    Effects of nanosuspension and inclusion complex techniques on the in vitro protease inhibitory activity of naproxen

    This study investigated the effects of nanosuspension and inclusion complex techniques on in vitro trypsin inhibitory activity of naproxen—a member of the propionic acid derivatives, which are a group of antipyretic, analgesic, and non-steroidal anti-inflammatory drugs. Nanosuspension and inclusion complex techniques were used to increase the solubility and anti-inflammatory efficacy of naproxen. The evaporative precipitation into aqueous solution (EPAS) technique and the kneading methods were used to prepare the nanosuspension and inclusion complex of naproxen, respectively. We also used an in vitro protease inhibitory assay to investigate the anti-inflammatory effect of modified naproxen formulations. Physiochemical properties of modified naproxen formulations were analyzed using UV, IR spectra, and solubility studies. Beta-cyclodextrin inclusion complex of naproxen was found to have a lower percentage of antitryptic activity than a pure nanosuspension of naproxen did. In conclusion, nanosuspension of naproxen has a greater anti-inflammatory effect than the other two tested formulations. This is because the nanosuspension formulation reduces the particle size of naproxen. Based on these results, the antitryptic activity of naproxen nanosuspension was noteworthy; therefore, this formulation can be used for the management of inflammatory disorders.O objetivo do presente estudo foi investigar a atividade anti-inflamatória in vitro de nanossuspensões e do complexo de inclusão contendo naproxeno. Esse fármaco é derivado de ácido propiônico, com ação analgésica, antipirética e antiinflamatória. A obtenção dessas formulações teve por finalidade o aumento da solubilidade e da atividade anti-inflamatória do fármaco. Os métodos por precipitação em solução aquosa por evaporação e por empastagem foram modificados para a obtenção da nanossuspensão e do complexo de inclusão, respectivamente. Para a avaliação da atividade anti-inflamatória das formulações utilizou-se ensaio in vitro modificado de inibição de tripsina. As propriedades físico-químicas das formulações propostas foram determinadas utilizando espectroscopia UV e de infravermelho, além de estudos de solubilidade. O complexo de inclusão de naproxeno apresentou menor atividade antitripsina, quando comparado ao composto livre e à nanossuspensão. Em conclusão, entre as formulações avaliadas, a nanossuspensão de naproxeno apresentou maior efeito anti-inflamatório. Esse efeito foi devido à redução da dimensão das partículas de naproxeno para a escala nanométrica. Com base nos resultados obtidos, a atividade da nanossuspensão de naproxeno foi notável. Dessa forma, essa formulação apresenta potencial para o tratamento de distúrbios inflamatórios

    EFFICACY OF CHUNDAI VATRAL CHOORNAM A SIDDHA POLY HERBAL FORMULATION IN TREATING TENSION TYPE OF HEADACHE (TTH) -A CASE REPORT

    Siddha system is an ancient system of medicine which is popularly practised around south India particularly in Tamilnadu. Siddhars were considered as the pioneer of Siddha system, this system of medicine mainly encompasses for a healthy life for human beings. Among the broad spectrum of treatment aspects in Siddha system Siddhars classify the forms of internal medicines into 32 types which are all unique by its preparations. Choornam is one of the forms of internal medicine which can be used as single as well as poly herbal formulations. In this case report a poly-herbal formulation in Siddha system was used to treat tension type of headache (TTH). A 30 years old male patient visited out-patient department of Ayothidoss Pandither Hospital in National Institute of Siddha, Chennai. Patient reported with the complaints of episodic and chronic headache which is band like around the head, the intensity becomes mild to moderate, pain increased during working hours for past 2 years. Patient advised to follow the internal medicine Chundai vatral choornam it relives the TTH immediately when he got those symptoms.       &nbsp

