    RosettaBackrub--a web server for flexible backbone protein structure modeling and design.

    The RosettaBackrub server (http://kortemmelab.ucsf.edu/backrub) implements the Backrub method, derived from observations of alternative conformations in high-resolution protein crystal structures, for flexible backbone protein modeling. Backrub modeling is applied to three related applications using the Rosetta program for structure prediction and design: (I) modeling of structures of point mutations, (II) generating protein conformational ensembles and designing sequences consistent with these conformations and (III) predicting tolerated sequences at protein-protein interfaces. The three protocols have been validated on experimental data. Starting from a user-provided single input protein structure in PDB format, the server generates near-native conformational ensembles. The predicted conformations and sequences can be used for different applications, such as to guide mutagenesis experiments, for ensemble-docking approaches or to generate sequence libraries for protein design

    Ground-based and in-flight simulator studies of low-speed handling characteristics of two supersonic cruise transport concepts

    Conventional and powered lift concepts for supersonic approach and landing tasks are considered. Results indicated that the transport concepts had unacceptable low-speed handling qualities with no augmentation, and that in order to achieve satisfactory handling qualities, considerable augmentation was required. The available roll-control power was acceptable for the powered-lift concept

    Tinigible Body Macrophages in Regulation of Germinal Center Reactions

    Tingible body macrophages (TBM), long thought simply as scavengers of apoptotic lymphocytes, are located in the unique microenvironment of germinal centers in close proximity to antigen-retaining follicular dendritic cells (FDC). Observations that TBM endocytose FDC-iccosomal (immune-complex coated bodies) antigen suggested that TBM might present this antigen and help regulate the germinal center reaction. To test for antigen presentation, the ovalbumin (OVA)-specific TH hybridoma, 3DO-54.8, which produces IL-2 on receiving effective presentation of OVA, were used as responders to OVA-bearing TBM. Results showed that OVA-bearing TBM failed to induce IL-2 production. Furthermore, addition of TBM to IL- 2-inducing positive controls (B cells) not only failed to augment IL-2 production, but rather TBM significantly (55-90%) reduced B-cell induction of IL-2. We found that TBM were rich in prostaglandin by comparison with other nongerminal center lymph node macrophages and that addition of indomethacin to the cultures reversed the inhibitory effect of TBM. Depletion of TBM from enriched preparations, prior to addition to positive control cultures, also abrogated the inhibitory effect on IL-2 production. These data support the concept that TBM, within the unique microenvironment of germinal centers, may be specialized to downregulate the germinal center reaction

    Refrigerants with Low Environmental Impact for Refrigeration Applications

    New refrigerants with considerable low environmental impact are currently under evaluation by the refrigeration industry. Such refrigerants could be used in place of high global warming fluids like R404A. Among these new fluids are 1234ze, N-13, L-40 and N-40. Initial laboratory evaluations of these refrigerants showed good energy efficiency due to their good thermal properties. These initial evaluations were followed by larger scale (field) trials, refrigerant–oil property measurements, and also material compatibility. This study presents and discusses results of such evaluations, and extends the assessment to LCCP using different refrigeration technologies and refrigerants

    Predatory bacteria in the haemolymph of the cultured spiny lobster Panulirus ornatus

    Bdellovibrio and like organisms (BALOs) are Gram-negative obligate predators of other bacteria in a range of environments. The recent discovery of BALOs in the circulatory system of cultured spiny lobster P. ornatus warrants more investigation. We used a combination of co-culture agar and broth assays and transmission electron microscopy to show a Halobacteriovorax sp. strain Hbv preyed upon the model prey bacterium Vibrio sp. strain Vib. The haemolymph microbiome of juvenile P. ornatus was characterised following injection of phosphate buffered saline (control) or prey and/or predator bacteria for 3 d. The predator Hbv had no effect on survival compared to the control after 3 d. However, when compared to the prey only treatment group, lobsters injected with both prey and predator showed significantly lower abundance of genus Vibrio in the haemolymph bacterial community composition. This study indicates that predatory bacteria are not pathogenic and may assist in controlling microbial population growth in the haemolymph of lobsters

    Use of Blends in Commercial Refrigeration Systems: Fractionation characteristics and material compatibility of R448A

    The development of lower global warming potential (GWP) blends offers an alternative for the widely used high GWP fluids such as R404A. Commercial refrigeration systems for supermarkets are a common example for such systems. These kinds of systems show relatively large refrigerant leakage rates of 15% to 20% per year. The newly developed R404A lower GWP alternatives are zeotropic blends, meaning they show a temperature glide during phase change processes in the evaporator and condenser of the system. Because of the glide, there is a concern within end users that the blend components could separate, or fractionate, during a leak event. Â This paper will describe the results from tests carried out to determine the effect of refrigerant fractionation and its effect on the performance of an actual refrigeration system subject to multiple leak events. The second part of this paper provides characteristics for the solubility and miscibility of R448A with POE oil and results from a material compatibility study of R448A with common materials used in refrigeration equipment

    Water absorption and desorption in shuttle ablator and insulation materials

Water accumulation in these materials after a soak for 24 hours ranged from +1.1% for orbiter tile to +161% for solid rocket booster MSA-1.
