    Invasion of a Virulent Phytophthora infestans Genotype at the Landscape Level; Does Spatial Heterogeneity Matter?

    Proper landscape-scale deployment of disease resistant genotypes of agricultural crop species could make those crops less vulnerable to invasion by resistance breaking genotypes. Here we develop a multi-scale, spatiotemporal model of the potato late blight pathosystem to investigate spatial strategies for the deployment of host resistance. This model comprises a landscape generator, a potato late blight model, and a suite of aerobiological models, including an atmospheric dispersion model. Within individual growing regions, increasing the number of host genotypes caused the greatest reduction in epidemic extent, followed by reduction of the proportion of potato in the landscape, lowering the clustering of host fields, and reducing the size of host fields. Deployment of host resistance in genotype mixtures had a large effect on disease invasion. The use of space as an isolation barrier was effective in scenarios involving two distinct potato growing regions. It was possible to completely eliminate the risk of epidemic spread from one region to another using inter-regional separation distances ranging from 8 to 32 km. The overall efficacy of this strategy was highly dependent, however, on the degree of spatial mixing of potato genotypes within each region.     Regional spore dispersal as a factor in disease risk warnings for potato late blight: a proof of concept

    This study develops and tests novel approaches that significantly reduce the fungicide input necessary for potato late blight control while maintaining the required high level of disease control. The central premise is that fungicide inputs can be reduced by reducing dose rates on more resistant cultivars, by omitting applications on days when conditions are unsuitable for atmospheric transport of viable sporangia and by adapting the dose rate to the length of the predicted critical period. These concepts were implemented and tested in field experiments in 2007 and 2008 in the North Eastern potato growing region in the Netherlands which is known for its high potato late blight disease pressure. Field experiments contained three starch potato cultivars, representing a range in resistance to potato late blight from susceptible to highly resistant, and a series of decision rules determining spray timing and incorporating an increasing number of variables such as: remaining fungicide protection level, critical weather, atmospheric capacity for viable transport of sporangia and the length of the predicted critical period. The level of cultivar resistance was used to reduce the dose rate of the preventive fungicide Shirlan (a.i. fluazinam) by default. A 50% – 75% reduction of the fungicide input proved possible in both years without adverse consequences to the crop or yield.     Improvement in eating disorder risk and psychological health in people with class 3 obesity : effects of a multidisciplinary weight management program

    This study aimed to evaluate the risk of eating disorders, psychological distress, and healthrelated quality of life (HRQoL) in people with class 3 obesity (body mass index (BMI) ≥ 40 kg/m2 ), and the effect of multidisciplinary weight management over 12 months. This retrospective cohort study included all adults with class 3 obesity who enrolled in a weight management program from March 2018 to December 2019. Questionnaires included the Eating Disorder Examination Questionnaire Short (EDE-QS), Kessler Psychological Distress Scale (K10), and 36-Item Short Form Survey (SF-36) for HRQoL. Physical and Mental Component Summary scores (PCS and MCS) were derived from the SF-36. Of 169 participants who completed 12 months in the program, 65.7% (n = 111) completed questionnaires at baseline and 12 months, with 6.0 ± 6.8% weight loss over this period. Compared to baseline, there was significant improvement at 12 months in EDE-QS (15.7 ± 6.6 vs. 13.6 ± 6.2, p = 0.002), K10 (25.7 ± 9.7 vs. 21.2 ± 9.4, p < 0.001), PCS (29.4 ± 10.1 vs. 36.1 ± 10.9, p < 0.001), and MCS scores (40.2 ± 12.4 vs. 44.0 ± 13.4, p = 0.001). All, apart from EDE-QS scores, remained significant after adjusting for weight change. This study highlights the importance of multidisciplinary management in people with class 3 obesity to help reduce eating disorder risk and psychological distress, and improve HRQoL, in addition to weight loss

    Class 3 obesity in a multidisciplinary metabolic weight management program : the effect of preexisting Type 2 diabetes on 6-month weight loss

    Introduction. Class 3 obesity (BMI ≥ 40 kg/m2) is a growing health problem worldwide associated with considerable comorbidity including Type 2 diabetes mellitus (T2DM). The multidisciplinary medical management of obesity can be difficult in T2DM due to potential weight gain from medications including sulphonylureas and insulin. However, newer weight-neutral/losing diabetes medications can aid additional weight loss. The aim of this study was to compare weight loss outcomes of patients with and without T2DM, and in patients with T2DM, to compare diabetes outcomes and change in medications at 6 months. Methods. All patients entering a multidisciplinary weight management metabolic program in a publicly funded hospital clinic in Sydney between March 2018 and March 2019, with BMI ≥ 40 kg/m2 and aged ≥18 years were included. Data was collected from patient clinical and electronic notes at baseline and 6 months. Results. Of the 180 patients who entered the program, 53.3% had T2DM at baseline. There was no difference in percentage weight loss in those with or without T2DM (4:2±4:9% vs. 3:6±4:7%, p = 0:35). Additionally, T2DM patients benefited from a 0.47% reduction in HbA1c (p < 0:01) and a reduction in the number of medications from baseline to 6 months (1:8±1:0/patient vs. 1:0±1:2/patient, p < 0:001). T2DM patients who started on weigh-neutral/losing medications in the program lost more weight than those started on weight-gaining medications (7:7±5:3% vs. 2:4±3:8%, p = 0:015). Conclusions. Patients with class 3 obesity had significant weight loss at 6 months in this program. Patients with T2DM at baseline had comparable weight loss at 6 months, a significant improvement in glycaemic control, and a reduction in diabetes medication load. Additionally, patients with T2DM who were started on weight-neutral/losing medications lost significantly more weight than those started on weight-gaining medications, and these medications should be preferentially used in class 3 obesity and comorbid T2DM

