    To B or Not to B: Understanding B Cell Responses in the Development of Malaria Infection

    Malaria is a widespread disease caused mainly by the Plasmodium falciparum (Pf) and Plasmodium vivax (Pv) protozoan parasites. Depending on the parasite responsible for the infection, high morbidity and mortality can be triggered. To escape the host immune responses, Plasmodium parasites disturb the functionality of B cell subsets among other cell types. However, some antibodies elicited during a malaria infection have the potential to block pathogen invasion and dissemination into the host. Thus, the question remains, why is protection not developed and maintained after the primary parasite exposure? In this review, we discuss different aspects of B cell responses against Plasmodium antigens during malaria infection. Since most studies have focused on the quantification of serum antibody titers, those B cell responses have not been fully characterized. However, to secrete antibodies, a complex cellular response is set up, including not only the activation and differentiation of B cells into antibody-secreting cells, but also the participation of other cell subsets in the germinal center reactions. Therefore, a better understanding of how B cell subsets are stimulated during malaria infection will provide essential insights toward the design of potent interventions

    Cuidados de Enfermagem Veterinária a uma cria de doninha (Mustela nivalis) em Centro de Recuperação de Animais Selvagens

    A doninha (Mustela nivalis) é um pequeno carnívoro, de espécie autóctone e protegida1, cuja entrada em Centros de Recuperação de Animais Selvagens não é frequente. Desta forma, torna-se pertinente a descrição dos cuidados de enfermagem veterinária aplicados na recuperação de uma cria desta espécie, que deu entrada no CERAS – Centro de Estudos e Recuperação de Animais Selvagens – com cerca de 10 dias de idade e 20 gramas de peso. O acompanhamento do desenvolvimento da cria é um dos papéis a desempenhar pelo Enfermeiro Veterinário, sobretudo ao nível da alimentação (desde as refeições com leite de substituição, o desmame, a introdução gradual de novos alimentos e o treino de caça), higiene e desenvolvimento comportamental (prevenção de imprinting e estímulo sociocognitivo). A monitorização do crescimento da cria foi feita através de ferramentas como controlo do peso e da temperatura corporal, exame visual e exame coprológico. A evolução do peso corporal foi positiva, com um ganho médio diário de 1,21g e um peso final de 95g. O exame coprológico detetou a presença de oocistos de Eimeria spp. (1800 opg), ovos (1150 Opg) e formas larvares (50 Lpg) de Strongyloides spp. Uma das maiores exigências na prestação destes cuidados foi corresponder às elevadas necessidades energéticas características desta espécie2. Ainda assim, a recuperação foi bem-sucedida e culminou com a libertação da doninha, fisicamente saudável e exibindo os comportamentos naturais da espécie, apenas 2 meses após a entrada no CERAS.info:eu-repo/semantics/publishedVersio

    Acute exposition to Roundup Transorb® induces systemic oxidative stress and alterations in the expression of newly sequenced genes in silverside fish (Odontesthes humensis)

    Roundup Transorb® (RDT) is a glyphosate-based herbicide commonly used in agricultural practices worldwide. This herbicide exerts negative effects on the aquatic ecosystem and affects bioenergetic and detoxification pathways, oxidative stress, and cell damage in marine organisms. These effects might also occur at the transcriptional level; however, the expression of genes associated with oxidative stress has not been studied well. Odontesthes humensis is a native Brazilian aquatic species naturally distributed in the habitats affected by pesticides, including Roundup Transorb® (RDT). This study evaluated the toxic effects of short-term exposure to RDT on O. humensis. Moreover, the genes related to oxidative stress were sequenced and characterized, and their expressions in the gills, hepatopancreas, kidneys, and brain of the fish were quantified by quantitative reverse transcription-polymerase chain reaction. The animals were exposed to two environmentally relevant concentrations of RDT (2.07 and 3.68 mg L−1) for 24 h. Lipid peroxidation, reactive oxygen species (ROS), DNA damage, and apoptosis in erythrocytes were quantified by flow cytometry. The expression of the target genes was modulated in most tissues in the presence of the highest tested concentration of RDT. In erythrocytes, the levels of lipid peroxidation, ROS, and DNA damage were increased in the presence of both the concentrations of RDT, whereas cell apoptosis was increased in the group exposed to 3.68 mg L−1 RDT. In conclusion, acute exposure to RDT caused oxidative stress in the fish, induced negative effects on cells, and modulated the expression of genes related to the enzymatic antioxidant system in O. humensis.Fil: Martins, Amanda Weege S..     Macadamia Oil Supplementation Attenuates Inflammation and Adipocyte Hypertrophy in Obese Mice

