    Myocardin immunohistochemistry index is associated with clinical prognosis in nasopharyngeal carcinoma: a clinical practice-based cohort study

    Purpose: Recent findings have implicated the role of myocardin re-expression in carcinogenesisHowever, the clinical functions of myocardin in nasopharyngeal carcinoma (NPC) is not known yet. The purpose for the cohort research was to investigate whether myocardin re-expression level may predict clinical prognosis in NPC patients.Methods: 148 NPC patients were recruited from September, 2005 to September, 2011 with median follow-up time of 4.5 years in a clinical practice setting. At study entry myocardin re-expression of these patients was determined using immunohistochemistry (IHC) and additional 20 normal nasopharyngeal tissues were included as control. Two-sample t-test was used to compare mean myocardin reexpression levels and Chi-square test was used for comparing tumour recurrence rate. Logistic regression analysis was used for tumour local control rate, and log-rank test, Kaplan-Meier estimates and Cox proportional hazard model for disease-free survival and overall survival.Results: Myocardin IHC index was significantly downregulated in NPC samples than in normalnasopharyngeal tissues (mean ± standard deviation, 61.2 ±31.5 vs. 109.9 ±73.6, P= 0.009). However, among NPC patients was observed a roughly V-shaped change of myocardin IHC index according to Tumour T-stage (P=0.067); meanwhile higher IHC level was associated with more tumour recurrence rate in NPC patients (High vs. Low: 21.6% vs. 8.1%; P=0.021). Logistic regression analysis equally showed high myocardin IHC level was an independent risk factor for local tumour control rate regardless of adjustments [High vs. Low: unadjusted Odds Ratio (OR) 0.320, 95% confidence interval (CI): 0.117 to 0.871; P=0.026]. Moreover, higher myocardin IHC level was associated with a marginal but not significant risk increase of disease-free survival [High vs. Low: adjusted Hazard Ratio (HR) 1.760, 95% CI: 0.981 to 3.158; log-rank: P=0.129]. A less obvious trend was observed with regard to overall survival [adjusted HR 1.409, 95% CI: 0.715 to 2.77; log-rank: P=0.745].Conclusion: The study results suggested that high myocardin IHC index level could be a potential clinically prognostic intermediate biomarker for tumour recurrence for NPC patients in routine practice. Large well-designed cohort studies involving IHC re-expression change over time is needed

    Probiotic Therapy for Treating Behavioral and Gastrointestinal Symptoms in Autism Spectrum Disorder: A Systematic Review of Clinical Trials

    The therapeutic potentials of probiotics in autism spectrum disorder (ASD) remains controversial, with the only existing systematic review on this topic published in 2015. Results from new trials have become available in recent years. We therefore conducted an updated systematic review, to assess the efficacy of probiotics in relieving behavioral symptoms of ASD and gastrointestinal comorbidities. Our review includes two randomized controlled trials, which showed improvement of ASD behaviors, and three open trials, all which exhibited a trend of improvement. Four of these trials concluded from subjective measures that gastrointestinal function indices showed a trend of improvement with probiotic therapy. Additional rigorous trials are needed to evaluate the effects of probiotic supplements in ASD

    Analysis of intravitreal injection of triamcinolone acetonide before vitrectomy for retinal detachment associated with choroidal detachment

    AIM:To observe the clinical effects of intravitreal injection of triamcinolone acetonide(TA)before vitrectomy for retinal detachment associated with choroidal detachment.METHODS: Totally 23 cases(23 eyes)of retinal detachment associated with choroidal detachment in our hospital were treated by intravitreal injection of TA 4-5d before 23-Gauge micro-invasive vitrectomy combined with silicone oil injection. All the cases were followed up between 6 to 9mo. The anatomic retinal reattachment, visual acuity, intraocular pressure and postoperative complications were observed and analyzed. RESULTS: After the surgery, the visual acuity of all patients were improved, with 9 eyes better than 0.3(39%), and 18 eyes better than 0.05(78%). The BCVA at 1wk, 1 and 3mo and last follow up were different compared with before operations(PPPCONCLUSION: Intravitreal injection of TA before vitrectomy for retinal detachment associated with choroidal could improve the clinical effects, and decrease the difficulty of surgery while the injection itself is pretty safe

    MicroRNA-148b is frequently down-regulated in gastric cancer and acts as a tumor suppressor by inhibiting cell proliferation

    <p>Abstract</p> <p>Background</p> <p>MicroRNAs (miRNAs) are involved in cancer development and progression, acting as tumor suppressors or oncogenes. Our previous studies have revealed that miR-148a and miR-152 are significantly down-regulated in gastrointestinal cancers. Interestingly, miR-148b has the same "seed sequences" as miR-148a and miR-152. Although aberrant expression of miR-148b has been observed in several types of cancer, its pathophysiologic role and relevance to tumorigenesis are still largely unknown. The purpose of this study was to elucidate the molecular mechanisms by which miR-148b acts as a tumor suppressor in gastric cancer.</p> <p>Results</p> <p>We showed significant down-regulation of miR-148b in 106 gastric cancer tissues and four gastric cancer cell lines, compared with their non-tumor counterparts by real-time RT-PCR. <it>In situ </it>hybridization of ten cases confirmed an overt decrease in the level of miR-148b in gastric cancer tissues. Moreover, the expression of miR-148b was demonstrated to be associated with tumor size (P = 0.027) by a Mann-Whitney U test. We also found that miR-148b could inhibit cell proliferation <it>in vitro </it>by MTT assay, growth curves and an anchorage-independent growth assay in MGC-803, SGC-7901, BGC-823 and AGS cells. An experiment in nude mice revealed that miR-148b could suppress tumorigenicity <it>in vivo</it>. Using a luciferase activity assay and western blot, CCKBR was identified as a target of miR-148b in cells. Moreover, an obvious inverse correlation was observed between the expression of CCKBR protein and miR-148b in 49 pairs of tissues (P = 0.002, Spearman's correlation).</p> <p>Conclusions</p> <p>These findings provide important evidence that miR-148b targets CCKBR and is significant in suppressing gastric cancer cell growth. Maybe miR-148b would become a potential biomarker and therapeutic target against gastric cancer.</p

Прикладний інтерес викликаний їх здатністю поєднувати у собі властивості магнітом’яких і магнітотвердих матеріалів, які суттєво залежать від структури і фазового складуз разків.У даній роботі, в якості досліджуваних магнітних структур, було використано тришарові плівкові системи Ni(d) / Au(15 нм) / Ni(25 нм) отримані шляхом пошарової конденсації, де d – товщина верхнього шару Ni, яка змінювалась від 5 до 15 нм
