    Interactions between metabolic, reward and cognitive processes in appetite control:Implications for novel weight management therapies

    Traditional models of appetite control have emphasised the role of parallel homeostatic and hedonic systems, but more recently the distinction between independent homeostatic and hedonic systems has been abandoned in favour of a framework that emphasises the cross talk between the neurochemical substrates of the two systems. In addition, evidence has emerged more recently, that higher level cognitive functions such as learning, memory and attention play an important role in everyday appetite control and that homeostatic signals also play a role in cognition. Here, we review this evidence and present a comprehensive model of the control of appetite that integrates cognitive, homeostatic and reward mechanisms. We discuss the implications of this model for understanding the factors that may contribute to disordered patterns of eating and suggest opportunities for developing more effective treatment approaches for eating disorders and weight management

    Construction of coronal hole and active region magnetohydrostatic solutions in two dimensions: Force and energy balance

    Coronal holes and active regions are typical magnetic structures found in the solar atmosphere. We propose several magnetohydrostatic equilibrium solutions that are representative of these structures in two dimensions. Our models include the effect of a finite plasma-β and gravity, but the distinctive feature is that we incorporate a thermal structure with properties similar to those reported by observations. We developed a semi-analytical method to compute the equilibrium configuration. Using this method, we obtain cold and under-dense plasma structures in open magnetic fields representing coronal holes, while in closed magnetic configurations, we achieve the characteristic hot and over-dense plasma arrangements of active regions. Although coronal holes and active regions seem to be antagonistic structures, we find that they can be described using a common thermal structure that depends on the flux function. In addition to the force balance, the energy balance is included in the constructed models using an a posteriori approach. From the two-dimensional computation of thermal conduction and radiative losses in our models, we infer the required heating function to achieve energy equilibrium. We find that the temperature dependence on height is an important parameter that may prevent the system from accomplishing thermal balance at certain spatial locations.     Tryptophan depletion disinhibits punishment but not reward prediction: implications for resilience

    Item does not contain fulltextRATIONALE: We have previously shown that tryptophan depletion enhances punishment but not reward prediction (Cools et al. in Neuropsychopharmacology 33:2291-2299, 2008b). This provided evidence for a valence-specific role of serotonin (which declines under depleted tryptophan) in aversive processing. Recent theoretical (Dayan and Huys in PLoS Comput Biol 4:e4, 2008) and experimental (Crockett et al. in J Neurosci 29:11993-11999, 2009) approaches have, however, further specified this role by showing that serotonin is critical for punishment-induced inhibition. OBJECTIVES: We sought to examine the role of serotonin in punishment-induced inhibition. We also examined the impact of induced mood on this effect to assess whether effects of tryptophan depletion on affective inhibition are moderated by mood. METHODS: Healthy females consumed a balanced amino acid mixture with (N = 20) or without (N = 21) the serotonin precursor tryptophan. Each subject completed either negative or neutral mood induction. All subjects completed the reward and punishment reversal learning task adopted in the previous study. RESULTS: We demonstrate a punishment prediction impairment in individuals who consumed tryptophan which was absent in individuals who were depleted of tryptophan. This effect was impervious to mood state. CONCLUSIONS: Our results suggest that serotonin promotes the inhibition of responses to punishing outcomes. This may lead to reduced punishment prediction accuracy in the presence of tryptophan and may contribute to resilience to affective disorders. Reduction of serotonin via tryptophan depletion then removes this inhibition. As such, we highlight a mechanism by which reduced serotonin can contribute to disorders of impulsivity and compulsivity as well as disorders of emotion.1 januari 201

    Beyond the disk: EUV coronagraphic observations of the Extreme Ultraviolet Imager on board Solar Orbiter

