Sodium dodecyl sulfate allows the persistence and recovery of biofilms of Pseudomonas fluorescens formed under different hydrodynamic conditions
The effect of the anionic surfactant sodium dodecyl sulfate (SDS) on Pseudomonas fluorescens biofilms was
investigated using flow cell reactors with stainless steel substrata, under turbulent (Re=5200) and laminar
(Re=2000) flow. Steady-state biofilms were exposed to SDS in single doses (0.5, 1, 3 and 7 mM) and biofilm
respiratory activity and mass measured at 0, 3, 7 and 12 h after the SDS application. The effect of SDS on biofilm
mechanical stability was assessed using a rotating bioreactor. Whilst high concentrations (7 mM) of SDS promoted
significant biofilm inactivation, it did not significantly reduce biofouling. Turbulent and laminar flow-generated
biofilms had comparable susceptibility to SDS application. Following SDS exposure, biofilms rapidly recovered over
the following 12 h, achieving higher respiratory activity values than before treatment. This phenomenon of posttreatment
recovery was more pronounced for turbulent flow-generated biofilms, with an increase in SDS
concentration. The mechanical stability of the biofilms increased with surfactant application, except for SDS
concentrations near the critical micellar concentration, as measured by biofilm removal due to an increase in external
shear stress forces. The data suggest that although SDS exerts antimicrobial action against P. fluorescens biofilms,
even if only partial and reversible, it had only limited antifouling efficacy, increasing biofilm mechanical stability at
low concentrations and allowing significant and rapid recovery of turbulent flow-generated biofilms.
Microsatellite markers for studies with the carnivorous plant Philcoxia minensis (Plantaginaceae)
Premise of the study: Microsatellite markers were developed for the critically endangered carnivorous species Philcoxia minensis (Plantaginaceae) for further population genetic studies aiming at its conservation. Methods and Results: We identifi ed 29 clones containing 40 microsatellites from a genomic enriched library. A total of 27 primer pairs were developed and evaluated in 30 individuals of a natural P. minensis population. Seventeen markers successfully presented amplifi cation products within the expected size range, of which 12 were polymorphic. The expected and observed heterozygosities ranged from 0.03 to 0.65 and from 0.00 to 0.77, respectively. Positive transferability with the related species P. bahiensis was observed for the same 17 markers. Conclusions: The 12 polymorphic microsatellite markers are suitable for studies in genetic diversity and structure, mating system, and gene fl ow in P. minensis and also may be useful for similar issues regarding the related species P. bahiensis38CONSELHO NACIONAL DE DESENVOLVIMENTO CIENTÍFICO E TECNOLÓGICO - CNPQCOORDENAÇÃO DE APERFEIÇOAMENTO DE PESSOAL DE NÍVEL SUPERIOR - CAPESFUNDAÇÃO DE AMPARO À PESQUISA DO ESTADO DE SÃO PAULO - FAPESPSem informaçãoSem informação2012/12927-

Comparative antibacterial potential of selected aldehyde-based biocides and surfactants against planktonic Pseudomonas fluorescens
The antimicrobial efficacy of two aldehydebased biocides (glutaraldehyde, GTA, and orthophthalaldehyde, OPA) and two surfactants (cetyltrimethyl ammonium bromide, CTAB, and sodium dodecyl sulphate,
SDS) was tested against planktonic Pseudomonas fluorescens. The antimicrobial effects were evaluated by respiratory activity as a measure of the oxygen uptake
rate, adenosine triphosphate (ATP) release, outer membrane proteins (OMP) expression and cellular colour changes. The results were compared with the bacterial
characteristics without chemical treatment. Tests in the presence of bovine serum albumin (BSA), in order to mimic a disinfection process in the real situation under dirty conditions, were performed according to the
European Standard EN-1276. P. fluorescens was completely inactivated with OPA (minimum bactericidal concentration, MBC = 0.5 mM) and CTAB (MBC = 5 mM) and was resistant to GTA and SDS. Only CTAB promoted cellular disruption and consequent
ATP release. The antimicrobial action of the
chemicals tested was significantly reduced when BSA was introduced into the bacterial cultures, increasing markedly the MBC values. Additionally, the presence of BSA
acted as a disruption protective agent when CTAB was applied and stimulated the bacterial respiratory activity when lower concentrations of SDS were tested. The
OMP of the bacterial cells was affected by the application of both surfactants. OMP expression remained unaltered after biocide treatment. Bacterial colour change was
noticed after treatment with biocides and surfactants. In summary, P. fluorescens was extremely resistant to GTA and SDS, with antimicrobial action being quenched
markedly by the reaction with BSA.Instituto de Biotecnologia e Química Fina (IBQF).Fundação para a Ciência e a Tecnologia (FCT) - (Project CHEMBIO - POCI/BIO/61872/2004
Signature for heavy Majorana neutrinos in hadronic collisions
The production and decay of new possible heavy Majorana neutrinos are
analyzed in hadronic collisions. New bounds on the mixing of these particles
with standard neutrinos are estimated according to a fundamental representation
suggested by grand unified models. A clear signature for these Majorana
neutrinos is given by same-sign dileptons plus a charged weak vector boson in
the final state. We discuss the experimental possibilities for the future Large
Hadron Collider (LHC) at CERN.Comment: Latex2e(epsfig), 12 pages, 8 figures, to appear Physical Review
Effect of an extract of Centella asiatica on the biodistribution of sodium pertechnetate (Na99mTcO4) and on the fixation of radioactivity on blood constituents
This study evaluates the effects of an acute treatment with a Centella asiatica (CA) extract on the biodistribution of the radiopharmaceutical Na99mTcO4 and on the fixation of technetium-99m on blood constituents. Wistar rats were treated with CA extract and, 1 hour after, Na99mTcO4 was administered; organs/tissues were withdrawn and weighted. The radioactivity was counted to calculate the percentage of activity per gram (%ATI/g). Also, blood samples were withdrawn, plasma (P), blood cells (BC), insoluble fraction (IF) and soluble fractions of P and BC were isolated and the radioactivity was counted to calculate the percentage of activity (%ATI). Data indicated that the acute treatment with CA extract changed significantly (p99mTcO4 and the fixation of the technetium-99m on blood constituents in an acute treatment