    A REVIEW ON HYPO HIDROTIC EFFECT OF SIDDHA FORMULATION - KUNGUMAPOO MATHIRAI

    Hyperhidrosis is a condition present with excess sweating of palms on the hands, soles of the feet, forehead and axillae. This is socially embarrassing. There are few herbs to treat hyperhidrosis in Siddha system of medicine. The drug Kungumapoo Mathirai is being used in Siddha system of medicine for many years to treat hyperhidrosis. The ingredients used in the preparation of Kungumapoo Mathirai are Crocus sativus, Zingiberofficinale, Piper nigrum, Piper cubeba, Costus speciosus, Carum capticum, Piper longum, Syzygium aromaticu, Elettaria cardomom, Korosanai (Oxbile), Vengaram (Sodium biborate), Lingam (Red sulphide of mercury), Sambrani poo (Dry powder of Styrax benzoin). This article reviews the hypo hydration effect of the drug Kungumapoo Mathirai and its ingredients. This review will help to get much information on hypohydrosis effect of the chemical constituents of the drug. The antioxidant properties of the ingredients which is acting as hypo hydrating agent and their mechanism in controlling the nervous excitement by reducing the action potential in detail. The use of herbal drugs is becoming more popular due to the adverse effects of synthetic Anticholenergics and Antispasmodic drugs which are used to treat hyperhidrosis worldwide. Excellence of Siddha, formulated each and every medicines on the basis of Thiridhosa theory and five elemental theory, which are the basic principles of Siddha medicine.     ANTI ARTHRITIC ACTIVITY OF HERBO MINERAL SIDDHA PREPARATION ARUMUGA CHENDURAM AGAINST TYPE II COLLAGEN INDUCED ARTHRITIS IN EXPERIMENTAL RATS

    Siddha system of medicine is the eternal science of life. It is a system that has its extensive bonding with Dravidian culture, language and beliefs. The system of medicine mostly prevailed and prospered in the regions of Dravidian cultures by the great Siddhars. It’s unique as one only than other AYUSH traditional systems of medicine across India with its distinctive abundant usage of medicinal plants, metals, minerals and animal products. Siddhars used the steps of Alchemy to prepare various medicines from metallic and mineral origin for attainment elixir and various rare diseases. Siddha medicine is classified into 32 types of internal and external medicine each. Among the 32 types of internal medicine Chendhuram is a medicine shelf life of 75 years usually from herbo-mineral combinations. Arumuga Chendhuram (ARC) is a herbo-mineral formulation cited in Siddha literature ‘Siddha Vaithiya Thirattu’. ARC was orally administered at higher dose 2gm/kg to the Wistar Albino rats in acute toxicity study and during 28 days of repeated (sub acute) toxicity study, at daily doses of 12, 24 & 48mg/kg of body weight to the Wistar Albino rats. Type II collagen arthritis is another model for developing autoimmune arthritis. The immune pathogenesis mediated by T cell and B cell response to collagen. By this model, nearly 100% arthritis can be achieved. In our study, ARC after 42 days treatment reduced the arthritic swelling significantly and degree of inflammation evident to act against auto immune disorder

    A REVIEW ON SIDDHA EXTERNAL THERAPY - NASIYAM (NASAL INSTILLATION)

    Siddha system of medicine, one of the ancient, traditional Indian systems of medicine has unique diagnostic methods, therapeutics and treatment procedures. It has a vast range of external modalities of treatment for health management. This external management is classified into 32 types. They include minor surgical procedures and treatment procedures. These therapies are used both as mainstream and supportive therapies - both curative and prophylactic. Most of the therapies are aimed to maintain the equilibrium of the three humours (Vatham, Pitham, Kabam). Among them Nasiyam (Nasal instillation) is one of the external therapies which is a non-invasive procedure. Nasiyam is the process by which the drug is administered through nostrils. This is the treatment procedure to balance the Thirithodam in its normal level. This paper documents the efficacy of Nasiyam, methods of application, shelf life, effect of Nasiyam in treating various diseases, the list of single drug and compound drug formulations that can be used as Nasiyam, Indications and contraindications are discussed in detail.     NEERKURI BY SAGE THERAIYAR - A REVIEW ON SIDDHA WAY OF URINE EXAMINATION IN THE LIGHT OF CONTEMPORARY CLINICAL METHODS

    The Siddha system is said to have emerged in antiquity, from the highly evolved consciousness of the Siddhars. The clarified intellect and heightened intuition of Sage Theraiyar, resulting from Yogic powers, enabled to expound to the world the signs of a disease in the Urine of a person in his treatise “Theraiyar Neerkuri Vaithyam†a book that details urine examination findings for diagnostic and prognostic purposes. The present paper deals with scientific validation of Neerkuri (urine examination), an important diagnostic tool in Siddha system of medicine. The methodology of diagnosis in Siddha system is based on eight fold examination of pulse, tactile perception, tongue, color and complexion, speech, eyes, stools and urine. Of all these parameters, Urine examination has gained paramount importance next to pulse examination. This paper parallels analyses the signs of urine examination between a few quotes from Sage Theraiyar text and modern text. This is an attempt to understand the Siddha system of diagnosing pathological conditions which are a non-invasive, highly cost effective procedure which can be used for both diagnostic and prognostic purposes

