    After his St Andrews lecture on ‘Fairy Stories’ Tolkien projected his theory of sub-creation into his legendarium. In this projection the theory of what the human sub-creator does with thought and words became a model for what miraculous and magical beings can do with spirit and matter. For example, Fëanor’s making of the Silmarils and Sauron’s forging of the Ring were conceived (or in the first case, re-conceived) as acts analogous to human fantasy. Middle-earth thus became a world in which the magical potential of human words is revealed in the visible being of magical things

    This essay proposes the idea of incarnation as a key to unlocking Tolkien’s conception of fantasy as set out in the \u27Origins\u27 section of On Fairy Stories. Tolkien\u27s intellectual context is explored and his conception of mythology as a blending of imagination and history examined. The essay also establishes the differences between mortal and Elvish fantasy and argues that Tolkien\u27s Elves engage in a different kind of incarnational art than do mortals. In conclusion it is claimed that in ‘Origins’ Tolkien reworked the speculations of mid-Victorian comparative philology into an aesthetic theory of artistic creation grounded upon the notion of incarnation

    Flagellin induces β-defensin 2 in human colonic ex vivo infection with enterohemorrhagic Escherichia coli

    Enterohemorrhagic E. coli (EHEC) is an important foodborne pathogen in the developed world and can cause life-threatening disease particularly in children. EHEC persists in the human gut by adhering intimately to colonic epithelium and forming characteristic attaching/effacing lesions. In this study, we investigated the innate immune response to EHEC infection with particular focus on antimicrobial peptide and protein expression by colonic epithelium. Using a novel human colonic biopsy model and polarized T84 colon carcinoma cells, we found that EHEC infection induced expression of human β-defensin 2 (hBD2), whereas hBD1, hBD3, LL-37 and lysozyme remained unchanged. Infection with specific EHEC deletion mutants demonstrated that this was dependent on flagellin, and apical exposure to purified flagellin was sufficient to stimulate hBD2 and also interleukin (IL)-8 expression ex vivo and in vitro. Flagellin-mediated hBD2 induction was significantly reduced by inhibitors of NF-κB, MAP kinase p38 and JNK but not ERK1/2. Interestingly, IL-8 secretion by polarized T84 cells was vectorial depending on the side of stimulation, and apical exposure to EHEC or flagellin resulted in apical IL-8 release. Our results demonstrate that EHEC only induces a modest immune response in human colonic epithelium characterized by flagellin-dependent induction of hBD2 and low levels of IL-8

    The GTPase Ras is a molecular switch engaged downstream of G-protein coupled receptors and receptor tyrosine inases that controls multiple cell fate-determining signalling athways. Ras signalling is frequently deregulated in cancer underlying associated changes in cell phenotype. Although Ca2+ signalling pathways control some overlapping functions with Ras, and altered Ca2+ signalling pathways are emerging as important players in oncogenic transformation, how Ca2+ signalling is remodelled during transformation and whether it has a causal role remains unclear. We have investigated Ca2+ signalling in two human colorectal cancer cell lines and their isogenic derivatives in which the mutated K-Ras allele (G13D) has been deleted by homologous recombination. We show that agonist-induced Ca2+ release from intracellular stores is enhanced by loss of K-RasG13D through an increase in the ER store content and a modification of IP3R subtype abundance. Consistently, uptake of Ca2+ into mitochondria and sensitivity to apoptosis was enhanced as a result of KRasG13D loss. These results suggest that suppression of Ca2+ signalling is a common response to naturally occurring levels of K-RasG13D that contributes to a survival advantage during oncogenic transformation

    The Willingness to Pay for Mortality Risk Reductions: A Comparison of the United States and Canada

    We present results for two contingent valuation surveys conducted in Hamilton, Canada and the US to elicit WTP for mortality risk reductions. We find similar Value of Statistical Life estimates across the two studies, ranging from USD 930,000 to USD 4.8 million (2000 US dollars). WTP increases with risk reduction size, but varies little with respondent age: individuals aged over 70 years hold WTP values approximately one-third lower than other respondents. Respondent health status has limited effect on WTP. These results provide little or no evidence for adjusting VSL estimates used in policy analyses for the affected population’s age or health status.value of a statistical life, mortality risks, benefit-cost analysis

    Neighbouring Communities: Interaction, Lessons and Opportunities

    Building, understanding and sharing software that works in creative spaces is increasingly popular and widespread, with many communities outside of academic research interested in pursuing questions highly relevant to Computational Creativity. We report here on several notable communities in the area: the Procedural Generation Jam, the National Novel Generating Month, the Twitterbot community and the #CreativeAI movement. By studying these communities, we benefit from different perspectives on building creative software, as well as how communities of like-minded people form, grow and sustain themselves. We reflect on these communities as sources of lessons for our field and opportunities for future growth and knowledge exchange, as well as raising awareness of sources of inspiration beyond academia
