    Modeling the effects of a Staphylococcal Enterotoxin B (SEB) on the apoptosis pathway

    BACKGROUND: The lack of detailed understanding of the mechanism of action of many biowarfare agents poses an immediate challenge to biodefense efforts. Many potential bioweapons have been shown to affect the cellular pathways controlling apoptosis [1-4]. For example, pathogen-produced exotoxins such as Staphylococcal Enterotoxin B (SEB) and Anthrax Lethal Factor (LF) have been shown to disrupt the Fas-mediated apoptotic pathway [2,4]. To evaluate how these agents affect these pathways it is first necessary to understand the dynamics of a normally functioning apoptosis network. This can then serve as a baseline against which a pathogen perturbed system can be compared. Such comparisons can expose both the proteins most susceptible to alteration by the agent as well as the most critical reaction rates to better instill control on a biological network. RESULTS: We explore this through the modeling and simulation of the Fas-mediated apoptotic pathway under normal and SEB influenced conditions. We stimulated human Jurkat cells with an anti-Fas antibody in the presence and absence of SEB and determined the relative levels of seven proteins involved in the core pathway at five time points following exposure. These levels were used to impute relative rate constants and build a quantitative model consisting of a series of ordinary differential equations (ODEs) that simulate the network under both normal and pathogen-influenced conditions. Experimental results show that cells exposed to SEB exhibit an increase in the rate of executioner caspase expression (and subsequently apoptosis) of 1 hour 43 minutes (± 14 minutes), as compared to cells undergoing normal cell death. CONCLUSION: Our model accurately reflects these results and reveals intervention points that can be altered to restore SEB-influenced system dynamics back to levels within the range of normal conditions

    INTRODUCTION: Cabotegravir and rilpivirine (CAB+RPV long-acting (LA)) is recommended as a treatment for HIV-1 allowing people living with HIV to receive 2 monthly injectable treatment, rather than daily pills. Providing injectable therapy in a system designed to provide and manage study participants on oral treatments poses logistical challenges namely how resources are used to accommodate patient preference within constrained health economies with capacity limitations. In this pragmatic multicentre study, we aim to understand the implementation of CAB-RPV-LA administration in two settings via mixed methods to explore perspectives of participants and the clinical team delivering CAB+RPV LA. METHODS AND ANALYSIS: Women, racially minoritised people and older people are chronically under-represented in HIV clinical trials so the ILANA trial has set recruitment caps to ensure recruitment of 50% women, 50% ethnically diverse people and 30% over 50 years of age to include a more representative study population. Using a mixed-methods approach, the primary objective is to identify and evaluate the critical implementation strategies for CAB+RPV LA in both hospital and community settings. Secondary objectives include evaluating feasibility and acceptability of CAB+RPV LA administration at UK clinics and community settings from the perspective of HIV care providers, nurses and representatives at community sites, evaluating barriers to implementation, the utility of implementation strategies and adherence. ETHICS AND DISSEMINATION: Ethical approval has been obtained from the Health Research Authority Research Ethics Committee (REC reference: 22/PR/0318). The dissemination strategy has been formulated with the SHARE Collaborative Community Advisory Board to maximise the impact of this work on clinical care and policy. This strategy draws on and leverages existing resources within the participating organisations, such as their academic infrastructure, professional relationships and community networks.     Quantitative trait loci for bone traits segregating independently of those for growth in an F-2 broiler X layer cross

    An F broiler-layer cross was phenotyped for 18 skeletal traits at 6, 7 and 9 weeks of age and genotyped with 120 microsatellite markers. Interval mapping identified 61 suggestive and significant QTL on 16 of the 25 linkage groups for 16 traits. Thirty-six additional QTL were identified when the assumption that QTL were fixed in the grandparent lines was relaxed. QTL with large effects on the lengths of the tarsometatarsus, tibia and femur, and the weights of the tibia and femur were identified on GGA4 between 217 and 249 cM. Six QTL for skeletal traits were identified that did not co-locate with genome wide significant QTL for body weight and two body weight QTL did not coincide with skeletal trait QTL. Significant evidence of imprinting was found in ten of the QTL and QTL x sex interactions were identified for 22 traits. Six alleles from the broiler line for weight- and size-related skeletal QTL were positive. Negative alleles for bone quality traits such as tibial dyschondroplasia, leg bowing and tibia twisting generally originated from the layer line suggesting that the allele inherited from the broiler is more protective than the allele originating from the layer

    Geochemical proxies of North American freshwater routing during the Younger Dryas cold event

The cause of this event, which has long been viewed as the canonical example of abrupt climate change, was initially attributed to the routing of freshwater to the St. Lawrence River with an attendant reduction in Atlantic meridional overturning circulation. However, this mechanism has recently been questioned because current proxies and dating techniques have been unable to confirm that eastward routing with an increase in freshwater flux occurred during the Younger Dryas. Here we use new geochemical proxies (ΔMg/Ca, U/Ca & 87Sr/86Sr) measured in planktonic foraminifera at the mouth of the St. Lawrence Estuary as tracers of freshwater sources to further evaluate this question. Our proxies, combined with planktonic δ18Oseawater and δ13C, confirm that routing of runoff from western Canada to the St. Lawrence River occurred at the start of the Younger Dryas, with an attendant increase in freshwater flux of 0.06 ± 0.02 Sverdrup (1 Sverdrup (Sv) = 106 m3 s-1). This base discharge increase is sufficient to have reduced Atlantic meridional overturning circulation and caused the Younger Dryas cold interval. In addition, our data indicate subsequent fluctuations in the freshwater flux to the St. Lawrence River of ~0.06 to 0.12 Sv, thus explaining the variability in the overturning circulation and climate during the Younger Dryas.This research was funded by the NSF Paleoclimate Program (P.U.C.) and the NSF (G.P.K.)

