    The Environmental Biorefinery: Using Microalgae to Remediate Wastewater, a Win-Win Paradigm

    Microalgae have been shown to be a source of multiple bio-based products ranging from high value molecules to commodities. Along with their potential to produce a large variety of products, microalgae can also be used for the depollution of wastewaters of different origins (urban, industrial, and agricultural). This paper is focused on the importance of harnessing the bioremediation capacity of microalgae to treat wastewaters in order to develop the microalgae industry (especially the microalgae biofuel industry) and to find other alternatives to the classic wastewater treatment processes. The current research on the potential of microalgae to treat a specific wastewater or a targeted pollutant is reviewed and discussed. Then, both strategies of selecting the best microalgae strain to treat a specific wastewater or pollutant and using a natural or an artificial consortium to perform the treatment will be detailed. The process options for treating wastewaters using microalgae will be discussed up to the final valorization of the biomass. The last part is dedicated to the challenges which research need to address in order to develop the potential of microalgae to treat wastewaters

    Exploring the microbiome of the “star” freshwater diatom Asterionella formosa in a laboratory context

    International audienceMost of our knowledge on the mechanisms underlying diatom-bacterial interactions has been acquired through studies involving isolation of culturable partners. Here, we established a laboratory model of intermediate complexity between complex natural communities and laboratory pure culture models. We investigated the whole community formed by the freshwater diatom Asterionella formosa and its associated bacteria in a laboratory context, including both culturable and unculturable bacteria. Combining cellular and molecular approaches, we showed that in laboratory cultures, A. formosa microbiome was dynamic and comprised of numerous bacterial species (mainly Proteobacteria and Bacteroidetes). Using metagenomics, we explored several metabolic potentials present within the bacterial community. Our analyses suggested that bacteria were heterotrophic although a third of them (Alpha- and Beta-proteobacteria) could also be phototrophic. About 60% of the bacteria, phylogenetically diverse, could metabolize glycolate. The capacity to synthesize molecules such as B vitamins appeared unevenly distributed among bacteria. Altogether, our results brought insights into the bacterial diversity found in diatom-bacterial communities and hinted at metabolic interdependencies within the community that could result in diatom-bacterial and bacterial-bacterial interactions. The present work allowed us to explore the functional architecture of the bacterial community associated with A. formosa in culture and is complementary to field studies.     Microalgal pigments: A source of natural food colors

    Naturally sourced colorants and dyes are currently gaining demand over synthetic alternatives due to an increase in consumer awareness brought forward by health and environmental issues. Microalgae are unicellular organisms which are microscopic in size and represent major photosynthesizers with the ability to efficiently convert available solar energy to chemical energy. Due to their distinct advantages over terrestrial plants such as faster growth rates, ability to grow on non-arable land, and diversity in the production of various natural bioactive compounds (e.g., lipids, proteins, carbohydrate, and pigments), microalgae are currently gaining promise as a sustainable source for the production of natural food-grade colorants. The versatility of microalgae to produce various pigments (e.g., chlorophylls, carotenoids, xanthophylls, and phycobiliproteins) that can be commercially exploited as a source of natural colorant is there to be explored. Various growth factors such as temperature, pH, salinity, and light in terms of both quality and quantity have been shown to significantly impact pigment production. In this chapter, we comprehensively review the characteristics of microalgal pigments and factors that affect pigment production in microalgae while evaluating the overall feasibility of exploiting them as a natural source of food colorants

    Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)

    In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide.
