    The millipede species Oxidus gracilis (Polydesmida:Paradoxosomatidae) is recorded for the first time in Uruguay. This constitutes the first record of the Order Polydesmida and the family Paradoxosomatidae for the country. The species was found in synantropic environments.     [Objective]: The lack of success with classical targeted therapies in cancer treatment has forced researchers to employ either combined therapies or agents that interfere with multiple pathways. In this study, a proteomic evaluation was performed to understand the in vitro antiproliferative effect of dietary compounds against leukemia cells in proteome level. Typical human cell models of leukemia (i.e., wild K562 cells and multi-drug resistant leukemia cells K562/R) were treated with rosemary (Rosmarinus officinalis L.) extracts rich in polyphenols and a comparative proteomic study was performed to identify up- or down-regulated proteins by in vitro antiproliferative effect of polyphenols. [Methods]: The protein fraction from the polyphenol-treated and control K562 and K562/R cells were separated through two dimensional polyacrylamide gel electrophoresis (2DE) and their proteomic profiles compared by image analysis. The proteins identified to be overexpressed or underexpressed in K562 and K562/R cells after the polyphenol treatment were identified by MALDI-TOF/TOF MS. [Results]: The results in this study were evaluated to understand the mode of action of these dietary constituents against leukemia cell proliferation. Dietary polyphenols extracted from rosemary bring about the up regulation or down regulation of different proteins on leukemia cells. These proteins are related to tumorigenesis, cancer proliferation and antioxidant activity. [Conclusion]: Our results revealed that the studied rosemary extract rich in polyphenols induced the down regulation of adenine phosphoribosyl transferase and annexin A1 in K562/R cell lines and tubulin alpha-1C chain in K562 cell lines. According to the expression proteomics results in this study, it could be concluded that the rosemary polyphenols shows in vitro antiproliferative activity in K562 and K562/R leukemia cell lines.Mustafa Çelebier thanks to The Council of Higher Education, Turkey for his grant.     Capillary electrophoresis-mass spectrometry of Spirulina platensis proteins obtained by pressurized liquid extraction

    In this work, the usefulness of CE-MS to monitor and optimize the pressurized liquid extraction (PLE) of proteins from Spirulina platensis microalga is demonstrated. Crude and purified PLE extracts from microalga were analyzed by CE-MS. It was observed that the use of purification protocols of phycobiliproteins (namely, ultrafiltration or precipitation- dialysis-freeze drying) resulted in better CE resolution and MS signals, demonstrating that sample matrix plays an important role in CE-MS of proteins in real samples. Ultrafiltration was found less laborious and much faster than precipitationdialysis- freeze drying (1 vs. 48 h). Direct analysis of crude extracts was demonstrated to be also possible by CE-MS, providing less-quality information but enough to characterize PLE extracts in a much faster way. Therefore, the latter protocol was selected to monitor and optimize the extraction process of phycobiliproteins from S. platensis. To do that, different extraction conditions were tested, including time, temperature and pressure of extraction, nature of pressurized liquid, distribution of microalga inside the extraction cell, type of packing, etc. It is demonstrated that the combined use of PLE and CE-MS allows the attainment of extracts rich in phycobiliproteins in short extraction times (namely, yields of 20% can be obtained in less than 2 h under the optimum PLE process in an automatic way).     Las arañas en agroecosistemas: bioindicadores terrestres de calidad ambiental

    Las arañas son predadores generalistas en ecosistemas terrestres, ampliamente utilizadas en estudios de biodiversidad. En el último siglo, el paisaje de Uruguay se transformó en un mosaico de áreas formadas por ambientes naturales y agroecosistemas. ¿Qué nivel de cambio se espera hallar en la comunidad de arañas si las áreas de estudio están próximas? ¿Qué porcentaje de especies nativas serán reemplazadas con la plantación? ¿Cómo afecta esto a la biodiversidad local? Se realizó un estudio en cuatro áreas vecinas en INIA Las Brujas, Canelones: un bosque ribereño, un bosque de “Espinillo”, una plantación de Eucalyptus globulus y una pradera artificial de Trifolium pratense y Avena sativa. Las arañas fueron capturadas con trampas de caída, trampas de tronco, segado, batido de follaje y colecta manual nocturna. Un total de 3.023 adultos fueron colectados. La mayor abundancia y riqueza de especies se observó en el bosque ribereño. A pesar de la proximidad de los sitios estudiados, se observó un alto reemplazo de especies, especialmente entre áreas naturales y agroecosistemas. Los protocolos de colecta utilizados podrían ser útiles para evaluar el impacto de agroecosistemas con diferentes tipos de manejos sobre la biodiversidad local y su aplicación en la certificación de calidad ambiental

