Tématem diplomové práce je Analýza DPH ve vybrané obci. Cílem diplomové práce je analýza DPH v podmínkách obce Velké Heraltice a zhodnocení, zda by mohla obec zrušit registraci k DPH. V teoretické části jsou definovány základní pojmy problematiky DPH. Dále jsou vymezena zdanitelná plnění, osvobozená plnění, plnění s nárokem na odpočet daně a plnění v režimu přenesení daňové povinnosti. Také je zde popsáno kontrolní hlášení a daňové přiznání. Teoretická část také charakterizuje obec, její orgány a rozpočtovou skladbu. V praktické části je uvedena charakteristika vybrané obce a analýza jednotlivých zdanitelných plnění obce. Dále se práce zabývá uplatněním nároku na odpočet daně v poměrné a krácené výši, zkoumá režim přenesení daňové povinnosti u obce a analyzuje kontrolní hlášení. V závěru diplomové práce je uvedeno, zda obec může nebo nemůže zrušit registraci plátcovství DPH.The topic of thesis is the Analysis of VAT in selected municipality. The thesis aim is the analysis of VAT in terms of Velké Heraltice and assessing whether the municipality could cancel the VAT registration. The theoretical part defines the basic concepts of VAT issues. Furthermore, there are defined taxable transactions, exempt transactions, transactions with a right to deduction and reverse charge. There is also check report and a tax return. The theoretical part also characterizes the municipality, its institutions and the budget structure. The practical part describes the characteristics of the selected municipality and the analysis of individual taxable municipalities. Further, the thesis deals with the application of the right to deduction in proportional and reduced amount, examines the reverse charge regime at the municipality and analyzes the control report. In the conclusion of the thesis is stated whether the municipality can or can not cancel registration of the VAT payer.117 - Katedra účetnictví a danívýborn

The Role of Mitochondrial NADPH-Dependent Isocitrate Dehydrogenase in Cancer Cells

Isocitrate dehydrogenase 2 (IDH2) is located in the mitochondrial matrix. IDH2 acts in the forward Krebs cycle as an NADP+-consuming enzyme, providing NADPH for maintenance of the reduced glutathione and peroxiredoxin systems and for self-maintenance by reactivation of cystine-inactivated IDH2 by glutaredoxin 2. In highly respiring cells, the resulting NAD+ accumulation then induces sirtuin-3-mediated activating IDH2 deacetylation, thus increasing its protective function. Reductive carboxylation of 2-oxoglutarate by IDH2 (in the reverse Krebs cycle direction), which consumes NADPH, may follow glutaminolysis of glutamine to 2-oxoglutarate in cancer cells. When the reverse aconitase reaction and citrate efflux are added, this overall “anoxic” glutaminolysis mode may help highly malignant tumors survive aglycemia during hypoxia. Intermittent glycolysis would hypothetically be required to provide ATP. When oxidative phosphorylation is dormant, this mode causes substantial oxidative stress. Arg172 mutants of human IDH2—frequently found with similar mutants of cytosolic IDH1 in grade 2 and 3 gliomas, secondary glioblastomas, and acute myeloid leukemia—catalyze reductive carboxylation of 2-oxoglutarate and reduction to D-2-hydroxyglutarate, which strengthens the neoplastic phenotype by competitive inhibition of histone demethylation and 5-methylcytosine hydroxylation, leading to genome-wide histone and DNA methylation alternations. AMNESTY AND PARDON UNDER THE SLOVAK LAW AND THEIR UNIQUE STORY

The article analyses a unique phenomenon in the Slovak society - amnesty that has been abolished nearly twenty years later after it was granted and this procedure was confirmed by the highest authority that oversees constitutionality - the Constitutional Court of the Slovak Republic. The authors focus on the analysis of this phenomenon from a comparative law perspective and from the perspective of national constitutional and criminal law. An interesting and sadly amusing case when the amnesty granted by the deputy president was abolished twenty years later is apparently unique worldwide. The uniqueness of this case lies in several aspects which deserve a more detailed legal analysis. There is a limited number of cases worldwide when amnesties were abolished but these cases have always concerned either an amnesty related to a war conflict genocide or an amnesty related to a mass destruction of people for political reasons. This was not the case in Slovakia. In 1998, Vladimir Mečiar, holding the office of the prime minister and, at the same time, the deputy president, firstly granted amnesty in connection with the dismissal of the referendum on the direct election of the president and, in particular, in connection with the kidnapping of the son of the former President Michal Kovac. The case also featured an individual pardon granted by President Michal Kováč to his son on suspicion of economic crime. This is just the beginning of the story with very serious and far-reaching legal consequences

