    Preliminary assessment of cardiac short term safety and efficacy of manganese chloride for cardiovascular magnetic resonance in humans

    <p>Abstract</p> <p>Background</p> <p>Manganese based agents are intracellular and accumulate inside myocytes allowing for different imaging strategies compared to gadolinium contrasts. While previous agents release manganese very slowly in the circulation, MnCl<sub>2 </sub>allows for rapid Mn2<sup>+ </sup>uptake in myocytes, creating a memory effect that can be potentially explored. Data on animal models are very encouraging but the safety and efficacy of this approach in humans has not yet been investigated. Therefore, our objectives were to study the safety and efficacy of a rapid infusion of manganese chloride (MnCl<sub>2</sub>) for cardiovascular magnetic resonance (CMR) in humans.</p> <p>Methods</p> <p>Fifteen healthy volunteers underwent a CMR scan on a 1.5 T scanner. Before the infusion, cardiac function was calculated and images of a short axis mid-ventricular slice were obtained using a 2D and 3D gradient-echo inversion recovery (GRE-IR) sequence, a phase-sensitive IR sequence and a single breath-hold segmented IR prepared steady-state precession acquisition for T<sub>1 </sub>calculations. MnCl<sub>2 </sub>was infused over three minutes at a total dose of 5 μMol/kg. Immediately after the infusion, and at 15 and 30 minutes later, new images were obtained and cardiac function re-evaluated.</p> <p>Results</p> <p>There was a significant decrease in T<sub>1 </sub>values compared to baseline, sustained up to 30 minutes after the MnCl<sub>2 </sub>infusion (pre,839 ± 281 ms; 0 min, 684 ± 99; 15 min, 714 ± 168; 30 min, 706 ± 172, P = 0.003). The 2D and 3D GRE-IR sequence showed the greatest increase in signal-to-noise ratio compared to the other sequences (baseline 6.6 ± 4.2 and 9.7 ± 5.3; 0 min, 11.3 ± 4.1 and 15.0 ± 8.7; 15 min, 10.8 ± 4.0 and 16.9 ± 10.2; 30 min, 10.6 ± 5.2 and 16.5 ± 8.3, P < 0.001 for both). There was a slight increase in systolic pressure and heart rate after three and four minutes of the infusion with normalization of these parameters thereafter. Patients showed good tolerance to MnCl<sub>2 </sub>with no major adverse events, despite all reporting transient facial flush.</p> <p>Conclusions</p> <p>In the short term, MnCl<sub>2 </sub>appears safe for human use. It effectively decreases myocardium T<sub>1</sub>, maintaining this effect for a relatively long period of time and allowing for the development of new imaging strategies in CMR, especially in ischemia research.</p

    Curraleiro Pé Duro cattle breed (Bos taurus) differ in phagocytic Neutrophil function related to crossbreed cattle (Bos taurus X Bos indicus).

    This work aimed to evaluate hematological parameters and phagocytic neutrophil functions of naturally selected Curraleiro Pé Duro cattle breed (Bos taurus) and crossbreed cattle (Bos taurus X Bos indicus). Two groups of adult cows: Natural= Curraleiro Pé Duro cattle breed (n=10) and Selected = ¾ Nelore X Simental crossbreed animals (n=10) were evaluated. Blood was sampled to make hemogram, in vitro evaluation of neutrophil phagocytosis and basal Nitroblue Tetrazolium - NBT reduction (without any stimulus). The results showed no difference in hematological parameters between these groups. Natural group had greater phagocytic index of neutrophils through pathogen-associated molecular patterns receptors (PAMPs) while in Selected group this same index was greater using opsonin receptors. The reduction of NBT was lower for Natural group than Selected group. In conclusion, we suggest that both animal groups showed differences in neutrophil phagocytic functions in relation to pathway activation and Curraleiro Pé Duro cattle breed produced more oxygen radicals (superoxide, O2-) than the other tested group

    Metabolizable energy and metabolizability coefficients of moringa and bocaiuva for slow-growing broilers at different ages.

    This study aimed to determine nutrient metabolizability coefficients and apparent metabolizable energy (AME) and nitrogen-corrected AME (AMEn) values of moringa leaf meal and bocaiuva pulp for slow-growing broilers. Three hundred and fifty 1-day-old chicks of the Red Naked Neck line were allocated to five treatments in a completely randomized design with seven replicates of 10 birds. Treatments consisted of a control diet and four test diets. The proportion of ingredients evaluated in the test diets was 20% moringa leaf meal and bocaiuva pulp and 40% corn and soybean meal. The total excreta collection method was applied with 1% ferric oxide included in the diets as a marker of start and end of collection, in the starter, grower and finisher phases. The metabolizability coefficients of dry matter, crude protein and ether extract and the AME and AMEn values of moringa leaf meal, bocaiuva pulp, corn and soybean meal were determined. Data were subjected to analysis of variance and means were compared by Tukey?s test at the 5% probability level. The protein from moringa leaf meal and the fat from bocaiuva pulp are highly metabolizable by birds at different ages. The AME and AMEn values of moringa leaf meal ranged from 2557 to 2868 Kcal/kg and 2205 to 2479 Kcal/kg, respectively. In bocaiuva pulp, AME and AMen ranged between 2680 and 3119 Kcal/kg and 2483 and 2490 Kcal/kg, respectively. Therefore, the alternative ingredients can be used in the formulation of diets for slow-growing broilers to partially replace corn and soybean meal

    A large Venous-Arterial PCO2 Is Associated with Poor Outcomes in Surgical Patients

    Background. This study evaluated whether large venous-arterial CO2 gap (PCO2 gap) preoperatively is associated to poor outcome. Method. Prospective study which included adult high-risk surgical patients. The patients were pooled into two groups: wide [P(v-a)CO2] versus narrow [P(v-a)CO2]. In order to determine the best value to discriminate hospital mortality, it was applied a ROC (receiver operating characteristic) curve for the [P(v-a)CO2] values collected preoperatively, and the most accurate value was chosen as cut-off to define the groups. Results. The study included 66 patients. The [P(v-a)CO2] value preoperatively that best discriminated hospital mortality was 5.0 mmHg, area = 0.73. Preoperative patients with [P(v-a)CO2] more than 5.0 mmHg presented a higher hospital mortality (36.4% versus 4.5% P = 0.004), higher prevalence of circulatory shock (56.8% versus 22.7% P = 0.01) and acute renal failure postoperatively (27.3% versus 4.5% P = 0.02), and longer hospital length of stays 20.0 (14.0–30.0) versus 13.5 (9.0–25.0) days P = 0.01.
