    How far do they go? The outer structure of dark matter halos

    We study the density profiles of collapsed galaxy-size dark matter halos with masses 1e11-5e12 Msun focusing mostly on the halo outer regions from the formal virial radius Rvir up to 5-7Rvir. We find that isolated halos in this mass range extend well beyond Rvir exhibiting all properties of virialized objects up to 2-3Rvir: relatively smooth density profiles and no systematic infall velocities. The dark matter halos in this mass range do not grow as one naively may expect through a steady accretion of satellites, i.e., on average there is no mass infall. This is strikingly different from more massive halos, which have large infall velocities outside of the virial radius. We provide accurate fit for the density profile of these galaxy-size halos. For a wide range (0.01-2)Rvir of radii the halo density profiles are fit with the approximation rho=rho_s exp(-2n[x^{1/n}-1])+rho_m, where x=r/r_s, rho_m is the mean matter density of the Universe, and the index n is in the range n=6-7.5. These profiles do not show a sudden change of behavior beyond the virial radius. For larger radii we combine the statistics of the initial fluctuations with the spherical collapse model to obtain predictions for the mean and most probable density profiles for halos of several masses.     Riociguat for the treatment of chronic thromboembolic pulmonary hypertension.

    BACKGROUND: Riociguat, a member of a new class of compounds (soluble guanylate cyclase stimulators), has been shown in previous clinical studies to be beneficial in the treatment of chronic thromboembolic pulmonary hypertension. METHODS: In this phase 3, multicenter, randomized, double-blind, placebo-controlled study, we randomly assigned 261 patients with inoperable chronic thromboembolic pulmonary hypertension or persistent or recurrent pulmonary hypertension after pulmonary endarterectomy to receive placebo or riociguat. The primary end point was the change from baseline to the end of week 16 in the distance walked in 6 minutes. Secondary end points included changes from baseline in pulmonary vascular resistance, N-terminal pro-brain natriuretic peptide (NT-proBNP) level, World Health Organization (WHO) functional class, time to clinical worsening, Borg dyspnea score, quality-of-life variables, and safety. RESULTS: By week 16, the 6-minute walk distance had increased by a mean of 39 m in the riociguat group, as compared with a mean decrease of 6 m in the placebo group (least-squares mean difference, 46 m; 95% confidence interval [CI], 25 to 67; P<0.001). Pulmonary vascular resistance decreased by 226 dyn · sec · cm-5in the riociguat group and increased by 23 dyn · sec · cm-5in the placebo group (least-squares mean difference, -246 dyn · sec · cm-5; 95% CI, -303 to -190; P<0.001). Riociguat was also associated with significant improvements in the NT-proBNP level (P<0.001) and WHO functional class (P = 0.003). The most common serious adverse events were right ventricular failure (in 3% of patients in each group) and syncope (in 2% of the riociguat group and in 3% of the placebo group). CONCLUSIONS: Riociguat significantly improved exercise capacity and pulmonary vascular resistance in patients with chronic thromboembolic pulmonary hypertension. The transfer of pump noise by four-wave mixing was clearly demonstrated. A numerical model was developed to simulate the transfer of pump noise and validated by these measurements.     We demonstrate a novel convenient nondestructive method based on optical parametric amplification that allows retrieval of the zero-dispersion wavelength map along a short optical fiber span with a high-spatial resolution.     Impact of Holocene climate variability on lacustrine records and human settlements in South Greenland

