest, these sensilla might be involved in the perception of mechanical stimuli (possibly air-borne vibrations), temperature and humidity variations and CO2 concentration. The strong reduction in sensillum number in this species is discussed as possible functional specialization of the flagellum itself. Le strutture sensoriali del flagello di Hyalesthes obsoletus: una riduzione funzionale?
Hyalesthes obsoletus Signoret (Hemiptera, Fulgoromorpha, Cixiidae), vettore naturale
del fitoplasma agente causale del Legno nero della vite, è un ampelofago occasionale. In
Europa e nel bacino del Mediterraneo lâinsetto può svolgere il suo ciclo di sviluppo
preimmaginale a carico di diverse piante spontanee (Urtica dioica, Convolvulus
arvensis, Calystegia sepium, Lavandula hybrida, Lavandula angustifolia, Cardaria
draba e Vitex agnus-castus). I meccanismi sensoriali che permettono al fitofago di
localizzare i conspecifici per lâaccoppiamento e le potenziali piante ospiti per
lâalimentazione e lâovideposizione fanno capo agli organi di senso localizzati sulle
antenne. Le conoscenze relative alle strutture antennali di questo gruppo di insetti sono
abbastanza scarse, pertanto scopo di questo lavoro è stata lâindagine ultrastrutturale dei
sensilli presenti sul flagello di H. obsoletus. In questa specie il flagello è in realtà ridotto
ad un singolo articolo antennale piuttosto allungato, che presenta una parte apicale che
tende ad assottigliarsi fino allâapice, mentre nella parte basale si allarga a formare una
sorta di bulbo. Il bulbo è caratterizzato dalla presenza di unâapertura distale e, sempre
nella parte distale, da uno sperone cuticolare tripartito. Sezioni seriate longitudinali e
trasversali osservate al microscopio elettronico a trasmissione hanno messo in evidenza
la presenza di un singolo scolopidio localizzato nel lume del flagello. A livello del bulbo,
in corrispondenza dellâapertura, sono stati riscontrati due sensilli celoconici con
caratteristiche morfologiche ben distinte. Lo sperone è costituito dalla fusione di tre
sensilli stiloconici separati nella parte apicale. Il ruolo che queste strutture sensoriali
potrebbero avere nella biologia di H. obsoletus viene discusso, con particolare
riferimento alla riduzione funzionale dei sensilli a livello del flagello e la totale assenza
del comparto sensoriale olfattivo