    Synthons and Design in Metal Phosphates and Oxalates with Open Architectures

    We briefly describe the structures of open-framework metal phosphates with different dimensionalities, such as the one-dimensional linear-chain and ladder structures, two-dimensional layer structures and three-dimensional structures with channels. We demonstrate the role of the zero-dimensional four-membered ring monomer and of the one-dimensional ladder structure as the starting building units or synthons involved in the formation of the complex architectures. Thus, we show how the one-dimensional ladder structure transforms to two- and three-dimensional structures under mild conditions. The two-dimensional layer structures also transform to three-dimensional structures, while the zero-dimensional monomer transforms to layered and three-dimensional structures under ordinary reaction conditions. These transformations provide an insight into the possible pathways involved in the building up of the complex structures of metal phosphates. The isolation of amine phosphates during the hydrothermal synthesis of metal phosphates and also the facile reactions between amine phosphates and metal ions to yield a variety of open-framework materials have thrown light on the mechanism of formation and design of these structures. The existence of a hierarchy of open-framework metal oxalates and their ready formation by employing amine oxalates as intermediates provides additional support to the observations made earlier with regard to the phosphates

    DETERMINATION OF DIAGNOSTIC SIGNIFICANCE OF WRIST CIRCUMETRIC SIGN IN SIDDHA SYSTEM OF MEDICINE

    Aim: Determination of diagnostic significance of wrist circumetric sign in siddha system of medicine.Materials and methods: Patients are randomly selected between the age 13years to 70 years with selected diagnoses like Madhumegam, Athikuruthiazhutham (Hypertension), Thamaraganoi, Putrunoi, Kalanjagapadai (Psoriasis), Moolam, Powthiram (Perianal fistula), Vali azhal keel vayu, Pakkavatham, Karupaisathaikatti (Uterine fibroids).Results and observation: Most of the patients MKN values fell within the range of 9-9¾ fb. This range of 9-9¾ fb MKN, was observed in Athikuruthiazhutham, Putrunoi, Moolam, Powthiram, Thamaraganoi, Madhumegam, Pakkavatham, healthy volunteers. 8-8¾fb MKN range was observed in 41.5% of Kalanjagapadai and vali azhal keel vayu cases. 10 - 10¾fb was observed in 43.1 % of patients with Karuppaisathaikattigal (Uterine fibroids).Conclusion: Since all other diseases had non-specific Manikkadai nool range except the condition Karupaisathaikattigal (uterine fibroid), Manikkadai nool can be used to assess screening test in detecting the presence of uterine fibroids and cardiac diseases according to this study.

    Anti-Urolithiatic Activity of Melia Azedarach Linn Leaf Extract in Ethylene Glycol-Induced Urolithiasis in Male Albino Rats

    Purpose: To investigate the anti-urolithiatic activity of the aqueous and alcoholic extracts of Melia azedarach Linn leaves in calcium oxalate urolithiasis in male albino rats.Methods: The effect of oral administration of aqueous and ethanol extracts of Melia azedarach Linn leaves on calcium oxalate urolithiasis has been investigated. Lithiasis was induced by oral adminstration of ethylene glycol (0.75 %v/v) in male albino rats for 28 days. Each of the extract (250 mg/kg) was administered orally day 0 as a prophylactic regimen and from day 15 as a curative regimen. Regular administration of ethylene glycol caused hyperoxaluria in ethylene glycol-fed animals, leading to increased renal retention and excretion of oxalate, calcium and phosphate. Histopathological study, urine microscopy, serum analysis and biochemical analysis of kidney homogenate were performed.Results: Oxalate and calcium excretion in urine increased (p < 0.01) to 3.68 ± 0.01 and 4.5 ± 0.01 mg/24 h, respectively, in lithiatic control animals compared to (0.37 ± 0.01 and 1.27 ± 0.12 mg/24 h) for the normal control group. Treatment with aqueous or ethanol extract (250 mg/kg, p.o.) significantly (p <0.01) reduced the elevated levels of calcium, oxalate and phosphate excretion in urine to 0.79 ± 0.01 and 1.09 ± 0.04 mg/24 h, respectively. Following treatment with the ethanol extract (250mg/kg), serum creatinine excretion was restored from 0.95 ± 0.01 mg/24 h to the normal level of 0.87 ± 0.01 mg/24 h. The results were comparable to those of the standard drug, allopurinol (50 mg/kg p.o.).Histopathological data for the kidney supported the foregoing results.Conclusions: The results demonstrate that the aqueous and ethanol extracts of Melia azedarach Linn leaves have potent antiurolithiatic activity against ethylene glycol-induced calcium oxalate urolithiasis in male albino rats.Keywords: Melia azedarach, Antiurolithiatic, Ethylene glycol, Urolithiasis, Excretion, Kidne