    International audienceDue to its sensitivity to climate changes, south Greenland is a particularly suitable area to study past global climate changes and their influence on locale Human settlements. A paleohydrological investigation was therefore carried out on two river-fed lakes: Lake Qallimiut and Little Kangerluluup, both located close to the Labrador Sea in the historic 5 farming center of Greenland. Two sediment cores (QAL-2011 and LKG-2011), spanning the last four millennia, were retrieved and showed similar thin laminae, described by high magnetic susceptibility and density, high titanium and TOC / TN atomic ratio, and coarse grain size. They are also characterized either by inverse grading followed by normal grading or by normal grading only and a prevalence of red amorphous particles 10 and lignocellulosic fragments, typical of flood deposits. Flood events showed similar trend in both records: they mainly occurred during cooler and wetter periods characterized by weaker Greenlandic paleo-temperatures, substantial glacier advances, and a high precipitation on the Greenlandic Ice Sheet and North Atlantic ice-rafting events. They can therefore be interpreted as a result of ice and 15 snow-melting episodes. They occurred especially during rapid climate changes (RCC) such as the Middle to Late Holocene transition around 2250 BC, the Sub-boreal/Sub-atlantic transition around 700 BC and the Little Ice Age (LIA) between AD 1300 and AD 1900, separated by cycles of 1500 years and driven by solar forcing. These global RCC revealed by QAL-2011 and LKG-2011 flood events may have influenced Human 20 settlements in south Greenland, especially the paleo-Eskimo cultures and the Norse settlement, and have been mainly responsible for their demise

    Effect of hypoxia/reoxygenation on the cytokine-induced production of nitric oxide and superoxide anion in cultured osteoarthritic synoviocytes

    SummaryObjectiveHypoxia/reoxygenation (H/R) is an important feature in the osteoarthritis (OA) physiopathology. Nitric oxide (NO) is a significant proinflammatory mediator in the inflamed synovium. The purpose of this study was to investigate the effects of H/R on inducible NO synthase (iNOS) activity and expression in OA synoviocytes. In addition we studied the relationship between nitrosative stress and NADPH oxidase (NOX) in such conditions.MethodsHuman cultured synoviocytes from OA patients were treated for 24 h with interleukin 1-β (IL-1β), tumour necrosis factor α (TNF-α) or neither; for the last 6 h, they were submitted to either normoxia or three periods of 1-h of hypoxia followed by 1-h of reoxygenation. NO metabolism (iNOS expression, nitrite and peroxynitrite measurements) was investigated. Furthermore, superoxide anion O2− production, NOX subunit expression and nitrosylation were also assessed.ResultsiNOS expression and nitrite (but not peroxynitrite) production were significantly increased under H/R conditions when compared with to normoxia (P < 0.05). H/R conditions decreased O2− production from ∼0.20 to ∼0.12 nmol min−1 mg proteins−1 (P < 0.05), while NOXs' subunit expression and p47-phox phosphorylation were increased. NOXs and p47-phox were dramatically nitrosylated under H/R conditions (P < 0.05 vs normoxia). Using NOS inhibitors under H/R conditions, p47-phox nitrosylation was prevented and O2− production was restored at normoxic levels (0.21 nmol min−1 mg of proteins−1).ConclusionsOur results provide evidence for an up-regulation of iNOS activity in OA synoviocytes under H/R conditions, associated to a down-regulation of NOX activity through nitrosylation. These findings highlight the importance of radical production to OA pathogenesis, and appraise the metabolic modifications of synovial cells under hypoxia

    Murine MPDZ-linked hydrocephalus is caused by hyperpermeability of the choroid plexus.

    Though congenital hydrocephalus is heritable, it has been linked only to eight genes, one of which is MPDZ Humans and mice that carry a truncated version of MPDZ incur severe hydrocephalus resulting in acute morbidity and lethality. We show by magnetic resonance imaging that contrast medium penetrates into the brain ventricles of mice carrying a Mpdz loss-of-function mutation, whereas none is detected in the ventricles of normal mice, implying that the permeability of the choroid plexus epithelial cell monolayer is abnormally high. Comparative proteomic analysis of the cerebrospinal fluid of normal and hydrocephalic mice revealed up to a 53-fold increase in protein concentration, suggesting that transcytosis through the choroid plexus epithelial cells of Mpdz KO mice is substantially higher than in normal mice. These conclusions are supported by ultrastructural evidence, and by immunohistochemistry and cytology data. Our results provide a straightforward and concise explanation for the pathophysiology of Mpdz-linked hydrocephalus