    Most observations of the solar corona beyond 2 Rs consist of broadband visible light imagery from coronagraphs. The associated diagnostics mainly consist of kinematics and derivations of the electron number density. While the measurement of the properties of emission lines can provide crucial additional diagnostics of the coronal plasma (temperatures, velocities, abundances, etc.), these observations are comparatively rare. In visible wavelengths, observations at these heights are limited to total eclipses. In the VUV range, very few additional observations have been achieved since the pioneering results of UVCS. One of the objectives of the Full Sun Imager (FSI) channel of the EUI telescope on board the Solar Orbiter mission has been to provide very wide field-of-view EUV diagnostics of the morphology and dynamics of the solar atmosphere in temperature regimes that are typical of the lower transition region and of the corona. FSI carries out observations in two narrowbands of the EUV spectrum centered on 17.4 nm and 30.4 nm that are dominated, respectively, by lines of Fe IX/X (formed in the corona around 1 MK) and by the resonance line of He II (formed around 80 kK in the lower transition region). Unlike previous EUV imagers, FSI includes a moveable occulting disk that can be inserted in the optical path to reduce the amount of instrumental stray light to a minimum. FSI detects signals at 17.4 nm up to the edge of its FOV (7~Rs), which is about twice further than was previously possible. Comparisons with observations by the LASCO and Metis coronagraphs confirm the presence of morphological similarities and differences between the broadband visible light and EUV emissions, as documented on the basis of prior eclipse and space-based observations. The very-wide-field observations of FSI are paving the way for future dedicated instruments

    Criteria of validity for animal models of psychiatric disorders: focus on anxiety disorders and depression

    Animal models of psychiatric disorders are usually discussed with regard to three criteria first elaborated by Willner; face, predictive and construct validity. Here, we draw the history of these concepts and then try to redraw and refine these criteria, using the framework of the diathesis model of depression that has been proposed by several authors. We thus propose a set of five major criteria (with sub-categories for some of them); homological validity (including species validity and strain validity), pathogenic validity (including ontopathogenic validity and triggering validity), mechanistic validity, face validity (including ethological and biomarker validity) and predictive validity (including induction and remission validity). Homological validity requires that an adequate species and strain be chosen: considering species validity, primates will be considered to have a higher score than drosophila, and considering strains, a high stress reactivity in a strain scores higher than a low stress reactivity in another strain. Pathological validity corresponds to the fact that, in order to shape pathological characteristics, the organism has been manipulated both during the developmental period (for example, maternal separation: ontopathogenic validity) and during adulthood (for example, stress: triggering validity). Mechanistic validity corresponds to the fact that the cognitive (for example, cognitive bias) or biological mechanisms (such as dysfunction of the hormonal stress axis regulation) underlying the disorder are identical in both humans and animals. Face validity corresponds to the observable behavioral (ethological validity) or biological (biomarker validity) outcomes: for example anhedonic behavior (ethological validity) or elevated corticosterone (biomarker validity). Finally, predictive validity corresponds to the identity of the relationship between the triggering factor and the outcome (induction validity) and between the effects of the treatments on the two organisms (remission validity). The relevance of this framework is then discussed regarding various animal models of depression

Comparisons with observations by the LASCO and Metis coronagraphs confirm the presence of morphological similarities and differences between the broadband visible light and EUV emissions, as documented on the basis of prior eclipse and space-based observations. Conclusions. The very-wide-field observations of FSI out to about 3 and 7 R, without and with the occulting disk, respectively, are paving the way for future dedicated instruments

    Bonsai Trees in Your Head: How the Pavlovian System Sculpts Goal-Directed Choices by Pruning Decision Trees

    When planning a series of actions, it is usually infeasible to consider all potential future sequences; instead, one must prune the decision tree. Provably optimal pruning is, however, still computationally ruinous and the specific approximations humans employ remain unknown. We designed a new sequential reinforcement-based task and showed that human subjects adopted a simple pruning strategy: during mental evaluation of a sequence of choices, they curtailed any further evaluation of a sequence as soon as they encountered a large loss. This pruning strategy was Pavlovian: it was reflexively evoked by large losses and persisted even when overwhelmingly counterproductive. It was also evident above and beyond loss aversion. We found that the tendency towards Pavlovian pruning was selectively predicted by the degree to which subjects exhibited sub-clinical mood disturbance, in accordance with theories that ascribe Pavlovian behavioural inhibition, via serotonin, a role in mood disorders. We conclude that Pavlovian behavioural inhibition shapes highly flexible, goal-directed choices in a manner that may be important for theories of decision-making in mood disorders
