Recommendations for reducing energy consumption and improving air quality through energy efficiency in Indian Country: Working paper series--04-04

The strongest recommendation stemming from the Air Pollution Prevention forum of the Western Regional Air Partnership (WRAP) was for tribes to develop and implement energy plans. This paper provides a discussion of the benefits of any such plan and itemized a variety of components of any said plan. A brief discussion of the WRAP is presented is the framework of improving air quality. Based on the background of Jacobs (2000) and Smith (2000), the primary and secondary benefits of energy efficiency plans are then discussed. Then a series of specific recommendations are itemized. These include tribally sponsored programs, collaborative opportunities, and tribal leadership programs

Economic analysis of energy efficiency measures: Tribal case studies with The Confederated Salish and Kootenai Tribes of The Flathead Reservation, The Pascua Yaqui Tribe, and The Yurok Tribe: Working paper series--04-02

The best way to reduce air pollution from the production of electricity is not to produce it in the first place. This paper presents three cases studies explaining how energy efficiency programs can be beneficial to tribes from a purely cost-saving basis, The Yurok, Flathead and Pascua Yaqui Reservations graciously provided an opportunity to investigate energy efficiency for their tribes. In each case the consumption of electricity was reduced such that a positive net present value was present following an initial capital investment. In the case of the irrigation systems on the Flathead Reservation, the savings were substantial even though no explicit savings of water was accounted for. It is estimated that tribes can reduce electricity costs by 10-50% by following energy efficiency programs. Many programs can be very simple involving small upfront capital outlays, such as changing light bulbs or installing motion sensors. The Yurok case showed a return of 7.5% after simply changing bulbs and the Pasqua Yaqui case showed a possible return of 10-80%. Other tribes could make similar investments and realize similar benefits in addition to improved air quality. The strongest recommendation from the Air Pollution Prevention Forum to tribes was to develop an energy plan. These cases show that some fairly simple analysis can make a plan very beneficial

Stuart Hall and the theory and practice of articulation

In this article, I argue that the idea of articulation links three different dimensions of Stuart Hall’s work: it is central to the work of cultural politics, to the work of hegemony, and to his practice of embodied pedagogy. I claim that his approach to pedagogy entails the art of listening combined with the practice of theorising in the service of expanding who belongs to the public. This involves the work of translation, finding ways of addressing different audiences. I treat each of these aspects in turn, drawing out the salience of articulation for each and suggest that these three dimensions are themselves articulated by Hall’s commitment to the theory and practice of articulation

Genome-wide CRISPR interference screen identifies long non-coding RNA loci required for differentiation and pluripotency

Although many long non-coding RNAs (lncRNAs) exhibit lineage-specific expression, the vast majority remain functionally uncharacterized in the context of development. Here, we report the first described human embryonic stem cell (hESC) lines to repress (CRISPRi) or activate (CRISPRa) transcription during differentiation into all three germ layers, facilitating the modulation of lncRNA expression during early development. We performed an unbiased, genome-wide CRISPRi screen targeting thousands of lncRNA loci expressed during endoderm differentiation. While dozens of lncRNA loci were required for proper differentiation, most differentially expressed lncRNAs were not, supporting the necessity for functional screening instead of relying solely on gene expression analyses. In parallel, we developed a clustering approach to infer mechanisms of action of lncRNA hits based on a variety of genomic features. We subsequently identified and validated FOXD3-AS1 as a functional lncRNA essential for pluripotency and differentiation. Taken together, the cell lines and methodology described herein can be adapted to discover and characterize novel regulators of differentiation into any lineage

Economic immorality and social reformation in English popular preaching, 1585-1625

Popular preachers, often drawing crowds of hundreds, frequently attempted to reform the relationship between rich and poor in Elizabethan and Jacobean England. Rather than accepting economic oppression as part of the divinely-ordained social order, many tried to convince their audiences that the extortions of merchants, landlords and creditors were crimes which should be punished severely by England’s earthly authorities. This paper demonstrates how the language of popular homiletics opened up a space for plebeian action with concrete socioeconomic consequences. By analysing the connotative idiom of social complaint found in homilies and other widely-heard sermons, the important but historiographically neglected role of ‘godliness’ in the early modern ‘moral economy’ is revealed

Geobiology of the late Paleoproterozoic Duck Creek Formation, Western Australia

The ca. 1.8 Ga Duck Creek Formation, Western Australia, preserves 1000 m of carbonates and minor iron formation that accumulated along a late Paleoproterozoic ocean margin. Two upward-deepening stratigraphic packages are preserved, each characterized by peritidal precipitates at the base and iron formation and carbonate turbidites in its upper part. Consistent with recent studies of Neoarchean basins, carbon isotope ratios of Duck Creek carbonates show no evidence for a strong isotopic depth gradient, but carbonate minerals in iron formations can be markedly depleted in C-13. In contrast, oxygen isotopes covary strongly with depth; delta O-18 values as positive as 2%. VPDB in peritidal facies systematically decline to values of 6 to 16% in basinal rocks, reflecting, we posit, the timing of diagenetic closure. The Duck Creek Formation contains microfossils similar to those of the Gunflint Formation, Canada; they are restricted to early diagenetic cherts developed in basinal facies, strengthening the hypothesis that such fossils capture communities driven by iron metabolism. Indeed, X-ray diffraction data indicate that the Duck Creek basin was ferruginous throughout its history. The persistence of ferruginous waters and iron formation deposition in Western Australia for at least several tens of millions of years after the transition to sulfidic conditions in Laurentia suggests that the late Paleoproterozoic expansion of sulfidic subsurface waters was globally asynchronous

Translational pharmacology of an inhaled small molecule αvβ6 integrin inhibitor for idiopathic pulmonary fibrosis

The αvβ6 integrin plays a key role in the activation of transforming growth factor-β (TGFβ), a pro-fibrotic mediator that is pivotal to the development of idiopathic pulmonary fibrosis (IPF). We identified a selective small molecule αvβ6 RGD-mimetic, GSK3008348, and profiled it in a range of disease relevant pre-clinical systems. To understand the relationship between target engagement and inhibition of fibrosis, we measured pharmacodynamic and diseaserelated end points. Here we report, GSK3008348 binds to αvβ6 with high affinity in human IPF lung and reduces downstream pro-fibrotic TGFβ signaling to normal levels. In human lung epithelial cells, GSK3008348 induces rapid internalization and lysosomal degradation of the αvβ6 integrin. In the murine bleomycin-induced lung fibrosis model, GSK3008348 engages αvβ6, induces prolonged inhibition of TGFβ signaling and reduces lung collagen deposition and serum C3M, a marker of IPF disease progression. These studies highlight the potential of inhaled GSK3008348 as an anti-fibrotic therapy