Ownership of land and its transfer from the perspective of tax legislation

Vlastnictví pozemků a jejich převody z pohledu daňových předpisů Abstrakt Předkládaná diplomová práce se zabývá vlastnictvím pozemků a jejich převody z pohledu daňových předpisů. Cílem této diplomové práce je podat přehled o daňových povinnostech, které souvisí s vlastnictvím pozemků a úplatnými převody vlastnictví k pozemkům na území České republiky a následně se zamyslet nad vhodností aktuální právní úpravy a jejími možnými změnami. Problematika daňových předpisů souvisejících s vlastnictvím pozemků a jejich převody je úzce provázána se soukromým právem. Úvodní část je proto věnována teoretickému úvodu do této problematiky, a to nejen z pohledu finančního, ale také občanského práva. V první části jsou konkrétně rozebírány pojmy nemovité věci a pozemku. Druhá část popisuje vlastnické právo včetně formy jeho převodu a krátce také problematiku katastru nemovitostí. V třetí části je obsažena obecná problematika daňového práva, vymezeny základní pojmy a charakteristika daňové soustavy. Čtvrtá část je věnována daním, jež zatěžují vlastnictví nemovitých věcí, a to zejména dani z pozemků. Rozebírány jsou konstrukční prvky daně z pozemků a obecně také správa daně z nemovitých věcí. Závěrem se autor práce zabývá opodstatněním daně a jejími možnými změnami. The aim of the thesis is to describe tax obligations connected with the ownership of land and with the transfers of ownership of land in the Czech Republic and further to consider the appropriateness of current legislation and its potential changes. The issue of tax regulations related to the ownership of land and its transfer is closely linked to the civil law. Introductory part of the thesis is therefore devoted to the theoretical introduction of the issue with respect to both financial law and civil law. The first chapter defines the term real estate and land. The second chapter describes and overviews forms of transfer of ownership of real estate and shortly also the issue of real estate cadaster. The fourth and fifth parts are the crucial ones with respect to the whole thesis. The fourth part overviews the responsibilities relating to the ownership of real estate particularly with regard to the land tax. Závěrečná práce je zpracována pro administrativní areál v Praze. Jedná se o přípravu a organizaci výstavby objektu v Hostivaři. V práci je řešeno zařízení staveniště na nejdůležitější technologické etapy, ekonomické srovnání jeřábů, finanční a časové zhodnocení stavby, výběr hlavních mechanismů a textová část zabývající se charakterem stavby, jejími technickými údaji, postupem výstavby objektu a celkovým pojednáním o budově. Technologický předpis je zpracován pro technologickou etapu výstavby montovaného skeletu.Final work is processed for an administrative center in Prague. It is the preparation and organization of the construction of the object in Hostivař. The work is done for the most important technological phases of site equipment, economic comparison of cranes, financial and time evaluation of building, selection of main mechanisms and main part of the text dealing with the character of the building, its technical data, process of constructing the object and the overall treatise about building. Technological process is made for the technological phase of construction of assembled skeleton.

V teoretické části je popsána charakteristika značky Apple a také teoretická východiska řízení značky. V praktické části jsou poté zpracovány dva výzkumy. Tato část je rozdělena na přípravnou fázi a realizační fázi. Je využito hloubkového individuálního rozhovoru a dotazníku. Základní soubor tvořili uživatelé značky Apple. Následující kapitola je věnována analýze výsledků výzkumu a jejich vyhodnocení. Primárním cílem této práce bylo zkoumat věrnost zákazníků značky Apple na trhu. Bylo zjištěno, že uživatelé značky jsou velice věrnými zákazníky, kteří značku Apple využívají víceméně ve všech oblastech, které jim Apple nabízí.This bachelor thesis deals with customer loyalty analysis of the Apple brand. The thesis is divided into theoretical and practical part. The theoretical part describes the characteristic of the Apple brand and theoretical bases of brand management. In the practical part are then processed two researches. This part is divided into preparatory and implementation phase. It is used in-depth individual interview and questionnaire survey. The basic group consisted of users of the Apple brand. The following chapter is devoted to analysing the results of research and evaluation. The primary objective of this study was to discover the customer loyalty of Apple brand on the market. Comparision of leachability of heavy metals from various types of alkali-activated alumino-silicate systems

Práce se zabývá stanovením a porovnáním vyluhovatelnosti těžkých kovů z různých typů alkalicky aktivovaných alumino-silikátových systémů (vysokoteplotního popílku, vysokopecní strusky). Tyto alkalicky aktivované materiály jsou schopné ve své struktuře účinně zapouzdřit těžké kovy, toxické látky nebo jiné kontaminanty, což může být přínosné pro životní prostředí. Náplní této práce je získat směs s přijatelnými mechanickými vlastnostmi, která by mohla být vhodnou pro inhibici těchto látek. Vyluhovatelnost byla testována dle normy ČSN EN – 12457-4 s následnou analýzou za použití ICP-OES. Struktura matrice byla zkoumána za použití metody SEM s detektorem EDS.The work deals with the determination and comparison of leaching of heavy metals from various types of alkali-activated alumino-silicate systems (high-temperature fly ash, blast furnace slag). Such alkali activated materials are capable in their structure effectively encapsulate heavy metals, toxic chemicals or other contaminants, which can be beneficial for the environment. The aim of this work is to obtain a mixture with acceptable mechanical properties that could be useful for the inhibition of these substances. Leachability was tested according to norm ČSN EN - 12457-4, followed by analysis by ICP-OES. The structure of the matrix was investigated using SEM equipped with EDS.