    Exploratory factor analysis of self-reported symptoms in a large, population-based military cohort

    <p>Abstract</p> <p>Background</p> <p>US military engagements have consistently raised concern over the array of health outcomes experienced by service members postdeployment. Exploratory factor analysis has been used in studies of 1991 Gulf War-related illnesses, and may increase understanding of symptoms and health outcomes associated with current military conflicts in Iraq and Afghanistan. The objective of this study was to use exploratory factor analysis to describe the correlations among numerous physical and psychological symptoms in terms of a smaller number of unobserved variables or factors.</p> <p>Methods</p> <p>The Millennium Cohort Study collects extensive self-reported health data from a large, population-based military cohort, providing a unique opportunity to investigate the interrelationships of numerous physical and psychological symptoms among US military personnel. This study used data from the Millennium Cohort Study, a large, population-based military cohort. Exploratory factor analysis was used to examine the covariance structure of symptoms reported by approximately 50,000 cohort members during 2004-2006. Analyses incorporated 89 symptoms, including responses to several validated instruments embedded in the questionnaire. Techniques accommodated the categorical and sometimes incomplete nature of the survey data.</p> <p>Results</p> <p>A 14-factor model accounted for 60 percent of the total variance in symptoms data and included factors related to several physical, psychological, and behavioral constructs. A notable finding was that many factors appeared to load in accordance with symptom co-location within the survey instrument, highlighting the difficulty in disassociating the effects of question content, location, and response format on factor structure.</p> <p>Conclusions</p> <p>This study demonstrates the potential strengths and weaknesses of exploratory factor analysis to heighten understanding of the complex associations among symptoms. Further research is needed to investigate the relationship between factor analytic results and survey structure, as well as to assess the relationship between factor scores and key exposure variables.</p

    The first legal mortgagor: a consumer without adequate protection?

    This article contends that the UK government’s attempt to create a well-functioning consumer credit market will be undermined if it fails to reform the private law framework relating to the first legal mortgage. Such agreements are governed by two distinct regulatory regimes that are founded upon very different conceptions of the mortgagor. The first, the regulation of financial services overseen by the Financial Conduct Authority, derives from public law and is founded upon a conception of the mortgagor as “consumer”. The other is land law, private law regulation implemented by the judiciary and underpinned by a conception of the mortgagor as “landowner”. Evidence suggests that the operation of these two regimes prevents mortgagors from receiving fair and consistent treatment. The current reform of financial services regulation therefore will change only one part of this governance regime and will leave mortgagors heavily reliant upon a regulator that still has to prove itself. What this article argues is that reform of the rules of private law must also be undertaken with the aim of initiating a paradigm shift in the conception of the mortgagor from “landowner” to “consumer”. Cultural shifts of this kind take time but the hope is that this conceptual transformation will occur in time to deter the predicted rise in mortgage possessions

    Spitzer IRAC Images and Sample Spectra of Cassiopeia A's Explosion

    We present Spitzer IRAC images, along with representative 5.27 to 38.5 micron IRS spectra of the Cassiopeia A supernova remnant. We find that various IRAC channels are each most sensitive to a different spectral and physical component. Channel 1 (3.6 micron) matches radio synchrotron images. Where Channel 1 is strong with respect to the other channels, the longer-wavelength spectra show a broad continuum gently peaking around 26 micron, with weak or no lines. We suggest that this is due to un-enriched progenitor circumstellar dust behind the outer shock, processed by shock photons and electrons. Where Channel 4 (8 micron) is bright relative to the other IRAC channels, the long-wavelength spectra show a strong, 2-3 micron-wide peak at 21 micron, likely due to silicates and proto-silicates, as well as strong ionic lines of [Ar II], [Ar III], [S IV] and [Ne II]. In these locations, the dust and ionic emission originate from the explosion's O-burning layers. The regions where Channels 2 (4.5 micron) and 3 (5.6 micron) are strongest relative to Channel 4 show a spectrum that rises gradually to 21 micron, and then flattens or rises more slowly to longer wavelengths, along with higher ratios of [Ne II] to [Ar II]. Dust and ionic emission in these locations arise primarily from the C- and Ne- burning layers. These findings are consistent with asymmetries in the explosion producing variations in the velocity structure in different directions, but preserving the nucleosynthetic layers. At each location, the dust and ionic lines in the mid-infrared, and the hotter and more highly ionized optical and X-ray emission are then dominated by the layer currently encountering the reverse shock in that direction.Comment: 28 pages, 10 figures, accepted in to the Astrophysical Journal.