    Multi-scale modelling of infection pressure from Phytophthora infestans

    Management of potato late blight could benefit from prediction of the risk posed to potato fields from external inoculum sources of Phytophthora infestans. Influx of inoculum depends on a complex interplay of population biological, atmospheric and spore survival processes, and is difficult to predict. This research aims at building tools for such prediction. BLIGHTSPACE is a spatio-temporal model (parameterized for potato late blight) that has been developed and utilized to study the progress of epidemics in individual fields and networks of fields. A quasi-Gaussian plume model was developed to provide long-range transport of spores within BLIGHTSPACE. Numerical results compared favorably with experimental data. A further submodel for the survival of spores during long-range transportation has been added. Integration of these three submodels will create an experimental arena for comparing control options for potato late blight

    If I Had My Way, I'd Have Been a Killer: Songwriting and its Motivations for Leisure and Work

    This paper builds on our earlier work (Henderson and Spracklen, 2014) which looked at folk musicians exercising agency in careers that can develop from casual leisure through serious leisure to work. In looking specifically at songwriting, it synthesises theories on the underlying motivations refelecting the view of Born (2010, p.171) who suggests that the ‘theory of cultural production requires reinvention’ citing key themes of ’aesthetics and the cultural object; agency and subjectivity; the place of institutions; history, temporality and change; and problems of value and judgement.’ It examines in a phenomenological way, through songwriter interviews, the cultural production of songs and the motivational factors within the songwriting process. Highlighting that this form of cultural production challenges the hierarchical view of Maslow’s needs and suggests the career arc proposed by Stebbins (1992, p. 68) is not a case of work simply following sequentially from serious and casual leisure

    T cell sensitivity to TGF-β is required for the effector function but not the generation of splenic CD8+ regulatory T cells induced via the injection of antigen into the anterior chamber

    The introduction of antigen into the anterior chamber (AC) of the eye induces the production of antigen-specific splenic CD8+ regulatory T cells (AC-SPL cells) that suppress a delayed-type hypersensitivity (DTH) reaction in immunized mice. Because the generation of these regulatory T cells is also induced by exposure to transforming growth factor (TGF)-β and antigen or F4/80+ cells exposed to TGF-β and antigen in vitro, we investigated (i) whether these cells are produced in dominant negative receptor for transforming growth factor β receptor type II (dnTGFβRII) or Cbl-b−/− mice whose T cells are resistant to TGF-β, (ii) whether DTH is suppressed by wild type (WT) CD8+ AC-SPL cells in Cbl-b−/− and dnTGFβRII mice and (iii) the effect of antibodies to TGF-β on the suppression of DTH by CD8+ AC-SPL cells. DnTGFβRII immunized and Cbl-b−/− mice produced splenic CD8+ regulatory cells after the intracameral injection of antigen and immunization. The suppression of a DTH reaction by CD8+ AC-SPL cells in WT mice was blocked by the local inclusion of antibodies to TGF-β when WT splenic CD8+ AC-SPL cells were injected into the DTH reaction site. Moreover, the DTH reaction in immunized dnTGFβRII and Cbl-b−/− mice was not suppressed by the transfer of WT CD8+ AC-SPL cells to the site challenged with antigen. In aggregate, these observations suggest that T cell sensitivity to TGF-β is not an obligate requirement for the in vivo induction of CD8+ AC-SPL T cells but the suppression of an in vivo DTH reaction by CD8+ AC-SPL cells is dependent on TGF-β

    Impacts of climate change on plant diseases – opinions and trends

    There has been a remarkable scientific output on the topic of how climate change is likely to affect plant diseases in the coming decades. This review addresses the need for review of this burgeoning literature by summarizing opinions of previous reviews and trends in recent studies on the impacts of climate change on plant health. Sudden Oak Death is used as an introductory case study: Californian forests could become even more susceptible to this emerging plant disease, if spring precipitations will be accompanied by warmer temperatures, although climate shifts may also affect the current synchronicity between host cambium activity and pathogen colonization rate. A summary of observed and predicted climate changes, as well as of direct effects of climate change on pathosystems, is provided. Prediction and management of climate change effects on plant health are complicated by indirect effects and the interactions with global change drivers. Uncertainty in models of plant disease development under climate change calls for a diversity of management strategies, from more participatory approaches to interdisciplinary science. Involvement of stakeholders and scientists from outside plant pathology shows the importance of trade-offs, for example in the land-sharing vs. sparing debate. Further research is needed on climate change and plant health in mountain, boreal, Mediterranean and tropical regions, with multiple climate change factors and scenarios (including our responses to it, e.g. the assisted migration of plants), in relation to endophytes, viruses and mycorrhiza, using long-term and large-scale datasets and considering various plant disease control methods