Nová tvář buněčné bioenergetiky

Cancer cells generally present abnormal bioenergetic properties including an elevated glucose uptake, a high glycolysis and a poorly efficient oxidative phosphorylation system. However, the determinants of cancer cells metabolic reprogramming remain unknown. The main question in this project was how environmental conditions in vivo can influence functioning of mitochondrial OXPHOS, because details of mitochondrial bioenergetics of cancer cells are poorly documented. We have combined two conditions, namely glucose and oxygen deprivation, to measure their potential interaction. We examined the impact of glucose deprivation and oxygen deprivation on cell survival, overall bioenergetics and OXPHOS protein expression. As a model, we have chosen a human breast carcinoma (HTB-126) and appropriate control (HTB-125) cultured cells, as large fraction of breast malignancies exhibit hypoxic tumor regions with low oxygen concentrations and poor glucose delivery. The results demonstrate that glucose presence or absence largely influence functioning of mitochochondrial oxidative phosphorylation. The level of mitochondrial respiration capacity is regulated by glucose; by Crabtree effect, by energy substrate channeling towards anabolic pathways that support cell growth and by mitochondrial biogenesis pathways. Both oxygen...Rakovinné buňky jsou charakteristické netypickými bioenergetickými vlastnostmi, jako je zvýšený metabolizmus glukózy a neefektivní oxidativní fosforylace. Přesný mechanizmus této metabolické proměny není doposud znám. Cílem tohoto projektu bylo ujasnit, jak prostředí tumoru ovlivňuje fungování mitochondriální oxidativní fosforylace. Zaměřovali jsme se na dvě základní podmínky, tj. deprivace glukózy a kyslíku, a analyzovali růst buněk, bioenergetické vlastnosti a expresi proteinů oxidativní fosforylace. Jako experimentální model jsme použili rakovinné buňky tumoru mléčné žlázy (HTB-126), které jsme porovnávali s příslušnou kontrolní linií (HTB-125), protože prostředí karcinomu prsu bývá často hypoxické a aglykemické. Výsledky této práce jasně ukazují závislost mitochondriální bioenergetiky na přítomnosti glukózy u rakovinných buněk. Mitochondriální respirace je regulována glukózou na několika úrovních, jako je Crabtree efekt, odvádění energetického substrátu z mitochondrií k podpoře buněčného růstu a proliferace a na úrovni biogeneze mitochondrií. Fungování oxidativní fosforylace je jasně ovlivněna přítomností glukózy i kyslíku. Součástí přizpůsobení se hypoxii je potlačení oxidativní fosforylace přítomností glukózy mnohem více, než v normoxii. Evaluation of the Leadership Style in a Selected Company

Diplomová práce je zaměřena na zhodnocení stylu vedení ve vybrané organizaci. Cílem práce je posoudit vhodnost stylů vedení v rámci různých oblastí organizace. Jsou zde brány v potaz, krom vlastností lídrů, také charakteristiky následovníků a struktura dané organizace. Diplomová práce se skládá z teoretické a praktické části. Teoretická část poukazuje na význam vedení lidí, objasňuje základní předpoklady lídra, vypovídá i o provázanosti leadershipu a managementu. Dále jsou zde objasněny koncepce leadershipu, efektivnost vedení a také metody posouzení vhodnosti. V poslední řadě je zde také popsána charakteristika organizace. Praktická část se pak skládá z několika částí, kde je využit Fiedlerův model, manažerská mřížka, analýza transakčního a transformačního vedení a analýza stylu vedení. U posledních dvou analýz byla využita metoda 360° zpětné vazba, aby došlo k objektivnímu posouzení. Výsledky praktické části byly zhodnocené a následně pak byly využity pro návrhy a doporučení pro jednotlivá oddělení.This diploma thesis is focused on the evaluation of the leadership style in a selected company. The main aim of the thesis is to assess a suitability of the leadership styles within different areas of the organization. The characteristics of the leaders and their followers are taken into account as well as the structure of the organization. The diploma thesis consists of theoretical and practical part. The theoretical part points to the importance of leadership, clarifies the basic assumptions of the leader, speaks about the interdependence of leadership and management. In addition, the leadership concepts, leadership efficiency, and suitability assessment methods are also explained. At the end of the theoretical part the organization`s characteristics are described. The practical part consists of several sections, where the Fiedler`s model, model of managerial grid, transactional and transformational leadership and analysis of the leadership styles were used. For the last two analyzes, the 360-degree feedback method was applied to make an objective assessment.