    Excess of saturated fatty acids in the diet has been associated with obesity, leading to systemic disruption of insulin signaling, glucose intolerance, and inflammation. Macadamia oil administration has been shown to improve lipid profile in humans. We evaluated the effect of macadamia oil supplementation on insulin sensitivity, inflammation, lipid profile, and adipocyte size in high-fat diet (HF) induced obesity in mice. C57BL/6 male mice (8 weeks) were divided into four groups: (a) control diet (CD), (b) HF, (c) CD supplemented with macadamia oil by gavage at 2g/Kg of body weight, three times per week, for 12 weeks (CD + MO), and (d) HF diet supplemented with macadamia oil (HF + MO). CD and HF mice were supplemented with water. HF mice showed hypercholesterolemia and decreased insulin sensitivity as also previously shown. HF induced inflammation in adipose tissue and peritoneal macrophages, as well as adipocyte hypertrophy.     Perception of hypertensive patients about their non-adherence to the use of medication

    This qualitative study aims to analyze the perception of patients with hypertension on their non-adherence to medication. 13 participants were interviewed, classified as non-adherent.The analysis was performed using the technique of thematic content analysis. Data points to contradictions in the approach of what is being adherent or not, the difficulty of adhering to the use of medication due to lifestyle habits, that forgetting is understood as a justification for non-compliance, and reinforces factors that hinder such practice, such as the use of many drugs, the presence of signs and symptoms and changes in daily routine. With complex conditions that involve non-adherence to treatment and the current context of the predominance of chronic diseases, it is essential to invest in innovative strategies of care for such people

    Fixed and volatile constituents oF genus Croton plants: C. adenoCalyx Baill -euphorBiaceae

    aBstract This work describes the phytochemical analysis of Croton adenocalyx Baill (Euphorbiaceae), a plant that is representative of the species from Ceará State (Brazil). The GC-MS analysis of the essential oil, obtained by hydrodistillation of the leaves from C. adenocalyx, allowed the identification of eleven volatile constituents; the main components were identified as α-pinene (32.63%); bicyclogermacrene (13.96%); trans-caryophyllene (10.23%); germacrene D (10.14%); β-pinene (10.11%) and β-elemene (8.31%). The chromatographic purification of the ethanolic extract from the trunk bark, allowed the isolation and identification of the 6-methoxy-7-hydroxycoumarin (1) and 3´,5-dihydroxy-3,4´,7-trimethoxyflavone (2). Los componentes mayoritarios fueron identificados como α-pineno (32.63%), biciclogermacreno (13.96%), trans-cariofileno (10.23%), germacreno D (10.14%), β-pineno (10.11%) y β-elemeno (8.31%). La purificación cromatográfica del extracto etanólico de la corteza del tronco, permitió aislar e identificar la coumarina 6-metoxi-7-hidroxicumarina (1) y el 3´,5-dihidroxi-3,4´,7-trimetoxiflavona (2)

    Fetal deaths in Brazil: a systematic review

    OBJECTIVE To review the frequency of and factors associated with fetal death in the Brazilian scientific literature. METHODS A systematic review of Brazilian studies on fetal deaths published between 2003 and 2013 was conducted. In total, 27 studies were analyzed; of these, 4 studies addressed the quality of data, 12 were descriptive studies, and 11 studies evaluated the factors associated with fetal death. The databases searched were PubMed and Lilacs, and data extraction and synthesis were independently performed by two or more examiners. RESULTS The level of completeness of fetal death certificates was deficient, both in the completion of variables, particularly sociodemographic variables, and in defining the underlying causes of death. Fetal deaths have decreased in Brazil; however, inequalities persist. Analysis of the causes of death indicated maternal morbidities that could be prevented and treated. The main factors associated with fetal deaths were absent or inadequate prenatal care, low education level, maternal morbidity, and adverse reproductive history. CONCLUSIONS Prenatal care should prioritize women that are most vulnerable (considering their social environment or their reproductive history and morbidities) with the aim of decreasing the fetal mortality rate in Brazil. Adequate completion of death certificates and investment in the committees that investigate fetal and infant deaths are necessary