    Characteristics of atheromatosis in the prediabetes stage: a cross-sectional investigation of the ILERVAS project

Our aim was to characterize atheromatous plaque burden by the number of affected territories and the total plaque area in the prediabetes stage. Methods Atheromatous plaque burden (quantity of plaques and total plaque area) was assessed in 12 territories from the carotid and femoral regions using ultrasonography in 6688 non-diabetic middle-aged subjects without cardiovascular disease. Prediabetes was defined by glycosylated hemoglobin (HbA1c) between 5.7 and 6.4% according to the American Diabetes Association guidelines. Results Prediabetes was diagnosed in 33.9% (n = 2269) of the ILERVAS participants. Subjects with prediabetes presented a higher prevalence of subclinical atheromatous disease than participants with HbA1c < 5.7% (70.4 vs. 67.5%, p = 0.017). In the population with prediabetes this was observed at the level of the carotid territory (p < 0.001), but not in the femoral arteries. Participants in the prediabetes stage also presented a significantly higher number of affected territories (2 [1;3] vs. 1 [0;3], p = 0.002), with a positive correlation between HbA1c levels and the number of affected territories (r = 0.068, p < 0.001). However, atheromatosis was only significantly (p = 0.016) magnified by prediabetes in those subjects with 3 or more cardiovascular risk factors. The multivariable logistic regression model showed that the well-established cardiovascular risk factors together with HbA1c were independently associated with the presence of atheromatous disease in participants with prediabetes. When males and females were analyzed separately, we found that only men with prediabetes presented both carotid and femoral atherosclerosis, as well as an increase of total plaque area in comparison with non-prediabetic subjects.This work was partially supported by grants from the Diputació de Lleida and the Generalitat de Catalunya (2017SGR696 and SLT0021600250) and Menarini Spain S.A.     Application of stepwise discriminant analysis to classify commercial orange juices using chiral micellar electrokinetic chromatography-laser induced fluorescence data of amino acids The use of chiral amino acids content and stepwise discriminant analysis to classify three types of commercial orange juices (i.e., nectars, orange juices reconstituted from concentrates, and pasteurized orange juices not from concentrates) is presented. Micellar electrokinetic chromatography with laser-induced fluorescence (MEKC-LIF) and b-cyclodextrins are used to determine L-and D-amino acids previously derivatized with fluorescein isothiocyanate (FITC). This chiral MEKC-LIF procedure is easy to implement and provides information about the main amino acids content in orange arginine, D-Arg, and the non-chiral g-amino-n-butyric acid (GABA), i.e., g-aminobutyric acid). From these results, it is clearly demonstrated that some D-amino acids occur naturally in orange juices. Application of stepwise discriminant analysis to 26 standard samples showed that the amino acids L-Arg, L-Asp and GABA were the most important variables to differentiate the three groups of samples. With these three selected amino acids a 100% correct classification of the samples was obtained either by standard or by leave-one-out cross-validation procedures. These classification functions based on the content in L-Arg, L-Asp and GABA were also applied to nine test samples and provided an adequate classification and/or interesting information on these samples. It is concluded that chiral MEKC-LIF analysis of amino acids and stepwise discriminant analysis can be used as a consistent procedure to classify commercial orange juices providing useful information about their quality and processing.     Phenotyping Type 2 Diabetes in Terms of Myocardial Insulin Resistance and Its Potential Cardiovascular Consequences: A New Strategy Based on 18F-FDG PET/CT

However, the role of myocardial insulin resistance (mIR) remains to be clarified. Methods: Two 18F-FDG PET/CT scans were performed on forty-three T2D patients at baseline and after hyperinsulinemic–euglycemic clamp (HEC). Myocardial insulin sensitivity (mIS) was determined by measuring the increment in myocardial 18F-FDG uptake after HEC. Coronary artery calcium scoring (CACs) and myocardial radiodensity (mRD) were assessed by CT. Results: After HEC, seventeen patients exhibited a strikingly enhancement of myocardial 18F-FDG uptake and twenty-six a marginal increase, thus revealing mIS and mIR, respectively. Patients with mIR showed higher mRD (HU: 38.95 [33.81–44.06] vs. 30.82 [21.48–38.02]; p = 0.03) and CACs > 400 (AU: 52% vs. 29%; p = 0.002) than patients with mIS. In addition, HOMA-IR and mIS only showed a correlation in those patients with mIR. Conclusions: 18F-FDG PET combined with HEC is a reliable method for identifying patients with mIR. This subgroup of patients was found to be specifically at high risk of developing cardiovascular events and showed myocardial structural changes. Moreover, the gold-standard HOMA-IR index was only associated with mIR in this subgroup of patients.     Uncoupled seasonal variability of transparent exopolymer and Coomassie stainable particles in coastal Mediterranean waters: Insights into sources and driving mechanisms

    Transparent exopolymer particles (TEP) and Coomassie stainable particles (CSP) are gel-like particles, ubiquitous in the ocean, that affect important biogeochemical processes including organic carbon cycling by planktonic food webs. Despite much research on both groups of particles (especially TEP) over many years, whether they exist as distinctly stainable fractions of the same particles or as independent particles, each with different driving factors, remains unclear. To address this question, we examined the temporal dynamics of TEP and CSP over 2 complete seasonal cycles at 2 coastal sites in the Northwestern Mediterranean Sea, the Blanes Bay Microbial Observatory (BBMO) and the L’Estartit Oceanographic Station (EOS), as well as their spatial distribution along a coast-to-offshore transect. Biological, chemical, and physical variables were measured in parallel. Surface concentrations (mean + standard deviation [SD]) of TEP were 36.7 + 21.5 µg Xanthan Gum (XG) eq L–1 at BBMO and 36.6 + 28.3 µg XG eq L–1 at EOS; for CSP, they were 11.9 + 6.1 µg BSA eq L–1 at BBMO and 13.0 + 5.9 µg BSA eq L–1 at EOS. Seasonal variability was more evident at EOS, where surface TEP and CSP concentrations peaked in summer and spring, respectively, and less predictable at the shore-most station, BBMO. Vertical distributions between surface and 80 m, monitored at EOS, showed highest TEP concentrations within the surface mixed layer during the stratification period, whereas CSP concentrations were highest before the onset of summer stratification. Phytoplankton were the main drivers of TEP and CSP distributions, although nutrient limitation and saturating irradiance also appeared to play important roles. The dynamics and distribution of TEP and CSP were uncoupled both in the coastal sites and along the transect, suggesting that they are different types of particles produced and consumed differently in response to environmental variability

    Flat Peach SUBLOR and SUBLIM and Flat Nectarine PERLA Series

    Flat peach and flat nectarine (Prunus persica L. Batsch.) production in Spain started from local cultivars at marginal sites, mainly in the Ebro Valley and Murcia regions. At the end of the 1990s, the introduction of improved cultivars from France and Italy, with more red skin color coverage and better regular shape, fruit size, and stylar cavity closure, together with superior handling and transport conditions led the Spanish peach sector to develop this fruit type on a commercial scale. Currently, Spain is the largest European producer and exporter of flat peach and nectarine, offering a wide range of cultivars covering the producing period from May to September. In 2020, a total of 10.200 ha for flat peach and 481 ha for flat nectarine together produced 263.651 t (20% of the total peach production) (MAPAMA, 2022). To guarantee adaptation to local growing conditions (Font i Forcada et al., 2021a), several national breeding programs started to develop new flat peach and flat nectarine cultivars. One of these breeding programs is the ASF-IRTA breeding program, which is located at Gimenells (Lleida) in the Ebro Valley (northeastern Spain), which is a major region of peach production in Spain (Batlle et al., 2012; Cantín et al., 2017). It was started in 2004, with the objective of developing new cultivars adapted to chilling between 1000 and 1500 chill units or between 42 and 75 chill portions and warm conditions similar to the Mediterranean area (32 °C mean daily summer temperature and 300–400 mm of annual rainfall). It aimed to produce high-quality fruits, both at harvest and after cold storage, with the ultimate goal of satisfying consumers (Font i Forcada et al., 2021a, 2021b). As a result of this collaborative effort, the ASF-IRTA scion peach breeding program has already released the SUBLOR (one flat yellow-fleshed peach), SUBLIM (six flat white-fleshed peach), and PERLA (two flat white-fleshed nectarine) series. These nine new flat peach and nectarine cultivars produce highly attractive yellow-fleshed or white-fleshed fruits with high coverage of red skin, a balanced or acid flavor, sweet taste, and good flesh firmness.
